1Stable carbon and hydrogen isotopic fractionations of alkane compounds and crude oil during aerobically microbial degradation显示文摘Normal alkane compounds dodecane, penta- decane, hexadecane, octadecane, tetracosane, isoprenoid alkane pristane and a crude oil sample were aerobically bio- degraded with a pure bacterial strain GIM2.5 and white rot fungus Phanerochaete Chrysosporium-1767 to monitor the kinetic fractionation of the molecular stable carbon (δ C) 13 and hydrogen (δ D) isotopes in the course of biodegradation. Both δ C (V-PDB ) and δ D (V-SMOW) remained stable for 13 the standard alkane compounds and n-alkane components (from n-C13 to n-C25) of the crude oil, generally varying in the range of ±0.5‰ and ±5‰ respectively, within the range of the instrumental precisions, especially for those molecularly heavier than n-C16 during microbial degradation. These re- sults indicate that molecular stable carbon and hydrogen isotopic fingerprints can be promising indicators for tracing the sources of petroleum-related contaminants in the envi- ronment, especially in the case of severe weathering when they are difficult to be unambiguously identified by the chemical fingerprints alone.PENGXianzhi ZHANGGan CHENFanzhong LIUGuoqing 2004Chinese Science Bulletin2004,49,24:2
2Characterization and mapping of a lesion mimic mutant in rice (Oryza sativa L.)显示文摘A rice initiation-type lesion mimic mutant (imi) was identified, which was isolated from an indica rice Zhongxian 3037 through γ radiation mutagenesis. Trypan blue staining and sterile culture revealed that the mutant spontaneously developed lesions on the leaves in a develop mentally regulated and light-dependent manner. Genetic analysis indicated that the lesion mimic trait was controlled by a single resessive locus. Using public molecular markers and an F2 population derived from lmi and 93-11, we mapped the lmi locus to the short arm of chromosome 8, nearby the centromere, between two SSR markers RM547 and RM331. The genetic distance was 1.2 and 3.2 Cm, respectively. Then according to the public rice genomic sequence between the two SSR markers, lmi was further finely tagged by three CAPS markers: C4135-8, C4135-9 and C4135-10. 5Measurements of leucocyte membrane elasticity based on the optical tweezers显示文摘A 1-mm in diameter polystyrene bead coated with lectin is trapped by optical tweezers which is formed by a focused laser beam (740 nm). A leucocyte adhered to the bottom of sample cell is chosen to close to the trapped bead. When their stable combination is confirmed, the leucocyte is displaced by moving the sample stage. A tether is then formed between the trapped bead and the membrane. The force acting on the tether is measured by a detector equipped on the optical tweezers system. The deformation of the membrane is diverse in different contact conditions. When the contact area is small, the tether is very thin and the force on it is 7.4 pN (10-12 N), while the tether is much thicker un-der a larger contact area and its corresponding force is about 14 pN.
