| 1 | Purification and Characterization of a New Thermostable κ-CarrageenasefromtheMarineBacterium Pseudoalteromonas sp. QY203显示文摘A new extracellular κ-carrageenase, namely CgkP, 34.0 kDa in molecular weight, was purified from Pseudoalteromonas sp. QY203. CgkP showed relatively high activity at acidities ranging from pH6.0 to pH9.0 and temperatures ranging from 30℃ to 50℃ with the highest activity at 45℃ and pH7.2. Sodium chloride increased its activity markedly, and KCl increased its activity slightly. The divalent and trivalent metal ions including Cu2+ , Ni2+ , Zn2+ , Mn2+ , Al3+ and Fe3+ significantly inhibited its activity, while Mg2+ did not. CgkP remained 70% of original activity after being incubated at 40℃ for 48 h, and remained 80% of the activity after being incubated at 45℃ for 1 h. It exhibited endo-κ-carrageenase activity, mainly depolymerizing the κ-carrageenan into disaccharide and tetrasaccharide. | 4 | Light-triggered NO-releasing nanoparticles for treating mice with liver fibrosis显示文摘Liver fibrosis, resulting from chronic liver damage and characterized by the accumulation of extracellular matrix (ECM) proteins, is a characteristic of most types of chronic liver diseases. The activation of hepatic stellate cells (HSC) is considered an essential pathological hallmark in liver fibrosis. Although nitric oxide (NO) can effectively induce HSC apoptosis, the systemic administration of NO is ineffective and may cause severe complications such as hypotension. To overcome this limitation, nanoparticles were designed to target HSCs and release NO locally under the exposure of near infrared light (NIR). To achieve this, upconversion nanoparticle (UCNP) cores were enveloped in mesoporous silica shells (UCNP@mSiO2), which were modified with hyaluronic acid (HA-UCNP@mSiO2) and Roussin’s black salt (RBS). HA molecules recognize and bind to CD44 proteins, which are overexpressed on activated HSCs. Under exposure to a 980-nm NIR laser, the UCNP cores convert the 980-nm wavelength into ultraviolet (UV) light, which then energizes the RBS (NO donors), resulting in an efficient release of NO inside of the HSCs. Once released, NO triggers HSC apoptosis and reverses the liver fibrosis. | 5 | Serum Metabolomic Characteristics of Primary Dysmenorrhea Rat Model Induced by Estradiol Benzoate Combined with Oxytocin显示文摘[Objectives]To investigate the serum metabolomic characteristics of primary dysmenorrhea rat model induced by estradiol benzoate combined with oxytocin,and to reveal its material basis.[Methods]20 female SD rats were randomly divided into control group and model group.The primary dysmenorrhea rat model was established by subcutaneous injection of estradiol benzoate for 10 consecutive days and intraperitoneal injection of oxytocin on the last day.The serum samples of rats in control group and model group were collected by ultra-performance liquid chromatography(UPLC)quadrupole time-of-flight mass spectrometry(Q-TOF-MS).The differential metabolites were identified by multivariable pattern recognition method and endogenous metabolite database,and the metabolic pathways were enriched by Metaboanalyst 5.0 platform.[Results]There were significant differences in serum metabolic profiles between the two groups.A total of 36 potential biomarkers of primary dysmenorrhea including L-tyrosine,glycocholic acid,citric acid,palmitoyl carnitine and cholesterol were screened and identified,mainly involving metabolic pathways such as phenylalanine,tyrosine and tryptophan biosynthesis,glycerophospholipid metabolism,and primary bile acid biosynthesis.[Conclusions]The serum metabolic profile of primary dysmenorrhea rats deviates significantly from that of healthy rats,and there are multiple metabolic pathway disorders,which are mainly related to phenylalanine,tyrosine and tryptophan biosynthesis,glycerophospholipid metabolism,and primary bile acid biosynthesis. | Zeming ZHANG Wei LIU Linna JIA Jiashuo JU Zilong QI Shuo LIU Jie WANG Zhiping NIU Hui XIONG | 2022 | Medicinal Plant2022,13,5: | 0 |
