| 2 | HIF-1α-induced expression of m6A reader YTHDF1 drives hypoxia-induced autophagy and malignancy of hepatocellular carcinoma by promoting ATG2A and ATG14 translation显示文摘N6-methyladenosine(m6A),and its reader protein YTHDF1,play a pivotal role in human tumorigenesis by affecting nearly everystage of RNA metabolism.Autophagy activation is one of the ways by which cancer cells survive hypoxia.However,the possibleinvolvement of m6A modification of mRNA in hypoxia-induced autophagy was unexplored in human hepatocellular carcinoma(HCO).In this study,specific variations in YTHDF1 expression were detected in YTHDF1-overexpressing,knockout,and-knockdownHCC cells,HCC organoids,and HCC patient-derived xenograft(PDX)murine models.YTHDF1 expression and hypoxia inducedautophagy were significantly correlated in vitro;signifhcant overexpression of YTHDF1 in HCC tissues was associated with poorprognosis,Multivariate cox regression analysis identihed YTHDF1 expression as an independent prognostic factor in patients withHCC.Multiple HC models conhrmed that YTHDF1 deficiency inhibited HCC autophagy,growth,and metastasis.Luciferase reporterassays and chromatin immunoprecipitation demonstrated that HlIF-1a regulated YTHDF1 transcription by directly binding to itspromoter region under hypoxia.The results of methylated RNA immunoprecipitation sequencing,proteomics,and polysomeprofling indicated that YTHDF1 contibuted to the translation of autophagy-related genes ATG2A and ATG14 by binding to m6A-modifhed ATG2A and ATG14 mRNA,thus facilitating autophagy and autophagy-related malignancy of HCC.Taken together,HlE-1d-induced YTHDF1 expression was associated with hypoxia-induced autophagy and autophagy-related HCC progression via promoting translation of autophagy-related genes ATG2A and ATG14 in a m6A-dependent manner.Our fndings suggest thatYTHDF1 is a potential prognostic biomarker and therapeutic target for patients with HCC. | Qing Li Yong Ni Liren Zhang Runqiu Jiang Jing Xu Hong Yang Yuanchang Hu Jiannan Qiu Liyong Pu Jinhai Tang Xuehao Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,3: | 24 |

| 4 | Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis. | TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang | 2016 | Biomedical and Environmental Sciences2016,29,12: | 21 |
| 5 | N-acetylcysteine attenuates reactive-oxygen-speciesmediated endoplasmic reticulum stress during liver ischemia-reperfusion injury显示文摘AIM:To investigate the effects of N-acetylcysteine(NAC) on endoplasmic reticulum(ER) stress and tissue injury during liver ischemia reperfusion injury(IRI).METHODS:Mice were injected with NAC(300 mg/kg) intraperitoneally 2 h before ischemia.Real-time polymerase chain reaction and western blotting determined ER stress molecules(GRP78,ATF4 and CHOP).To analyze the role of NAC in reactive oxygen species(ROS)-mediated ER stress and apoptosis,lactate dehydrogenase(LDH) was examined in cultured hepatocytes treated by H2O2 or thapsigargin(TG).RESULTS:NAC treatment significantly reduced the level of ROS and attenuated ROS-induced liver injury after IRI,based on glutathione,malondialdehyde,serum alanine aminotransferase levels,and histopathology.ROS-mediated ER stress was significantly inhibited in NAC-treated mice.In addition,NAC treatment significantly reduced caspase-3 activity and apoptosis after reperfusion,which correlated with the protein expression of Bcl-2 and Bcl-xl.Similarly,NAC treatment significantly inhibited LDH release from hepatocytes treated by H2O2 or TG.CONCLUSION:This study provides new evidence for the protective effects of NAC treatment on hepatocytes during IRI.Through inhibition of ROS-mediated ER stress,NAC may be critical to inhibit the ER-stressrelated apoptosis pathway. | Yong Sun Li-Yong Pu Ling Lu Xue-Hao Wang Feng Zhang Jian-Hua Rao | 2014 | World Journal of Gastroenterology2014,20,41: | 13 |

| 8 | The Role of Exportin-5 in MicroRNA Biogenesis and Cancer显示文摘Micro RNAs(mi RNAs) are conserved small non-coding RNAs that play an important role in the regulation of gene expression and participate in a variety of biological processes. The biogenesis of mi RNAs is tightly controlled at multiple steps, such as transcription of mi RNA genes,processing by Drosha and Dicer, and transportation of precursor mi RNAs(pre-mi RNAs) from the nucleus to the cytoplasm by exportin-5(XPO5). Given the critical role of nuclear export of premi RNAs in mi RNA biogenesis, any alterations of XPO5, resulting from either genetic mutation,epigenetic change, abnormal expression level or posttranslational modification, could affect mi RNA expression and thus have profound effects on tumorigenesis. Importantly, XPO5 phosphorylation by ERK kinase and its cis/trans isomerization by the prolyl isomerase Pin1 impair XPO50 s nucleo-to-cytoplasmic transport ability of pre-mi RNAs, leading to downregulation of mature mi RNAs in hepatocellular carcinoma. | 9 | Immunization of Male Mice with a New Recombinant GnRH Fusion Protein Reduces the Testicular Function显示文摘The objective of the present study was to evaluate the effect of an immunogenic maltose-binding protein-gonadotropin releasing hormone (GnRH-6-MBP) using genetic engineering. The synthetic mammalian tandem repeated GnRH hexamer gene was inserted into the expression plasmid pMAL-c2x. Recombinant GnRH-6-MBP protein was over-expressed in E.coli strain BL21. Amylose resin with affinity chromatograph was used to purify target protein. The reactiongenicity of fusion protein was identified by indirect enzyme linked immunosorbent assay (ELISA), and the antigenicity and biological effects of GnRH-6-MBP were tested in mice. In the