1The effects of water flow and temperature on thermal regime around a culvert built on permafrost显示文摘Temperature and water flow through a culvert beneath the Alaska Highway near Beaver Creek,Yukon,were measured at hourly intervals between June and October 2013.These data were used to simulate the effect of the culvert on the thermal regime of the road embankment and subjacent permafrost.A 2-D thermal model of the embankment and permafrost was developed with TEMP/W and calibrated using field observations.Empirical relations were obtained between water temperatures at the entrance to the culvert,flow into the culvert,and water temperatures inside the structure.Water temperatures at the entrance and inside the culvert had a linear relation,while water temperatures inside the culvert and water flow were associated by a logarithmic relation.A multiple linear regression was used to summarize these relations.From this relationship,changes in the flow rate and water temperatures at the entrance of the culvert were simulated to obtain predicted water temperatures in the culvert.The temperatures in the culvert were used in the thermal model to determine their effects on the ground thermal regime near the culvert.Variation of ±10% in water flow rate had no impact on the thermal regime underneath the culvert.Variation of water temperature at the entrance of the culvert had a noticeable influence on the thermal regime.A final simulation was conducted without insulation beneath the culvert.The thaw depth was 30 cm with insulation,and 120 cm without insulation,illustrating the importance of insulation to the ground thermal regime.Loriane Périer Guy Doré C.R.Burn Loriane Perier Guy Dore C. 15Protective effects of honey compound syrup on busulfan-induced azoospermia in male rats显示文摘Objective:To evaluate the protective effects of honey compound syrup on sperm count and testis tissue in rats.Methods:Thirty rats were randomly assigned to five groups.The control group received 1 mL normal saline with dimethyl sulfoxide intraperitoneally;the busulfan group received busulfan 10 mg/kg body weight at the first and twenty-first days of the experiment via intraperitoneal injection;the last three groups received busulfan 10 mg/kg body weight to induce azoospermia,and then received 1.0,1.5,or 2.0 mg/kg honey compound syrup,respectively,after induction of azoospermia.After administration,the testis and epididymis of all rats were removed.Then,reproductive organ weight and sperm parameters(sperm concentration,epididymal sperm reserve and daily sperm production)were measured.After hematoxylin-eosin staining,seminiferous tubule cells and diameters were assessed.Results:Busulfan damaged the testis tissue and impaired spermatogenesis.Administration of honey compound syrup in three doses improved testis tissue and spermatogenesis.The protective effects of honey compound syrup may relate to the antioxidant properties of honey and other compounds in this syrup.Conclusions:Administration of honey compound syrup could be an ameliorative agent for the side effects of chemotherapy drugs such as busulfan on the male reproductive system.Seyyed Shamsadin Athari Keivan Lorian Haniye Kashafroodi Saadat Ghafarzadeh Rasool Choopani 2021Asian pacific Journal of Reproduction2021,10,6:0
16Adult human liver slice cultures:Modelling of liver fibrosis and evaluation of new anti-fibrotic drugs显示文摘BACKGROUND Liver fibrosis can result in end-stage liver failure and death.AIM To examine human liver fibrogenesis and anti-fibrotic therapies,we evaluated the three dimensional ex vivo liver slice(LS)model.METHODS Fibrotic liver samples(F0 to F4 fibrosis stage according to the METAVIR score)were collected from patients after liver resection.Human liver slices(HLS)were cultivated for up to 21 days.Hepatitis C virus(HCV)infection,alcohol(ethanol stimulation)and steatosis(palmitate stimulation)were examined in fibrotic(F2 to F4)liver slices infected(or not)with HCV.F0-F1 HLS were used as controls.At day 0,either ursodeoxycholic acid(choleretic and hepatoprotective properties)and/or α-tocopherol(antioxidant properties)were added to standard of care on HLS and fibrotic liver slices,infected(or not)with HCV.Expression of the biomarkers of fibrosis and the triglyceride production were checked by quantitative reverse transcription polymerase chain reaction and/or enzymelinked immunosorbent assay.RESULTS The cultures were viable in vitro for 21 days allowing to study fibrosis inducers and to estimate the effect of anti-fibrotic drugs.Expression of the biomarkers of fibrosis and the progression to steatosis(estimated by triglycerides production)was increased with the addition of HCV and/or ethanol or palmitate.From day 15 of the follow-up studies,a significant decrease of both transforming growth factorβ-1 and Procol1A1 expression and triglycerides production was observed when a combined anti-fibrotic treatment was applied on HCV infected F2-F4 LS cultures.CONCLUSION These results show that the human three dimensional ex vivo model effectively reflects the in vivo processes in damaged human liver(viral,alcoholic,nonalcoholic steatohepatitis liver diseases)and provides the proof of concept that the LS examined model permits a rapid evaluation of new anti-fibrotic therapies when used alone or in combination.Daria Kartasheva-Ebertz Jesintha Gaston Loriane Lair-Mehiri Pierre-Philippe Massault Olivier Scatton Jean-Christophe Vaillant Vladimir Alexei Morozov Stanislas Pol Sylvie Lagaye 2021World Journal of Hepatology2021,13,2:0
