| 1 | Generation and Regulation of CD8^+ Regulatory T Cells显示文摘Research into the suppressive activity of CD4+FoxP3+ T regulatory cells (Treg) has defined a sublineage of CD4+ cells that contribute to self-tolerance and resistance to autoimmune disease. Much less attention has been given to the potential contribution of regulatory sublineages of CD8+ cells. Analysis of a small fraction of CD8+ cells that target autoreactive CD4+ cells through recognition of the MHC class Ib molecule Qa-1 in mouse and HLA-E in human has revitalized interest in CD8+ Treg. | 2 | Therapeutic efficacy of anti-CD19 CAR-T cells in a mouse model of systemic lupus erythematosus显示文摘Dysregulated B-cell activation plays pivotal roles in systemic lupus erythematosus(SLE),which makes B-cell depletion a potential strategy for SLE treatment.The clinical success of anti-CD19 CAR-T cells in treating B-cell malignancies has attracted the attention of researchers.In this study,we aimed to investigate the feasibility of applying anti-CD19 CAR-T cell therapy to SLE treatment in a mouse disease model.We constructed murine anti-CD19 CARs with either CD28 or 4-1BB as the intracellular costimulatory motif and evaluated the therapeutic function of the corresponding CAR-T cells by infusing them into MRL-lpr mice.Furthermore,anti-CD19 CAR-T cells were transferred to MRL-lpr mice before the onset of disease to determine their role in SLE prevention.According to our observations,compared with antibody treatment,the adoptive transfer of our anti-CD19 CAR-T cells showed a more sustained B-cell-depletion effect in MRL-lpr mice.The transfer of syngeneic anti-CD19 CAR-T cells not only prevented disease pathogenesis before the onset of disease symptoms but also displayed therapeutic benefits at a later stage after disease progression.We also tried to optimize the treatment strategy and found that compared with CAR-T cells with the CD28 costimulatory motif,CAR-T cells with the 4-1BB costimulatory motif showed better therapeutic efficiency without cell enrichment.Taken together,these results show that anti-CD19 CAR-T cell therapy was effective in the prevention and treatment of a murine model of SLE,indicating its potential for clinical use in patients. | Xuexiao Jin Qin Xu Chengfei Pu Kaixiang Zhu Cheng Lu Yu Jiang Lei Xiao Yongmei Han Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,8: | 12 |
| 3 | Determination of the coefficient of rolling friction of irregularly shaped maize particles by using discrete element method显示文摘The coefficient of rolling friction is a foundation parameter for conducting particles simulation,however,which of irregularly shaped maize seeds is difficult to measure.Furthermore,the coefficient of rolling friction between the simulation particles and the actual seeds is inconsistent due to the shaped difference of model and different position of gravity center.This paper use two methods to determinate the coefficient of rolling friction based on discrete element method(DEM)and physical experiments.Three types of maize models from five different shaped maize samples(including horse-tooth shape,spherical cone shape,spherical shape,oblate shape,irregular shape)were developed with the help of slice modeling and 3D modeling technology.Aluminum cylinder container is used to arrange the simulation experiments of angle of repose with taking the coefficient of rolling friction as independent variables and the simulation angle of repose as target values.After predicting detailed the coefficient of rolling friction(including horse-tooth shape,spherical cone shape,spherical shape,between horse-tooth shape and spherical cone shape,between horse-tooth shape and spherical shape,between spherical shape and spherical cone shape maize models),and forecasting a unified the coefficient of rolling friction among horse-tooth shape,spherical cone shape and spherical shape maize models,two types of materials(aluminum cylinder container and organic glass container)were used to validate the difference the angle of repose between the simulation maize models and actual maize seeds.Results show the relative error of the angle of repose between the maize models controlled by the coefficient of rolling friction through the detailed method and the actual maize seeds is 0.22%,0.33%in aluminum cylinder,organic glass container,respectively.The relative error of the angle of repose between the simulation maize models controlled by the coefficient of rolling friction through the united method and actual maize seeds is 2.47%,2.97%in aluminum cylinder,organic glass container,respectively.Although the difference of the angle of repose between two method is smaller,the detailed method is better.Moreover,From the accumulation process of the angle of repose we found that the difference on the contacts number between maize models and bottom plate,the change curve of the rotational kinetic energy,the potential energy of maize models controlled by the coefficient of rolling friction through the detailed and the united method are evidently.We can choose a better method to predict the coefficient of rolling friction of maize seeds according to the application situation and investigation objective of irregular maize seeds.The results can provide a theoretical basis for designing and optimizing the structure of the seed-metering machine with DEM. | Linrong Shi Wuyun Zhao Bugong Sun Wei Sun | 2020 | International Journal of Agricultural and Biological Engineering2020,13,2: | 5 |
