| 1 | Ginkgo biloba extract improved cognitive and neurological functions of acute ischaemic stroke:a randomised controlled trial显示文摘Purpose To evaluate the efficacy and safety of Ginkgo biloba extract(GBE)in acute ischaemic stroke and its impact on the recurrence of vascular events.Methods We conducted a multicentre,prospective,randomised,open label,blinded,controlled clinical trial enrollingpatients with an onset of acute stroke within 7 days from five hospitals in China Jiangsu Province.Participants were assigned to the GBE group(450 mg GBE with 100 mg aspirin daily)or the control group(100 mg aspirin daily)for 6 months.The primary outcome was the decline in the Montreal Cognitive Assessment score at 6 months.Secondary outcomes were other neuropsychological tests of cognitive and neurological function,the the incidence of adverse events and vascular events.results 348 patients were enrolled:179 in the GBE group and 169 in the control group.With 18 patients lost to follow-up,the dropout rate was 5.17%.Admission data between two groups were similar,but in the GBE group there was a marked slow down in the decline in the Montreal Cognitive Assessment scores(−2.77±0.21 vs−1.99±0.23,P=0.0116(30 days);−3.34±0.24 vs−2.48±0.26,P=0.0165(90 days);−4.00±0.26 vs−2.71±0.26,P=0.0004(180 days))compared with controls.The National Institutes of Health Stroke Scale scores at 12 and 30 days,the modified Rankin Scale scores for independent rate at 30,90 and 180 days,and the Barthel Index scores at 30,90 and 180 days in the GBE group were significantly improved compared with controls.Improvements were also observedin GBE groups for Mini-Metal State Examination scores of 30,90 and 180 days,Webster's digit symbol test scores at 30 days and Executive Dysfunction Index scores at 30 and 180 days.No significant differences were seen in the incidence of adverse events or vascular events.conclusions We conclude that GBE in combination with aspirin treatment alleviated cognitive and neurological deficits after acute ischaemic stroke without increasing the incidence of vascular events. | Shanshan Li Xinjiang Zhang Qi Fang Junshan Zhou Meijuan Zhang Hui Wang Yan Chen Biyun Xu Yanfeng Wu Lai Qian Yun Xu | 2017 | Stroke & Vascular Neurology2017,2,4: | 25 |

| 3 | Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. | 4 | Hydrogen Sulfide Positively Regulates Abscisic Acid Signaling through Persulf idation of SnRK2.6 in Guard Cells显示文摘The phytohormone abscisic acid(ABA)plays pivotal roles in triggering stomatal closure and facilitating adaptation of plants to drought stress.Hydrogen sulfide(H2S),a small signaling gas molecule,is involved in ABA-dependent stomatal closure.However,how H2S regulates ABA signaling remains largely unclear.Here,we show that ABA induces the production of H2S catalyzed by L-CYSTEINE DESULFHYDRASE1(DES1)in guard cells,and H2S in turn positively regulates ABA signaling through persulfidation of Open Stomata 1(OST1)/SNF1-RELATED PROTEIN KINASE2.6(SnRK2.6).Two cysteine(Cys)sites,Cys131 and Cys137,which are exposed on the surface of SnRK2.6 and close to the activation loop,were identified to be persulfidated,which promotes the activity of SnRK2.6 and its interaction with ABA response element-binding factor 2(ABF2),a transcription factor acting downstream of ABA signaling.When Cys131,Cys137,or both residues in SnRK2.6 were substituted with serine(S),H2S_induced SnRK2_6 activity and SnRK2.6-ABF2 interaction were partially(SnRK2.6c131s and SnRK2.6c137S)or completely(SnRK2.6c131sc137S)compromised.Introduction of SnRK2.6c131s,SnRK2.6c137S,or SnRK2.6c131sc137S into the ost1-3 mutant could not rescue the mutant phenotype:less sensitivity to ABA-and H2S-induced stomatal closure and Ca2+influx as well as increased water loss and decreased drought tolerance.Taken together,our study reveals a novel post-translational regulatory mechanism of ABA signaling whereby H2S persulfidates SnRK2.6 to promote ABA signaling and ABA-induced stomatal closure. | Sisi Chen Honglei Jia Xiaofeng Wang Cong Shi Xiao Wang Peiyun Ma Juan Wang Meijuan Ren Jisheng Li | 2020 | Molecular Plant2020,13,5: | 13 |
| 5 | Electroacupuncture reduces injury to the blood-brain barrier following cerebral ischemia/reperfusion injury显示文摘This study used electroacupuncture at Renzhong (DU26) and Baihui (DU20) in a rat model of cerebral ischemia/reperfusion injury. Neurological deficit scores, western blotting, and reverse transcription-PCR results demonstrated that electroacupuncture markedly reduced neurological deficits, decreased corpus striatum aquaporin-4 protein and mRNA expression, and relieved damage to the blood-brain barrier in a rat model of cerebral ischemia/reperfusion injury. | 9 | IFN-inducible p47 GTPases display differential responses to Schistosoma japonicum acute infection显示文摘Interferon gamma induced GTPase(IGTP)(also named lrgm3)and interferon gamma inducible protein 47(IRG-47)(also named lIrgd)are interferon(IFN)-inducible p47 GTPases that have been shown to regulate host resistance to intracellular pathogens.Little knowledge has been known about the role of p47 GTPases in host responses against extracellular pathogens.To investigate possible roles of IGTP and IRG-47 in the course of Schistosoma japonicum infection,IGTP and IRG-47 knockout and wild-type(WT)mice were challenged with cercariae of S.japonicum,and host responses were analyzed.At the acute stage of S.japonicum infection,mice that lacked IGTP displayed similar parasite burden and pathological damage to WT mice.Importantly,S.japonicum-infected lRG-47-deficient mice,in contrast to IGTP-deficient mice and WT mice,showed significantly reduced worms and lower egg-burden,but intense granulomatous reaction evoked by schistosome eggs in peripheral parts of liver lobes.In addition,upregulation of inflammation-related gene expression was observed in the spleen of IRG-47-deficient mice using oligonucleotide microarrays,in which multiple pathways of cytokine-cytokine receptor interaction,T-cell receptor signaling,complement,coagulation cascades and cell adhesion molecules were highlighted.Taken together,these data suggest that IGTP and IRG-47 might have distinct features that were differentially required for resistance to S.japonicum. | Xuesong Chen Xiaotang Du Meijuan Zhang Donghui Zhang Minjun Ji Guanling Wu | 2010 | Cellular & Molecular Immunology2010,7,1: | 2 |
