2PGE2 activates EP4 in subchondral bone osteoclasts to regulate osteoarthritis显示文摘Prostaglandin E2(PGE2), a major cyclooxygenase-2(COX-2) product, is highly secreted by the osteoblast lineage in the subchondral bone tissue of osteoarthritis(OA) patients. However, NSAIDs, including COX-2 inhibitors, have severe side effects during OA treatment. Therefore, the identification of novel drug targets of PGE2 signaling in OA progression is urgently needed. Osteoclasts play a critical role in subchondral bone homeostasis and OA-related pain. However, the mechanisms by which PGE2 regulates osteoclast function and subsequently subchondral bone homeostasis are largely unknown. Here, we show that PGE2 acts via EP4 receptors on osteoclasts during the progression of OA and OA-related pain. Our data show that while PGE2 mediates migration and osteoclastogenesis via its EP2 and EP4 receptors, tissue-specific knockout of only the EP4 receptor in osteoclasts(EP4 Lys M) reduced disease progression and osteophyte formation in a murine model of OA. Furthermore, OA-related pain was alleviated in the EP4 Lys M mice, with reduced Netrin-1 secretion and CGRP-positive sensory innervation of the subchondral bone. The expression of plateletderived growth factor-BB(PDGF-BB) was also lower in the EP4 Lys Mmice, which resulted in reduced type H blood vessel formation in subchondral bone. Importantly, we identified a novel potent EP4 antagonist, HL-43, which showed in vitro and in vivo effects consistent with those observed in the EP4 Lys Mmice. Finally, we showed that the Gαs/PI3 K/AKT/MAPK signaling pathway is downstream of EP4 activation via PGE2 in osteoclasts. 3Symmetry properties of Cayley graphs of small valencies on the alternating group A_5显示文摘The normality of symmetry property of Cayley graphs of valencies 3 and 4 on the alternating group A5 is studied. We prove that all but four such graphs are normal;that A5 is not 5-CI. 4Tauroursodeoxycholic acid(TUDCA) inhibits influenza A viral infection by disrupting viral proton channel M2显示文摘Influenza is a persistent threat to human health and there is a continuing requirement for updating antiinfluenza strategies. Initiated by observations of different endoplasmic reticulum(ER) responses of host to seasonal H1N1 and highly pathogenic avian influenza(HPAI) A H5N1 infections, we identified an alternative antiviral role of tauroursodeoxycholic acid(TUDCA), a clinically available ER stress inhibitor, both in vitro and in vivo. Rather than modulating ER stress in host cells, TUDCA abolished the proton conductivity of viral M2 by disrupting its oligomeric states, which induces inefficient viral infection. We also showed that M2 penetrated cells, whose intracellular uptake depended on its proton channel activity,an effect observed in both TUDCA and M2 inhibitor amantadine. 6Fractal characteristics and its application in electromagnetic radiation signals during fracturing of coal or rock显示文摘The present study analyzed the electromagnetic radiation(EMR) time series of the destruction process of coal or rock sample under uniaxial loading and the monitoring process in working face by means of fractal geometry,and results of the correlation dimension change curve of EMR time series were obtained.In the meantime,the current study also sought the fractal characteristic to the EMR signals by contrast to the change curve of EMR signals and explored the precursory phenomenon of rock burst.This paper concluded the main findings as followed:the EMR time series of the destruction process of coal or rock sample under uniaxial loading and the monitoring process in working face corresponded to fractal;the correlation dimension of EMR time series reflected the process of coal or rock damage deformation,that is,the inner damage of coal or rock made a change from random to order.In the field application,the correlation dimension served as a new index of forecasting the coal or rock dynamic disaster.Wang Chao Xu Jiankun Zhao Xiaoxia Wei Mingyao 2012International Journal of Mining Science and Technology2012,22,2:6
7Mesenchymal stem cell-based therapy for burn wound healing显示文摘Burns,with their high incidence and mortality rates,have a devastating effect on patients.There are still huge challenges in the management of burns.Mesenchymal stem cells(MSCs),which have multidirectional differentiation potential,have aroused interest in exploring the capacity for treating different intractable diseases due to their strong proliferation,tissue repair,immune tolerance and paracrine abilities,among other features.Currently,several animal studies have shown that MSCs play various roles and have beneficial effects in promoting wound healing,inhibiting burn inflammation and preventing the formation of pathological scars during burn healing process.The substances MSCs secrete can act on peripheral cells and promote burn repair.According to preclinical research,MSC-based treatments can effectively improve burn wound healing and reduce pain.However,due to the small number of patients and the lack of controls,treatment plans and evaluation criteria vary widely,thus limiting the value of these clinical studies.Therefore,to better evaluate the safety and effectiveness of MSC-based burn treatments,standardization of the application scheme and evaluation criteria of MSC therapy in burn treatment is required in the future.In addition,the combination of MSC pretreatment and dressing materials are also conducive to improving the therapeutic effect of MSCs on burns.In this article,we review current animal research and clinical trials based on the use of stem cell therapy for treating burns and discuss the main challenges and coping strategies facing future clinical applications.Mingyao Wang Xinxuan Xu Xiongxin Lei Jie Tan Huiqi Xie 2021Burns & Trauma2021,9,1:2
8Grain boundary optimization induced substantial squareness enhancement and high performance in iron-rich Sm-Co-Fe-Cu-Zr magnets显示文摘Increasing iron content has been witnessed an essential method to improve the remanence of 2:17-type Sm-Co-Fe-Cu-Zr magnets,however,the inferior squareness factor accompanied with the increased iron content turns into a neck sticking problem.In this work,the grain boundary optimization induced substantial squareness enhancement from 63.4%to 91.4%,and consequently an excellent maximum energy product of 32.63 MGOe have been achieved in iron-rich Sm-Co-Fe-Cu-Zr magnets via tuning solution process.It is clearly revealed that the grain boundary(GB)phases as well as the micro-twins’density in grain interiors can be controlled and interprets the enhancement mechanism of squareness.Jun Cao Tianli Zhang Jinghua Liu Hao Xu Mingyao Hu Wei Xia Ao Wang Hui Wang Chengbao Jiang 2021Journal of Materials Science & Technology2021,,26:2