| 4 | Effects of Structural Parameters on the Poisson’s Ratio and Compressive Modulus of 2D Pentamode Structures Fabricated by Selective Laser Melting显示文摘Metamaterials have been receiving an increasing amount of interest in recent years. As a type of metamaterial, pentamode materials (PMs) approximate the elastic properties of liquids. In this study, a finite-element analysis was conducted to predict the mechanical properties of PM structures by altering the thin wall thicknesses and layer numbers to obtain an outstanding load-bearing capacity. It was found that as the thin wall thickness increased from 0.15 to 0.45 mm, the compressive modulus of the PM structures increased and the Poisson’s ratio decreased. As the layer number increased, the Poisson’s ratio of the PM structures increased rapidly and reaches a stable value ranging from 0.50 to 0.55. Simulation results of the stress distribution in the PM structures confirmed that stress concentrations exist at the junctions of the thin walls and weights. For validation, Ti–6Al–4V specimens were fabricated by selective laser melting (SLM), and the mechanical properties of these specimens (i.e., Poisson’s ratio and elastic modulus) were experimentally studied. Good consistency was achieved between the numerical and experimental results. | 6 | Investigation of interaction effect between static and rolling friction of corn kernels on repose formation by DEM显示文摘The coefficient of static friction(SF),the coefficient of rolling friction(RF)for particles are two key parameters affecting the repose angle formation and flow characteristics.In this paper,the interaction effects of SF and RF on the formation process of corn repose angle was investigated by the discrete element method.Firstly,five shape kinds of corn models(horse tooth,spherical cone,spheroid,oblate,and irregular shape)were established.Secondly,aluminum cylinder and organic glass box were used to conduct the simulation experiments with taking SF and RF as independent factors and seeing the repose angle as dependent value.Based on simulation results the regression equations were established.Simulation results showed the relation between two factors and the rotational kinetic energy is not nonlinear,and SF does not significantly restrict the flow of corn models after increasing the flow direction,and the effect of SF on the contact number between corns and the bottom plate is remarkable,while the effect of RF on the contact number is not remarkable.Finally,the interaction effect of two factors on the repose angle was analyzed by variance analysis and results showed SF and RF all have a significant impact on the repose angle.Moreover,their interaction effect has an impact on the repose angle. | Linrong Shi Xiaoping Yang Wuyun Zhao Wei Sun Guanping Wang Bugong Sun | 2021 | International Journal of Agricultural and Biological Engineering2021,14,5: | 2 |
| 7 | Priming of NLRP3 inflammasome activation by Msn kinase MINK1 in macrophages显示文摘The nucleotide-binding domain,leucine-rich-repeat containing family,pyrin domain-containing 3(NLRP3)inflammasome is essential in inflammation and inflammatory disorders.Phosphorylation at various sites on NLRP3 differentially regulates inflammasome activation.The Ser725 phosphorylation site on NLRP3 is depicted in multiple inflammasome activation scenarios,but the importance and regulation of this site has not been clarified.The present study revealed that the phosphorylation of Ser725 was an essential step for the priming of the NLRP3 inflammasome in macrophages.We also showed that Ser725 was directly phosphorylated by misshapen(Msn)/NIK-related kinase 1(MINK1),depending on the direct interaction between MINK1 and the NLRP3 LRR domain.MINK1 deficiency reduced NLRP3 activation and suppressed inflammatory responses in mouse models of acute sepsis and peritonitis.Reactive oxygen species(ROS)upregulated the kinase activity of MINK1 and subsequently promoted inflammasome priming via NLRP3 Ser725 phosphorylation.Eliminating ROS suppressed NLRP3 activation and reduced sepsis and peritonitis symptoms in a MINK1-dependent manner.Altogether,our study reveals a direct regulation of the NLRP3 inflammasome by Msn family kinase MINK1 and suggests that modulation of MINK1 activity is a potential intervention strategy for inflammasomerelated diseases. | Kaixiang Zhu Xuexiao Jin Zhexu Chi Sheng Chen Songquan Wu Richard D.Sloan Xuai Lin Dante Neculai Di Wang Hu Hu Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,10: | 2 |

| 9 | Study on mechanical properties of soil-rock mixture of various compactness subjected to freeze-thaw cycles显示文摘The soil-rock mixture,a collection of soil particles and rock blocks,is inherently heterogeneous and anisotropic due to significant particle size and material strength differences.This study conducts triaxial tests on soil-rock mixture samples of various compactness subjected to varying freeze-thaw cycles.A mesoscopic simulation is carried out by particle flow code(PFC)to analyze the effects of freeze-thaw cycles on the mechanical properties of soil and rock particles.The results show that the mechanical properties of the soil-rock mixture under freeze-thaw cycles are greatly affected by the initial compaction.In general,when the degree of compaction is higher,the influence of freeze-thaw cycles on the soil-rock mixture is greater.The stress-strain curves of the samples with different compactness demonstrate strain-softening behavior.The freeze-thaw cycles greatly influence the failure strength of the samples with a higher degree of compaction but have little impact on the samples with a lower degree of compaction.On the microscopic level,during freeze-thaw cycles,the pore volume in the highly compacted sample is too small to accommodate the volume expansion from ice crystal formation,causing significant strength loss among the soil and rock particles and deterioration of the macroscopic properties of the soil-rock mixture. | Zhong Zhou HaoHui Ding WenYuan Gao LinRong Xu | 2021 | Research in Cold and Arid Regions2021,13,5: | 2 |

| 19 | JAK3 mediates c-myc gene expression induced by interleukin-2显示文摘C myc gene expression can be rapidly induced by IL 2 through intracellular signal transduction which is triggered by the interaction between IL 2 and its receptor (IL 2R). JAK3 which associates to the intracellular domain of IL2R γ may play a critical role in this process. To reveal the action of JAK3 in c myc induction, a chimeric receptor gene IL2R α/γ/Δ NJAK3 is constructed which consists of extracellular domain derived from IL 2R α subunit (IL2R α), the transmembrane sequence derived from IL2R γ and the cytoplasmic domain derived from the catalytic domain of JAK3 (Δ NJAK3), and then transfected this chimeric gene into mouse fibroblast cell line L929 β which had been transfected with IL2R β gene and stably expressed IL2R β in high level. In the transfectants coexpressing IL2R β and α/γ/Δ NJAK3, the stimulation of IL2 could intensively induce c myc gene expression.
