1Comparison of Two Methods to Assess the Size Structure of Phytoplankton Community Assemblages, in Liaodong Bay, China显示文摘Liaodong Bay is one of the largest semi-enclosed bays located in Bohai Sea,in northeast China.It is an important feeding and spawning place,even wintering and breeding migration base for many marine organisms.The size structure of phytoplankton communities is a key feature of marine ecosystems and can be used as an indicator for algae disaster,but the methods to evalute is still challenging.Here,we compared the size-fractionated chlorophyll-a method with a high-throughout sequencing method in an evaluation of the size structure of phytoplankton assemblages in Liaodong Bay,China,in 2014.The proportion of picophytoplankton was estimated at just 19%according to the results based on size-fractionated chlorophyll-a,which likely represents a severe underestimation.The sequencing method confirmed that many kinds of phytoplankton species appeared in more than one size-fractionated filter membrane,with some even dominating where the filter membrane pore size was greater than the individual phytoplankton size.According to the sequencing method,the phytoplankton assemblages in Liaodong Bay were dominated by picophytoplankton(average proportion=44%),followed by the nanophytoplankton(average 32%),a composition that is more consistent with other studies.The sequencing method provides a more robust way to assess the size structure and taxonomic diversity of marine phytoplankton communities.SONG Lun WU Jing DU Jing LI Nan WANG Kun WANG Peng 2019Journal of Ocean University of China2019,18,5:8
2Proteomic Analysis of Macrophages:A Potential Way to Identify Novel Proteins Associated with Activation of Macrophages for Tumor Cell Killing显示文摘One major mechanism through which macrophages effectively kill tumor cells requires cell to cell contact,indicating that certain molecules expressed on cell surface of activated macrophages may mediate the tumoricidal capability. Tumor necrosis factor (TNF) and nitric oxide (NO) are the two classical mediators of tumor cell death. However,evidence of discrepancy is accumulating indicating these known mediators do not appear to account for the broad and potent tumoricidal activity of macrophages. To obtain a full repertoire of tumoricidal activation-associated membrane proteins,we combined one-dimensional SDS-PAGE with capillary liquid chromatography-tandem mass spectrometry (LC-MS/MS). Using this technique,we identified 454 activated macrophage specifically expressed proteins with extremely high confidence,including most known activation markers of macrophages,such as NO synthase (iNOS),Ym1,cyclooxygenase,etc. Membrane bound TNF-α was also identified on activated macrophages. However,it was also detected on thioglycolate elicited macrophages,indicating this molecule may not play a key role in conjugation-dependent tumor cell killing. In contrast,although NO has not been assigned as an effector molecule of conjugation-dependent tumoricidal pathway,iNOS was identified from membrane fraction of activated macrophages,suggesting NO may be involved in conjugation-dependent tumoricidal mechanism,because iNOS association with plasma membrane is ideally suited to deliver NO directly into the contacted tumor cells. 3Highly Efficient Synthesis of Environmentally Friendly Ag-modified TiO2 Nanoflowers to Enhance Photocatalytic Performance显示文摘Ag-modified TiO2 nanoflowers were prepared using a two-step process. The experimental process is green and free from contamination and can be synthesized directly at room temperature. Compared with pure TiO2, Ag-modified TiO2 enhances the absorption of visible light and effectively promotes the detachment of photoelectron pairs, Ag-TiO2 has a significantly enhanced visible light response activity to photodecomposition of methyl orange(MO). It is shown that the strong interaction between Ag nanoparticles and TiO2 enhances the photocatalytic activity of TiO2 nanoflowers. The self-made open-air reactor was used to test the photocatalytic performance of different samples. The results showed that Ag-modified TiO2 nanoflowers had excellent photodegradation ability. 