1DYRK1A suppression restrains Mcl-1 expression and sensitizes NSCLC cells to Bd-2 inhibitors显示文摘Objective:Mcl-1 overexpression confers acquired resistance to Bcl-2 inhibitors in non-small cell lung cancer(NSCLC),but no direct Mcl-1 inhibitor is currently available for clinical use.Thus,novel therapeutic strategies are urgently needed to target Mcl-1 and sensitize the anti-NSCLC activity of Bcl-2 inhibitors.Methods:Cell proliferation was measured using sulforhodamine B and colony formation assays,and apoptosis was detected with Annexin V-FITC staining.Gene expression was manipulated using siRNAs and plasmids.Real-time PCR and Western blot were used to measure mRNA and protein levels.Immunoprecipitation and immunofluorescence were used to analyze co-localization o f dual specificity tyrosine-phosphorylation-regulated kinase 1A(DYRK1A)and Mcl-1.Results:Suppression of DYRK1A resulted in reduced Mcl-1 expression in NSCLC cells,whereas overexpression of DYRK1A significantly increased Mcl-1 expression.Suppression of DYRK1A did not alter Mcl-1 mRNA levels,but did result in an accelerated degradation o f Mcl-1 protein in NSCLC cells.Furthermore,DYRK1A mediated proteasome-dependent degradation o f Mcl-1 in NSCLC cells,and DYRK1A co-localized with Mcl-1 in NSCLC cells and was co-expressed with Mcl-1 in tumor samples from lung cancer patients,suggesting that Mcl-1 may be a novel DYRK1A substrate.We showed that combined therapy with harmine and Bcl-2 antagonists significantly inhibited cell proliferation and induced apoptosis in NSCLC cell lines as well as primary NSCLC cells.Conclusions:Mcl-1 is a novel DYRK1A substrate,and the role of DYRK1A in promoting Mcl-1 stability makes it an attractive target for decreasing Bcl-2 inhibitor resistance.Yangling Li Dongmei Zhou Shuang Xu Mingjun Rao Zuoyan Zhang Linwen Wu Chong Zhang Nengming Lin 2020Cancer Biology & Medicine2020,17,2:2
2Multifunctional interstitial-carbon-doped FeCoNiCu high entropy alloys with excellent electromagnetic-wave absorption performance显示文摘Electromagnetic-wave absorbing(EMA)materials that have efficient absorption performances,great me-chanical properties and chemical stability are rare and yet essential for communication security and pro-tection.Herein,flaky interstitial-carbon-doped FeCoNiCu high entropy alloys(HEAs)as novel EMA ma-terials were successfully prepared by high-energy ball-milling method.Interstitial-carbon doping as a modulating approach impacted the phase forming,morphology and electromagnetic properties of Fe-CoNiCu HEAs.Impedance matching was significantly optimized via tuning interstitial carbon contents.The carbon-doped FeCoNiCu HEAs with appropriate carbon contents delivered superior EMA performance compared with other HEAs EMA materials.Strong reflection loss as low as-61.1 dB in the Ku band,broad effective absorption bandwidth of 5.1 GHz was achieved for FeCoNiCuC_(0.04).Moreover,the carbon-doped FeCoNiCu HEAs exhibited excellent mechanical hardness and chemical stability.This work not only suggests that interstitial-carbon doping is an available approach to tuning electromagnetic properties of HEAs,but also presents carbon-doped FeCoNiCu HEAs as promising EMA materials for civilian and mili-tary due to the efficient absorption,broad bandwidth,great durability and stability.Jianping Yang Linwen Jiang Zhonghao Liu Zhuo Tang Anhua Wu 2022Journal of Materials Science & Technology2022,,18:1

4Bacterial community composition in gut content and ambient sediment of two tropical wild sea cucumbers(Holothuria atra and H.leucospilota)显示文摘Sea cucumbers play an important role in sediment bioturbation in coral reef and rocky intertidal ecosystems,and bacteria were a commonly-reported component of holothuroid diets.Bacterial community composition in the foregut and hindgut of two common tropical sea cucumbers(Holothuria atra and H.leucospilota)and the ambient surface sediment were investigated using high throughput sequencing based on 16S rRNA gene analysis.A total of 5584 operational taxonomic units(OTUs)were obtained from 25 samples based on a 97%threshold,and more than half of the OTUs(n=3694,66.2%)were shared by the gut contents of two species of sea cucumbers and surrounding sediments.Bacterial richness and diversity in sediment samples were signifi cantly higher than those in the gut content samples(P<0.05).Proteobacteria was the predominant phylum in most samples showing 45.69%±8.61%,70.09%±4.03%,45.88%±5.38%,and 55.19%±0.79%reads in the foregut of H.leucospilota,hindgut of H.leucospilota,hindgut of H.atra,and sediment libraries,respectively,but Bacteroidetes was the predominant phylum with the relative content of 34.98%±5.52%in the foregut of H.atra.Among the dominant genera,reads related to the genera Anderseniella,Ilumatobacter,and Ruegeria were detected in all the gut contents and sediment libraries.A comparison of gut bacteria community between the two species of sea cucumbers suggested that H.atra had stronger feeding preference than H.leucospilota,and the same types of microbes escaped digestion of the two sea cucumber species.Obvious diff erent bacterial community composition in the foreguts of the two species of sea cucumbers and the surrounding sediments might result from the animal’s selective feeding for sediment patches.Bacterial community structure in hindgut contents of H.atra and H.leucospilota both diff ered clearly from adjacent sediments,which indicated feeding activity of deposit-feeding sea cucumbers could change the sedimental bacterial composition.In conclusion,from the perspective of bacteria,sea cucumber H.atra and H.leucospilota had diff erent feeding preferences,yet they could both aff ect bacterial composition in sediments by feeding activity.The motivation for selective feeding and sea cucumber-sediment interaction might be explored in the future.Fei GAO Yue ZHANG Peilin WU Mengling CHEN Linwen HE Qiang XU Aimin WANG 2022Journal of Oceanology and Limnology2022,40,1:0
