| 1 | Establishment of in Vitro Regeneration System of Bitter Melon (Momordica Charantia L. )显示文摘Explants of bitter melon can produce three types of callus: green, yellow green and fragile yellow callus. During the period of inducing callus, the explants are not sensitive to the kinds and proportions of phytohormone. The rate of callus inductivity is about 90.0%. However, in the process of callus differentiating adventitious bud, the kind, proportion and quantity of phytohormone and the type of callus made different result. The adventitious buds were induced successfully on MS medium+6-BA (N6-benzyladenine) 4.0 mg l-1+KT(kinetin) 2.0 mg l-1) by yellow green callus. The frequency was about 66.7 %. Yellow callus did not differentiated adventitious bud. The frequency of green callus differentiation was very low (<15.0 %) even under the most suitable conditions. The MS + ZT (zeatin) 5.0 mgl-1 +KT 0.5 mgl-1 medium was suitable for the proliferation of bud and three weeks later, the coefficient of proliferation was about 5-6. The 1/2 MS + ZT 0.02 mgl-1 or 1/2 MS media were suitable for in vitro rooting of shoot, the shoot on them could produce 6-7 new roots in three weeks. | 3 | Structural basis for a homodimeric ATPase subunit of an ECF transporter显示文摘The transition metal cobalt,an essential cofactor for many enzymes in prokaryotes,is taken up by several specifi c transport systems.The CbiMNQO protein complex be-longs to type-1 energy-coupling factor(ECF)transporters and is a widespread group of microbial cobalt transport-ers.CbiO is the ATPase subunit(A-component)of the cobalt transporting system in the gram-negative thermo-philic bacterium Thermoanaerobacter tengcongensis.Here we report the crystal structure of a nucleotide-free CbiO at a resolution of 2.3Å.CbiO contains an N-terminal canonical nucleotide-binding domain(NBD)and C-termi-nal helical domain.Structural and biochemical data show that CbiO forms a homodimer mediated by the NBD and the C-terminal domain.Interactions mainly via conserved hydrophobic amino acids between the two C-terminal do-mains result in formation of a four-helix bundle.Structural comparison with other ECF transporters suggests that non-conserved residues outside the T-component bind-ing groove in the A component likely act as a specifi city determinant for T components.Together,our data provide information on understanding of the structural organiza-tion and interaction of the CbiMNQO system. | Chengliang Chai You Yu Wei Zhuo Haifeng Zhao Xiaolu Li Na Wang Jijie Chai Maojun Yang | 2013 | Protein & Cell2013,4,10: | 1 |

| 7 | Identification and functional analysis of miRNAs in developing kernels of a viviparous mutant in maize显示文摘Given the important roles of miRNAs in post-transcriptional gene regulation, identification of differentially expressed miRNAs will facilitate the elucidation of molecular mechanisms underlying kernel development. In this study, we constructed a small RNA library to comprehensively represent the full complement of individual small RNAs and to characterize miRNA expression profiles in pooled ears of maize(Zea mays L.) at 10, 15,20, 22, 25 and 30 days after pollination(DAP). At least 21 miRNAs were differentially expressed. The differential expression of three of these miRNAs, i.e., miR528a, miR167a and miR160b, at each stage was verified by qRT-PCR. The results indicated that these miRNAs might be involved in kernel development. In addition, the predicted functions of target genes indicated that most of the target genes are involved in signal transduction and cell communication pathways, particularly the auxin signaling pathway. The expression of candidate germination-associated miRNAs was analyzed by hybridization to a maize genome microarray, and revealed differential expression of genes involved in plant hormone signaling pathways. This finding suggests that phytohormones play a critical role in the development of maize kernels. We found that in combination with other miRNAs, miR528a regulated a putative laccase, a Ring-H2 zinc finger protein and a MADS box-like protein, whereas miR167a and miR160b regulated multiple target genes,including ARF(auxin response factor), a member of the B3 transcription factor family. All three miRNAs are important for ear germination, development and physiology.
