| 1 | A Simplified PCR-SSP Method for HLA-A2 Subtype in a Population of Wuhan,China显示文摘HLA-A2 is the most frequent HLA-A allele in all ethnic populations, and an important restriction element for peptide presentation to T cells in infectious disease and cancer. However, the HLA-A2 supertype consisting of up to 75 subtypes, mutation studies and analyses using cytotoxic T lymphocytes suggest the functional relevance of subtype-specific differences in HLA-A2 molecules for peptide binding and T-cell recognition. Therefore, it is necessary for T-cell response study to discriminate the HLA-A2 subtypes and to understand the profile of HLA-A2 allelic distribution in a given population. In this study, we developed a simple, robust approach based on the nested polymerase chain reaction using sequence-specific primers (PCR-SSP) to discriminate 17 HLA-A2 subtypes which cover the most HLA-A2 alleles (> 99% allele frequency) reported in Chinese, using 15 combinations of 19 allelic specific primers. In the first round of PCR, 3 combinations of 5 primers were used to determine whether the tested sample was HLA-A2 positive, meanwhile the subtypes of HLA-A*0209 and HLA-A*0215N were determined for the variant position of these two subtypes is in exon 4 instead of exon 2, 3. Samples of HLA-A2 positive were subtyped in the second round of PCR, using PCR products of the first round as templates. This strategy was applied to test the samples of 78 random HLA-A2 positive individuals for their HLA-A2 subtypes. Those samples were screened for HLA-A2 positive by the first round PCR-SSP from 154 healthy blood donors in Wuhan, China. The subtyping results were verified by using flow cytometric analysis (FCM) with HLA-A2 specific monoclonal antibody BB7.2 and DNA sequencing. The typing results of the samples show 50.7% random individuals in the population carry HLA-A2, HLA-A*0201 ranks the first (allele frequency = 15.5%), followed by A*0207 (5.8%), A*0206 (4.7%), A*0203 (2.6%), A*0210 (0.7%), and these 5 alleles account for 99.0% HLA-A2 subtypes of allele frequency. | 2 | Influence of Deposition Temperature and Pressure on Microstructure and Tribological Properties of Arc Ion Plated Ag Films显示文摘The films deposited at low temperature(LT-films) have increasingly attracted theoretical and technical interests since such films exhibit obvious difference in structure and performances compared to those deposited at room temperature.Studies on the tribological properties of LT-films are rarely reported in available literatures.In this paper,the structure,morphology and tribological properties of Ag films,deposited at LT(166 K) under various Ar pressures on AISI 440C steel substrates by arc ion plating(AIP),are studied by X-ray diffraction(XRD),atomic force microscopy(AFM) and a vacuum ball-on-disk tribometer,and compared with the Ag films deposited at RT(300 K).XRD results show that(200) preferred orientation of the films is promoted at LT and low Ar pressure.The Crystallite sizes are 70 nm-80 nm for LT-Ag films deposited at 0.2 Pa and 0.8 Pa and larger than 100 nm for LT-Ag films deposited at 0.4 Pa and 0.6 Pa,while they are 55 nm-60 nm for RT-Ag films deposited at 0.2 Pa-0.6 Pa and 37 nm for RT-Ag films deposited at 0.8 Pa.The surfaces of LT-Ag films are fibre-like at 0.6 Pa and 0.8 Pa,terrace-like at 0.4 Pa,and sphere-like at 0.2 Pa,while the surfaces of RT-Ag films are composed of sphere-like grains separated by voids.Wear tests reveal that,due to the compact microstructure LT-Ag films have better wear resistances than RT-Ag film.These results indicate that the microstructure and wear resistance of Ag films deposited by AIP can be improved by low temperature deposition. | HU Ming GAO Xiaoming SUN Jiayi WENG Lijun ZHOU Feng LIU Weimin | 2012 | Chinese Journal of Mechanical Engineering2012,25,4: | 2 |

| 5 | Macro-superlubricity in sputtered MoS_(2)-based films by decreasing edge pinning effect显示文摘To date,MoS_(2) can only be achieved at microscale.Edge pinning effect caused by structure defects is the most obvious barrier to expand the size of structural superlubricity to macroscale.Herein,we plan to pin edge planes of MoS_(2) with nanospheres,and then the incommensurate structure can be formed between adjacent rolling nanoparticles to reduce friction.The sputtered MoS_(2) film was prepared by the physical vapor deposition(PVD)in advance.Then enough Cu_(2)O nanospheres(~40 nm)were generated in situ at the edge plane of MoS_(2) layers by liquid phase synthesis.An incommensurate structure(mismatch angle(θ)=8°)caused by MoS_(2) layers was formed before friction.The friction coefficient of the films(5 N,1,000 r/min)was~6.0×10^(−3) at the most.During friction,MoS_(2) layers pinned on numerous of Cu_(2)O nanoparticles reduced its edge pinning effect and decreased friction.Moreover,much more incommensurate was formed,developing macro-superlubricity. | Chunmeng DONG Dong JIANG Yanlong FU Desheng WANG Qinqin WANG Lijun WENG Ming HU Xiaoming GAO Jiayi SUN | 2024 | Friction2024,12,1: | 0 |
| 6 | Co_(13)O_(8)—metalloxocubes:a new class of perovskite-like neutral clusters with cubic aromaticity显示文摘Exploring stable clusters to understand structural evolution from atoms to macroscopic matter and to construct new materials is interesting yet challenging in chemistry.Utilizing our newly developed deep-ultraviolet laser ionization mass spectrometry technique,here we observe the reactions of neutral cobalt clusters with oxygen and find a very stable cluster species of Co13O8 that dominates the mass distribution in the presence of a large flow rate of oxygen gas.The results of global-minimum structural search reveal a unique cubic structure and distinctive stability of the neutral Co13O8 cluster that forms a new class of metal oxides that we named as‘metalloxocubes’.Thermodynamics and kinetics calculations illustrate the structural evolution from icosahedral Co13 to the metalloxocube Co13O8 with decreased energy,enhanced stability and aromaticity.This class of neutral oxygen-passivated metal clusters may be an ideal candidate for genetic materials because of the cubic nature of the building blocks and the stability due to cubic aromaticity. | Lijun Geng Mouyi Weng Cong-Qiao Xu Hanyu Zhang Chaonan Cui Haiming Wu Xin Chen Mingyu Hu Hai Lin Zhen-Dong Sun Xi Wang Han-Shi Hu Jun Li Jiaxin Zheng Zhixun Luo Feng Pan Jiannian Yao | 2021 | National Science Review2021,8,1: | 0 |