7Numerical Simulation of Bohai Oil Spill in the Winter Sea Ice Period显示文摘The Bohai Sea is a seasonal icy sea area that has the lowest latitude of any sea experiencing icing in the northern hemisphere, and simulation studies on oil spills during its sea ice period are the key to analyzing winter oil spill accidents. This study applied the three-dimensional free surface to establish a high-resolution hydrodynamic model and simulate tidal distributions in the Bohai Sea. Then, the oil spill model of the open sea area and thermodynamic model were combined to establish a numerical model for the Bohai oil spill during the winter sea ice period. The hydrodynamic model and sea ice growth and melting model were verified, and the parameters were adjusted based on the measured values, which indicate that the numerical model established in this paper is of high accuracy,stability and ubiquity. Finally, after checking the calculations repeatedly, the diffusion coefficient for the Bohai Sea was determined to be 1.0×10^(–7 )m^2/s. It is better that the comprehensive weathering attenuation coefficient is lower than that of a non-winter oil spill, with 1.3×10^(–7 )m^2/s being the most appropriate coefficient. 9A two-dimensional simulation method for investigating charge transport behavior in 3-D charge trapping memory显示文摘This work presents a self-consistent two-dimensional(2-D) simulation method with unified physical models for different operation regimes of charge trapping memory. The simulation carefully takes into consideration the tunneling process, charge trapping/de-trapping mechanisms, and 2-D drift-diffusion transport within the storage layer. A string of three memory cells has been simulated and evaluated for different gate stack compositions and temperatures. The simulator is able to describe the charge transport behavior along bitline and tunneling directions under different operations. 14Isolation, Identification, and Quantitative Determination of Saponin in Apostichopus japonicus by HPLC-DAD显示文摘Apostichopus japonicus is an important invertebrate that is widely used as a tonic food in Asian countries.The purpose of this study is to purify and identify a class of compound,the saponins,from the body wall of A.japonicus,and to establish a new me-thod to determine the quantity of saponins in the sea cucumber.In this study,the saponins of A.japonicus,cladoloside A(CA),were ob-tained from 80%ethanol extract by column chromatography for the first time and were characterized using the spectral method.The resulting purified saponins were then profiled using ^(1)HNMR,^(13)CNMR,and ESI-MS,which revealed the CA molecular formula of C_(53)H_(82)O_(2) and contained a triterpenoid backbone,a methylglucopyranosyl moiety,a quinovopyranosyl,and two xylopyranosyls.A me-thod for the quantitative determination of CA,comprising microwave-assisted extraction,high-performance liquid chromatography,and diode array detector method,was established.Extraction efficiency was optimized by changing microwave power,extraction sol-vent,volume,time,and temperature.Results showed that under the optimum conditions(extraction time of 10 min,temperature of 45℃,and solvent of 25 mL 70%ethanol under 400 W),the detection limit of CA was 0.0015 mg mL^(−1),and the recoveries of CA from samples at spiking levels of 10,20,and 50μg g^(−1) ranged from 90.1%-104%.The proposed method was successfully applied to ana-lyze the saponins in different tissues of A.japonicus collected in different seasons.The method developed in this study can provide quantitative technical support for the quality control of A.japonicus.LIU Guiying WANG Xuda GE Kun WU Jinhao WANG Zhaohui DU Jing SONG Lun ZHOU Zunchun 2022Journal of Ocean University of China2022,21,2:1

16Microsurgical Resection of Pituitary Adenoma via Single-Nostril Transsphenoidal Approach显示文摘OBJECTIVE To explore the methods and experience of thesingle-nostril transsphenoidal approach for treating pituitaryadenomas.METHODS We retrospectively analyzed 46 patients whohad pituitary tumors and received surgery via the singlenostriltranssphenoidal approach and observed the effects andcomplications of surgery. The specific surgical methods are: a nasalspeculum is inserted slowly through the right nostril towards theanterior wall of the sphenoid sinus. A 1.5 cm incision is made intothe nasal mucosa in the right nasal cavity at the level of the middlenasal turbinate. By fracturing the bony septum, a space is formedbetween the bilateral nasal mucosa and the bony septum of thesphenoid sinus. Then, the inside of the sphenoid sinus is exposed.The remaining part of the bony septum, the anterior sphenoidsinus wall, and the sphenoid mucosa are gradually removed. Theanterior sphenoidotomy is less than 1.5 cm wide. After confirmingthe tumor by dural puncture, a cross incision of the dura is made,and the tumor is slowly removed by curette. The sella is usuallycollapsed and visible after the total tumor removal. When thetumor is resected satisfactorily, gelatin sponges are placed into theoperative cavity to stop bleeding.RESULTS Postoperative MRI scans revealed that among the 46cases, total resection of the tumor was achieved in 34 cases andsubtotal in 12. No deaths or disability occurred, and the hormonelevels of almost all patients improved. 