2Intravenous formulation of Panax notoginseng root extract:human pharmacokinetics of ginsenosides and potential for perpetrating drug interactions显示文摘XueShuanTong,a lyophilized extract of Panax notoginseng roots(Sanqi)for intravenous administration,is extensively used as add-on therapy in the treatment of ischemic heart and cerebrovascular diseases and comprises therapeutically active ginsenosides.Potential for XueShuanTong-drug interactions was determined;the investigation focused on cytochrome P450(CYP)3A induction and organic anion-transporting polypeptide(OATP)1 B inhibition.Ginsenosides considerably bioavailable for drug interactions were identifed by dosing XueShuanTong in human subjects and their interaction-related pharmacokinetics were determined.The CYP3A induction potential was determined by repeatedly dosing XueShuanTong for 15 days in human subjects and by treating cryopreserved human hepatocytes with circulating ginsenosides;midazolam served as a probe substrate.Joint inhibition of OATP1B by XueShuanTong ginsenosides was assessed in vitro,and the data were processed using the Chou-Talalay method,Samples were analyzed by liquid chromatography/mass spectrometry.Ginsenosides Rb1,Rd,and Rg1 and notoginsenoside R were the major circulating XueShuanTong compounds;their interaction-related pharmacokinetics comprised compound dose-dependent levels of systemic exposure and,for ginsenosides Rb,and Rd,long terminal hal-lives(32-57 and 58-307 h,respectively)and low unbound fractions in plasma(0.8%-2.9%and 0.4%-3.0%,respectively).Dosing XueShuanTong did not induce CYP3A.Based on the pharmacokinetics and inhibitory potency of the ginsenosides,XueShuanTong was predicted to have high potential for OATP1B3-mediated drug interactions(attributed chiefly to ginsenoside Rb,)suggesting the need for further model-based determination of the interaction potential for XueShuanTong and,if necessary,a clinical drug interaction study.Increased awareness of ginsenosides'pharmacokinetics and XueShuanTong-drug interaction potential will help ensure the safe use of XueShuanTong and coadministered synthetic drugs.Salisa Pintusophon Wei Niu Xiao-na Duan Olajide E Olaleye Yu-hong Huang Feng-qing Wang Yan-fen Li Jun-ling Yang Chuan Li 2019Acta Pharmacologica Sinica2019,40,10:13
3The maternally expressed polycomb group gene OsEMF2a is essential for endosperm cellularization and imprinting in rice显示文摘Cellularization is a key event in endosperm development.Polycomb group(PcG)genes,such as Fertilization-Independent Seed 2(FIS2),are vital for the syncytium-to-cellularization transition in Arabidopsis plants.In this study,we found that OsEMF2a,a rice homolog of the Arabidopsis PcG gene Embryonic Flower2(EMF2),plays a role similar to that of FIS2 in regard to seed development,although there is limited sequence similarity between the genes.Delayed cellularization was observed in osemf2a,associated with an unusual activation of type I MADS-box genes.The cell cycle was persistently activated in osemf2a caryopses,which was likely caused by cytokinin overproduction.However,the overaccumulation of auxin was not found to be associated with the delayed cellularization.As OsEMF2a is a maternally expressed gene in the endosperm,a paternally inherited functional allele was unable to recover the maternal defects of OsEMF2a.Many imprinted rice genes were deregulated in the defective hybrid seeds of osemf2a(♀)/9311(♂)(m9).The paternal expression bias of some paternally expressed genes was disrupted in m9 due to either the activation of maternal alleles or the repression of paternal alleles.These findings suggest that OsEMF2a-PRC2-mediated H3K27me3 is necessary for endosperm cellularization and genomic imprinting in rice.Xiaojun Cheng Meiyao Pan Zhiguo E Yong Zhou Baixiao Niu Chen Chen 2021Plant Communications2021,2,1:5
4OsbZIP76 interacts with OsNF-YBs and regulates endosperm cellularization in rice (Oryza sativa)显示文摘Following double fertilization,plant endosperm nuclei undergo syncytial divisions,followed by synchro-nous cellularization.Cellularization is a key event during endosperm development,but our understanding of its regulation is limited to Arabidopsis.In this study we show that OsbZIP76 regulates cellularization in rice(Oryza sativa).Activation of OsbZIP76 coincided with the initiation of cellularization,and its knockdown or knockout mutants exhibited precocious cellularization.Genes involved in en-dosperm development or starch biosynthesis were pre-maturely activated in the osbzip76 caryopsis.As a putative transcription factor,OsbZIP76 alone lacked transcriptional activation activity;however,it interacted with the nuclear factor Y(NF-Y)family transcription factors OsNF-YB9 and OsNF-YB1 in yeast and in planta.OsbZIP76 and OsNF-YB9 were predominantly expressed in the endosperm and the proteins colocalized.Seeds of osnf-yb1 and osbzip76 mu-tants showed reduced size and reduced apparent amylose content.The parent-of-origin-dependent expression of OsbZIP76 is variable in different rice accessions.In sum-mary,OsbZIP76 is an endosperm-expressed imprinted gene that regulates endosperm development in rice.Baixiao Niu Hui Deng Tingting Li Sandeep Sharma Qianbin Yun Qianru Li Zhiguo E Chen Chen 2020Journal of Integrative Plant Biology2020,62,12:4
