1Transformation of japonica rice with RHL gene and salt tolerance of the transgenic rice plant显示文摘Overexpression of the yeast HAL2 gene increases salt tolerance of yeast and plant. Rice HAL2-like (RHL) gene was introduced into a japonica rice cultivar HJ19 with Agrobacterium tumefaciens-mediated transformation. Transgenic plants in R0 generation were selected on the principle of GUS-positive, RHL gene PCR-positive and normal growth. Hygromycin-resistant plants of some transgenic lines in R1 generation increased salt tolerance during the seedling and booting stage, being less damaged in the cy-tomembrane and stronger in leaf tissue viability under salt stress during booting period. Southern analysis of transgenic lines tolerant to salt in R1 generation showed that the RHL gene expression cassette had been successfully integrated into rice genome. 2Hierarchical micro-nanostructures from polyoxometalates and polydopamine:Characterization,electrochemical and intrinsic peroxidase-like properties显示文摘Hybrid composite materials with hierarchical structures have attracted continuing attention for their enhanced features and various applications.The new hybrid composites XW_(9)@PDA(XW_(9) means Na_(8)H[α-PW_(9)O_(34)],Na_(10)[α-SiW_(9)O_(34)]and Na_(10)[α-GeW_(9)O_(34)];PDA means polydopamine)have been synthesized for potential sensing applications.The effect of polyoxometalates on the formation of the hybrid composites was investigated.Scanning electron microscopy(SEM)supported the obtained XW_(9)@PDA were hierarchical microsphere.By monitoring the reaction of classical peroxidase substrate o-phenylenediamine(OPD)to form yellow 2,3-diaminophenazine(DAB)in the presence of H_(2)O_(2),the peroxidase-like activity of XW_(9)@PDA was calculated.The results showed that XW_(9)@PDA still keep the peroxidase-like activity and have 505.98%,208.58%and 582.78%enhancement in intrinsic catalytic activity for PW_(9)@PDA,SiW_(9)@PDA and GeW_(9)@PDA.Among the three composites,PW_(9)@PDA showed the highest peroxidase-like activity.Therefore,PW_(9)@PDA was applicable to the colorimetric and electrochemical sensing for H_(2)O_(2).Ruijuan Lv Xinming Zhang Ruimeng Sun Lixia Chen Yang Zhang Rongtian Sheng Ting Du Yuhan Li Yanfei Qi 2022Particuology2022,20,5:1
3Expression Characteristics and Borer Resistance of Bt Gene of Transgenic cry1C~* Super Japonica Rice显示文摘[Objective] The paper was to study the expression characteristics and borer resistance of Bt gene of Songjing 9(1C~*). [Method] With super japonica rice Songjing 9 as the receptor, transgenic cry1C~* super japonica rice Songjing 9(1C~*) regulated by ubi promoter was created by agrobacterium-mediated genetic transformation method. Ten strains of transgenic super japonica rice formed from different transformation events were planted in the field; the Bt-m RNA of leaf, stem-sheath and young spikelet at booting stage were detected by real-time fluorescent quantitative PCR; the Bt protein content of leaf, stem-sheath and young spikelet and harvested brown rice were detected by enzyme-linked immunosorbent assay(ELISA); the white head rate damaged by Chilo suppressalis was manually investigated. [Result] The Bt-m RNA and Bt protein content of various organs and Bt protein content of mature brown rice at booting stage of Songjing 9(1C~*) significantly differed among strains; there was no correlation relationship in Bt-m RNA and Bt protein content between leaf and stem-sheath, between leaf and young spikelet, and between stem-sheath and young spikelet; there was no relation between Bt-m RNA or Bt protein content of leaf, stem-sheath and young spikelet and Bt protein content of brown rice; there was positive correlation between Bt-m RNA and Bt protein content in the same organ such as leaf, stem-sheath and young spikelet at booting stage; such strains as Songjing 9(1C~*), whether the Bt gene expression was high or low, showed good resistance against C. suppressalis.This indicated that there were differences in Bt gene expression in different strains of transgenic cry1C~* super japonica rice or different organs of the same strain; there was no relation in Bt gene expression among different organs; the Bt-m RNA and protein content of the same organ usually had consistent expression; the Bt gene expression of various organs had nothing to do with the Bt protein content of brown rice.