18Establishment of blood glucose control model in diabetic mice显示文摘AIM:To explore the established method of the diabetic mouse blood glucose control model and preliminar y observation of its influence on the retinas of diabetic mice.METHODS:The db/db BKS-DB(Lepr^(ko/ko))mice were randomly divided into two groups:the poor blood glucose control group(PG group,n=18)and the stable blood glucose control group(SG group,n=12),with BKS-DB(Lepr^(wt/wt))as the normal blood glucose control group(NG group,n=18).According to the blood glucose values for 5 intervals which were monitored during the period of adaption,the PG group was injected with insulin aspart twice daily,fasted for 2h and then returned to normal.The SG group was injected with insulin aspart twice and insulin glargine once daily and fed with a quantitative ration.Fundus images were collected after eight weeks.The glycosylated hemoglobin(Hb A1c),mean blood glucose level(MBG),standard deviation of blood glucose(SDBG),coefficient of variation of blood glucose(CVBG),and mean amplitude of glycemic excursion(MAGE)in each group were examined and calculated.RESULTS:The Hb A1c,MBG,SDBG,CVBG,and MAGE levels in the PG group were significantly higher than those in the NG and SG groups(all P<0.05).MBG,SDBG,CVBG,and MAGE levels in the SG group were higher than those in the NG group(all P<0.05).There was no significant difference in Hb A1c levels between the NG and SG groups(P>0.05).Preliminary observation of fundus images in the PG group and SG groups showed scattered retinal bleeding spots,while bleeding was more obvious in the PG group.CONCLUSION:The blood glucose control model of type 2 diabetes mellitus mice can be successfully established by subcutaneous injection of insulin aspart insulin glargine and rationed food,which is valuable for studying the mechanism of blood glucose fluctuations in diabetic complications in vivo.Cai-Hua Rao Lun Liu Jian Gao Zi-Hao Du Chen Gao 2022International Journal of Ophthalmology(English edition)2022,15,12:0
19One-step,in situ formation of WN-W_(2)C heterojunctions implanted on N doped carbon nanorods as efficient oxygen reduction catalyst for metal-air battery显示文摘Transition metal nitrides and carbides have attracted intensive attentions in metal-air battery application due to their metallic electron transport behavior and high chemical stability toward the oxygen reduction reaction(ORR).Herein,the polyoxometalate@polyaniline composite derived WN-W_(2)C heterostructured composite(WN-W_(2)C@pDC)has been fabricated through an in situ nitriding-carbonization strategy,with WN-W_(2)C nanoparticles implanted on N doped carbon nanorods.Asfabricated WN-W_(2)C@pDC demonstrates prominent electrocatalytic performance towards ORR and excellent cycling stability in metal-air battery,which possesses positive half-wave potential and large diffusion limiting current density(0.81 V and 5.8 mA·cm^(-2)).Moreover,it demonstrates high peak power density of 157.4 mW·cm^(-2)as Al-air primary cathode and excellent stability at the discharge-charge test(>500 h)of Zn-air secondary battery.The excellent activity and durability of WNW_(2)C@pDC catalyst should be attributed to the combined effect of intimate WN-W_(2)C heterointerfaces,unique embedded nanoparticles structure,and excellent electrical media of N doped carbon,confirmed by a series of contrast experiments.Yue Du Wenxue Chen Lina Zhou Rui Hu Shizhu Wang Xueqing Li Yunlong Xie Lun Yang Yisi Liu Zhenhui Liu 2023Nano Research2023,16,7:0
20Sonic hedgehog elevates N-myc gene expression in neural stem cells显示文摘Proliferation of neural stem cells is regulated by the secreted signaling molecule sonic hedgehog. In this study, neural stem cells were infected with recombinant adeno-associated virus expressing sonic hedgehog-N-enhanced green fluorescent protein. The results showed that over expression of sonic hedgehog in neural stem cells induced the increased expression of Gli1 and N-myc, a target gene of sonic hedgehog. These findings suggest that N-myc is a direct downstream target of the sonic hedgehog signal pathway in neural stem cells.
