1Development and application of wellbore heat transfer model considering variable mass flow显示文摘Dual-gradient drilling technology is being increasingly used in formations with narrow pressure margins.For dual-gradient drilling based on downhole separation,hollow spheres are separated into the annulus at the separator position,resulting in variable mass flow in the wellbore.Thus,existing heat transfer models are no longer suitable for describing wellbore temperature profiles in dual-gradient drilling.This study focused on developing a wellbore heat transfer model that fully considers separated hollow spheres entering the annulus,complex casing programs,and heat sources,for dual-gradient drilling based on downhole separation.The model was solved using an iterative method.Then,the accuracy of the model was verified using temperature data measured from two wells.Finally,the difference in the annular temperature distributions between dual-gradient drilling and conventional single-gradient drilling were investigated,as were the wellbore heat transfer characteristics for dual-gradient drilling.The following major conclusions were drawn:(1)for dualgradient drilling based on downhole separation,at the separator location,the annular fluid temperature does not decrease,but rather increase in the flow direction because of the inflow of hollow spheres;(2)a clear inflection point exists in the annular fluid temperature curve at the location where the separator would be;(3)the magnitude of the mutation of the temperature curve at the inflection point is considerably affected by the heat capacities of the hollow spheres and the pure drilling fluid;(4)under the same change in separation efficiency,distance between the bit and separator,flow rate,and thermal conductivity of formation,the variation range of the fluid temperature at the bottom hole is greater than that at the wellhead.Jiangshuai Wang Jun Li Gonghui Liu Shujie Liu Meipeng Ren Kuidong Luo Shanxun Tao 2021Underground Space2021,6,3:3

4Expression and immunoreactivity of an epitope of HCV in a foreign epitope presenting system显示文摘AIM: To construct and highly express an epitope of hepatitis C virus (HCV) in a foreign epitope presenting vectorbased on an insect virus, and to study the antigenicity of the epitope.METHODS: The HCV epitope sequence (amino acidresidues 315 to 328: EGHRMAWDMMMNWS) of the E1 region was constructed at different positions of a foreign epitope presenting vector based on an insect virus, flock house virus (FHV) capsid protein encoding gene as a vector, and expressed in E. coli cells. Western blottingand ELISA were used to detect the immunoreactivity of these recombinant proteins.RESULTS: The gene encoding of the concerned B-cell epitope of HCV E1 envelope protein was expressed on FHV capsid carrier protein at positions I1 (aa 106), I2 (aa153) and I3 (aa 305), respectively, on the surface of FHV capsid protein. The recombinant proteins in this system could be highly expressed in more than 40% of total cell protein of E. coli BL21. All the expressed recombinant proteins were in inclusion body form, and showed obvious immunoreactivity by Western blotting. Further purified recombinant proteins were detected by indirect ELISA as coating antigen respectively. All recombinant proteins could still show immunoreactivity.CONCLUSION: The epitope of HCV E1 envelope protein can be highly expressed in FHV carrier system as a chimeric protein with high immunoreactivity. 10Resistance Torque Based Variable Duty-Cycle Control Method for a Stage Ⅱ Compressor显示文摘The resistance torque of a piston stage Ⅱ compressor generates strenuous fluctuations in a rotational period, and this can lead to negative influences on the working performance of the compressor. To restrain the strenuous fluctuations in the piston stage Ⅱ compressor, a variable duty-cycle control method based on the resistance torque is proposed. A dynamic model of a stage Ⅱ compressor is set up, and the resistance torque and other characteristic parameters are acquired as the control targets. Then, a variable duty-cycle control method is applied to track the resistance torque, thereby improving the working performance of the compressor. Simulated results show that the compressor, driven by the proposed method,requires lower current, while the rotating speed and the output torque remain comparable to the traditional variable-frequency control methods. A variable duty-cycle control system is developed, and the experimental results prove that the proposed method can help reduce the specific power, input power, and working noise of the compressor to 0.97 k W·m^(-3)·min^(-1), 0.09 k W and 3.10 d B,respectively, under the same conditions of discharge pressure of 2.00 MPa and a discharge volume of 0.095 m^3/min. The proposed variable duty-cycle control method tracks the resistance torque dynamically, and improves the working performance of a Stage Ⅱ Compressor.
