1Connective tissue growth factor expression hints at aggressive nature of colorectal cancer显示文摘BACKGROUND Connective tissue growth factor(CTGF)is a mediator of transforming growth factor-beta signaling and plays a key role in connective tissue remodeling,inflammatory processes and fibrosis in various illnesses including cancer.AIM To investigate the role of CTGF in colorectal cancer(CRC)progression and to compare the CTGF expression with different clinicopathological parameters.METHODS Real-time polymerase chain reaction,immunohistochemistry and Western blotting was performed to evaluate the CTGF expression and the results were statistically analyzed against the clinicopathological variables of patient data using STATA software version 16.RESULTS CTGF expression levels in tumor specimens were significantly higher than their paired normal specimens.The higher protein expression levels showed a significant association with smoking,staging,tumor grade,invasion depth,necrosis of tumor tissue,and both lymphovascular and perineural invasion.As per the cox regression model and classification tree analysis,tumor-nodemetastasis stage and perineural invasion were important predictors for CTGF expression and prognosis of CRC patients.Survival analysis indicated that CTGF overexpression was associated with poorer overall and disease-free survival.CONCLUSION Expression of CTGF was increased in CRC and was linked with poor overall and disease-free survival of CRC patients.These findings support prior observations and thus CTGF may be a possible prognostic marker in CRC.Ishrat Parveiz Bhat Tahseen Bilal Rather Irfan Maqbool Gowhar Rashid Kulsum Akhtar Gulzar A Bhat Fazl Q Parray Besina Syed Ishrat Younas Khan Mohsin Kazi Muhammad D Hussain Mudassar Syed 2022World Journal of Gastroenterology2022,28,5:1
2Crystal Structure Analysis of 1-Hydroxy-3,5-dimethoxy-9H-xanthen-9-one Isolated from Ajuga bracteosa Root Extract and Its Biological Studies显示文摘The structure of 1-hydroxy-3,5-dimethoxy-9 H-xanthen-9-one isolated from chloroform extract of Ajuga bracteosa root was analyzed by single-crystal X-ray diffraction. DPPH(1,1-diphenyl-2-picryl hydrazyl), ABTS(2,2?-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities(RSA) and Fe2+ chelating activities were carried out to determine the antioxidant potential of the compound. RSA values for the compound were 96%, 96% and 96% for all the three activities respectively at maximum concentration of the compound(100 μ gmL-1) with the IC50 values of 3.40, 4.86 and 0.10(μ gmL-1). Antidiabetic activities including antiglycation and α-glucosidase inhibition were also performed where the antiglycation activity was performed using two techniques including spectrofluorometric as well as spectrophotometric technique. Spectrofluorometric technique provided 97% antiglycation potential while 92% antiglycation potential was observed by spectrophotometric technique for the isolated compound. The compound at a concentration of 10 μ gmL-1 exhibited 31% α-glucosidase inhibitory potential with IC50 of 15.56 μ gmL-1. 3Forkhead box P3 and indoleamine 2,3-dioxygenase co-expression in Pakistani triple negative breast cancer patients显示文摘BACKGROUND Forkhead box P3(FOXP3)is a specific marker for immunosuppressive regulatory T(T-reg)cells.T-regs and an immunosuppressive enzyme,indoleamine 2,3-dioxygenase(IDO),are associated with advanced disease in cancer.AIM To evaluate the co-expression of FOXP3 and IDO in triple negative breast cancer(TNBC)with respect to hormone-positive breast cancer patients from Pakistan.METHODS Immunohistochemistry was performed to analyze the expression of FOXP3,IDO,estrogen receptor,progesterone receptor,and human epidermal growth factor receptor on tissues of breast cancer patients(n=100):Hormone-positive breast cancer(n=51)and TNBC(n=49).A total of 100 patients were characterized as FOXP3 negative vs positive and further categorized based on low,medium,and high IDO expression score.Univariate and multivariate logistic regression models were used.RESULTS Out of 100 breast tumors,25%expressed FOXP3 positive T-regs.A significant coexpression of FOXP3 and IDO was observed among patients with TNBC(P=0.01)compared to those with hormone-positive breast cancer.Two variables were identified as significant independent risk factors for FOXP3 positive:IDO expression high(adjusted odds ratio(AOR)5.90;95%confidence interval(CI):1.22-28.64;P=0.03)and TNBC(AOR 2.80;95%CI:0.96-7.95;P=0.05).CONCLUSION Our data showed that FOXP3 positive cells might be associated with high expression of IDO in TNBC patients.FOXP3 and IDO co-expression may also suggest its involvement in disease,and evaluation of FOXP3 and IDO expression in TNBC patients may offer a new therapeutic option.Kashif Asghar Asif Loya Iftikhar Ali Rana Muhammad Abu Bakar Asim Farooq Muhammad Tahseen Muhammad Ishaq Iqra Masood Muhammad Usman Rashid 2020World Journal of Clinical Oncology2020,11,12:0
