1Molecular cloning and functional identification of sterol C24-methyltransferase gene from Tripterygium wilfordii显示文摘Sterol C24-methyltransferase(SMT) plays multiple important roles in plant growth and development. SMT1, which belongs to the family of transferases and transforms cycloartenol into 24-methylene cycloartenol, is involved in the biosynthesis of 24-methyl sterols. Here, we report the cloning and characterization of a cDNA encoding a sterol C24-methyltransferase from Tripterygium wilfordii(Tw SMT1). Tw SMT1(Gen Bank access number KU885950) is a 1530 bp cDNA with a 1041 bp open reading frame predicted to encode a 346-amino acid, 38.62 k Da protein. The polypeptide encoded by the SMT1 cDNA was expressed and purified as a recombinant protein from Escherichia coli(E. coli) and showed SMT activity. The expression of Tw SMT1 was highly up-regulated in T. wilfordii cell suspension cultures treated with methyl jasmonate(Me JA). Tissue expression pattern analysis showed higher expression in the phellem layer compared to the other four organs(leaf, stem, xylem and phloem), which is about ten times that of the lowest expression in leaf. 2Tailoring polysulfide trapping and kinetics by engineering hollow carbon bubble nanoreactors for high-energy Li-S pouch cells显示文摘Despite great progress of lithium-sulfur(Li-S)battery performance at the laboratory-level,both key parameters and challenges at cell scales to achieve practical high energy density require high-sulfur-loading cathodes and lean electrolytes.Herein,a novel carbon foam integrated by hollow carbon bubble nanoreactors with ultrahigh pore volume of 6.9 cm3·g−1 is meticulously designed for ultrahigh sulfur content up to 96 wt.%.Tailoring polysulfide trapping and ion/electron transport kinetics during the charge-discharge process can be achieved by adjusting the wall thickness of hollow carbon bubbles.And a further in-depth understanding of electrochemical reaction mechanism for the cathode is impelled by the in-situ Raman spectroscopy.As a result,the as-prepared cathode delivers high specific capacitances of 1,269 and 695 mAh·g−1 at 0.1 and 5 C,respectively.Furthermore,Li-S pouch cells with high areal sulfur loading of 6.9 mg·cm−2 yield exceptional practical energy density of 382 Wh·kg−1 under lean electrolyte of 3.5µL·mg−1,which demonstrates the great potential for realistic high-energy Li-S batteries.Lei Wang Shuangke Liu Jin Hu Xianan Zhang Xin Li Guanhua Zhang Yujie Li Chunman Zheng Xiaobin Hong Huigao Duan 2021Nano Research2021,14,5:3

5Multiple-statistical genome-wide association analysis and genomic prediction of fruit aroma and agronomic traits in peaches显示文摘‘Chinese Cling’is an important founder in peach breeding history due to the pleasant flavor.Genome-wide association studies(GWAS)combined with genomic selection are promising tools in fruit tree breeding,as there is a considerable time lapse between crossing and release of a cultivar.In this study,242 peaches from Shanghai germplasm were genotyped with 145456 single-nucleotide polymorphisms(SNPs).The six agronomic traits of fruit flesh color,fruit shape,fruit hairiness,flower type,pollen sterility,and soluble solids content,along with 14 key volatile odor compounds(VOCs),were recorded for multiple-statistical GWAS.Except the reported candidate genes,six novel genes were identified as associated with these traits.Thirty-nine significant SNPs were associated with eight VOCs.The putative candidate genes were confirmed for VOCs by RNA-seq,including three genes in the biosynthesis pathway found to be associated with linalool,soluble solids content,and cis-3-hexenyl acetate.Multiple-trait genomic prediction enhanced the predictive ability forγ-decalactone to 0.7415 compared with the single-trait model value of 0.1017.One PTS1-SSR marker was designed to predict the linalool content,and the favorable genotype 187/187 was confirmed,mainly existing in the‘Shanghai Shuimi’landrace.Overall,our findings will be helpful in determining peach accessions with the ideal phenotype and show the potential of multiple-trait genomic prediction to improve accuracy for highly correlated genetic traits.The diagnostic marker will be valuable for the breeder to bridge the gap between quantitative trait loci and marker-assisted selection for developing strong-aroma cultivars.Xiongwei Li Jiabo Wang Mingshen Su Minghao Zhang Yang Hu Jihong Du Huijuan Zhou Xiaofeng Yang Xianan Zhang Huijuan Jia Zhongshan Gao Zhengwen Ye 2023Horticulture Research2023,10,7:0
