2Extracellular-superoxide dismutase DNA methylation promotes oxidative stress in homocysteine-induced atherosclerosis显示文摘In the present study,we investigate the effect of homocysteine(Hcy)on extracellular-superoxide dismutase(ECSOD)DNA methylation in the aorta of mice,and explore the underlying mechanism in macrophages,trying to identify the key targets of Hcy-induced EC-SOD methylation changes.ApoE--mice are fed different diets for 15 weeks,EC-SODand DNAmethyltransferase1(DNMT1)expression levels aredetected byRT-PCRandwestern blot analysis.EC-SOD methylation levels are assessed by ntMS-PCR.After EC-SOD overexpression or knockdown in macrophages,following the transfection of macrophages with pEGFP-N1-DNMT1,the methylation levels of ECSOD are detected.Our data show that the concentrations of Hcy and the area of atherogenic lesions are significantly increased in ApoE--mice fed with a high-methionine diet,and have a positive correlation with the levels of superoxide anions,which indicates that Hcy-activated superoxide anions enhance the development of atherogenic lesions.EC-SOD expression is suppressed by Hcy,and the content of superoxide anion is increased when EC-SOD is silenced by RNAi in macrophages,suggesting that EC-SOD plays a major part in oxidative stress induced by Hcy.Furthermore,the promoter activity of EC-SOD is increased following transfection with the-1/-110o fragment,and EC-SOD methylation level is significantly suppressed by Hcy,and more significantly decreased upon DNMT1 overexpression.In conclusion,Hcy may alter the DNA methylation status and DNMT1 acts as the essential enzyme in the methyl transfer process to disturb the status of EC-SOD DNA methylation,leading to decreased expression of Ec-sODandincreasedoxidativestressandatherosclerosis.Shengchao Ma Guanjun Lu Qing Zhang Ning Ding Yuzhen Jie Hui Zhang Lingbo Xu Lin Xie Xiaoling Yang Huiping Zhang Yideng Jiang 2022Acta Biochimica et Biophysica Sinica2022,54,9:1
3Sensitive and rapid detection of two toxic microalgae Alexandrium by loop-mediated isothermal amplification显示文摘A loop-mediated isothermal amplification (LAMP) assay was designed and evaluated for rapid detection of the toxic microalgae Alexandrium catenella and A.minutum,which can produce paralytic shellfish poisoning (PSP).Two sets of four specific primers targeting these two species were derived from the sequence of internal transcribed spacer (ITS) of ribosomal DNA.The method worked well in less than an hour under isothermal conditions of 65 C.LAMP specificity was validated in closely related algae as a comparison,suggesting the strict specificity of the LAMP primers.Two visual inspection approaches were feasible to interpret the positive or negative results.The detection limits of A.catenella and A.minutum samples using the LAMP assay were found to be 5.6 and 4.5 pg DNA,respectively.The sensitivity of this LAMP assay was 10 or 100-fold higher than Polymerase Chain Reaction (PCR) method in detecting the two microalgae.These characteristics of species specificity,sensitivity,and rapidity suggest that this method has the potentiality in the monitoring of red tide caused by A.catenella and A.minutum.ZHANG Fengying SHI Yanhong JIANG Keji XU Zhaoli MA Lingbo 2012Acta Oceanologica Sinica2012,31,2:1
4Electrochemical analysis of microRNAs with hybridization chain reaction-based triple signal amplification显示文摘Selective and sensitive detection of trace microRNA is important for early diagnosis of diseases due to its expression level related to diseases.Herein,a triple signal amplification strategy is developed for trace microRNA-21 (miRNA-21) detection by combining with target-triggered cyclic strand displacement reaction (TCSDR),hybridization chain reaction (HCR) and enzyme catalytic amplification.Four DNA hairpins(H1,H2,H3,H4) are employed to form an ultralong double-strand DNA (dsDNA) structure,which is initiated by target miRNA-21.As H3 and H4 are labeled with horseradish peroxidase (HRP),numerous HRPs are loaded on the long dsDNA,producing significantly enhanced electrocatalytic signals in the hydrogen peroxide (H_(2)O_(2)) and 3,3,5,5-tetramethylbenzidine (TMB) reaction strategy.Compared with single signal amplification,the triple signal amplification strategy shows higher electrochemical response,wider dynamic range and lower detection limit for miRNA-21 detection with excellent selectivity,reproducibility and stability.Taking advantage of the triple signal amplification strategy,the proposed electrochemical biosensor can detect miRNA-21 in 10 He La cell lysates,suggesting that it is a promising method for fruitful assay in clinical diagnosis.Jianfeng Ma Lingbo Gong Yingying Cen Lin Feng Yan Su Xingfen Liu Jie Chao Ying Wan Shao Su Lianhui Wang 2023Chinese Chemical Letters2023,34,7:0