experiment, 20 male Kunming white mice of 20 d old were randomly divided into treatment and control group. Ten mice were immunized with 100 μg GnRH-6-MBP administered subcutaneously (s.c.) thrice at 2-week intervals with GnRH-6-MBP. Mice were sacrificed after 3 weeks following the booster injection, the testis was removed, weighed and measured, and the histological structure was observed. The reactiongenicity of fusion protein to GnRH antibody was much higher than the control. Active immunization against GnRH-6-MBP reduced remarkably (P < 0.01) the length and weight of the testis, and shortened the girth and width of the testis (P < 0.05), and suppressed testicular spermatogenesis compared to the control mice. | 10 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 14 | Assessment on the Effects of Hepatitis B Prevention and Control Measures in Western China:A Comparison of Three Population-based Serosurveys显示文摘Objective Despite the remarkable progress in efforts to control disease spread,the nationwide elimination of hepatitis B in China is still hindered by the persistently high rate of hepatitis B virus(HBV)infection in Western China.This study aimed to evaluate the strategy of hepatitis B prevention and control in Western China and identify potential areas and strategies for improvement.Methods Susceptible population vaccination,health education,professional training of doctors,and other prevention and control measures have been implemented in Wuwei city since 2010.Data were obtained from three representative cross-sectional serosurveys conducted in 2010,2013,and 2015.The serum samples were subjected to enzyme-linked immunosorbent assays to detect the following seromarkers:HBV surface antigen(HBsAg),antibody against hepatitis B surface antigen(anti-HBs),and antibody against hepatitis B core antigen(anti-HBc).Estimates of variance were determined using Taylor series linearization methods.Results The three serosurveys revealed decreases in the prevalence of HBsAg(7.19% in 2010 vs.6.51% in 2013 vs.5.87% in 2015)and anti-HBc positivity(43.89%vs.32.87%vs.28.46%)and an increase in the prevalence of anti-HBs positivity(49.07%vs.53.66%vs.53.72%)over time.From 2010 to 2015,the legally reported incidence of hepatitis B in Wuwei city decreased from 686.53/100,000 to 53.72/100,000.Notably,persistently high HBsAg-positive rates(above 5.40%)were observed among subjects aged 20–69 years old in the three serosurveys;the prevalence of HBsAg was above 1% among children younger than 10 years old.Furthermore,rural subjects had higher rates of HBsAg and anti-HBc positivity than their urban counterparts(6.04%vs.4.83% and 30.26%vs.20.35%,respectively)in 2015 but had a lower rate of anti-HBs positivity(49.68 vs.55.18%).Multivariate regression analysis showed that age,urban and rural areas,and education level were the main factors affecting HBV infection.Conclusion Although vaccine-based prevention and control measures reduced the rate of HBV infection in Wuwei City over time,the hepatitis B infection rate in children younger than 10 years was still higher than the national average level.Therefore,the prevention and control of mother-to-child transmission and the management of the infected should be the focus of future prevention and control work. | CHEN Hui LIU Nan JI Zhao Hua PU Zhong Shu GUO Zhi Wen GAO Jie SHAO Zhong Jun LIU Yi Wen YAN Yong Ping | 2020 | Biomedical and Environmental Sciences2020,33,10: | 5 |
| 15 | Elevated Platelet Activating Factor Level in Ischemia-Related Arrhythmia and Its Electrophysiological Effect on Myocardium显示文摘Objective The mechanism through which platelet activating factor (PAF) induces cardiac electrical activity and arrhythmia is not well understood and previous studies have suggested a potential involvement of ion channels in its action. The present study was aimed to clarify the role of PAF in fatal arrhythmias following acute myocardia infarction (AMI) and the underlying mechanism. Methods (1) Blood PAF levels were measured among 72 AMI patients at the time of diagnosis with AMI and 48 h later, and their electrocardiogram (ECG) was recorded continuously. (2) Ischemia simulation and surface electrocardiogram were conducted in 20 pigs and their PAF levels were measured. (3) PAF perfusion and standard microelectrode recording were performed on guinea pig papillarymuscles. Results In both humans and pigs, elevated PAF levels were detected in AMI and simulated ischemia, respectively, and even higher PAF levels were found when fatal arrhythmias occurred. In guinea pig myocardium, PAF induced a shortening of action potential duration at 90% level of repolarization (APD 90 )under non-ischemic conditions and a more pronounced shortening under early simulated ischemic conditions. Conclusion AMI and ischemia are associated with increased PAF levels in humans and pigs, which are further raised when fatal arrhythmia follows. | 17 | Precipitation Behavior and Microstructural Evolution of Ferritic Ti–V–Mo Complex Microalloyed Steel显示文摘Precipitation behavior of(Ti,V,Mo)C and microstructural evolution of the ferritic Ti–V–Mo complex microalloyed steel were investigated through changing coiling temperature(CT).It is demonstrated that the strength of the Ti–V–Mo microalloyed steel can be ascribed to the combination of grain refinement hardening and precipitation hardening.The variation of hardness(from 318 to 415 HV,then to 327 HV) with CT(from 500 to 600–625 °C,then to 700 °C) was attributed to the changes of volume fraction and particle size of(Ti,V,Mo)C precipitates.The optimum CT was considered as 600–625 °C,at which the maximum hardness value(415 HV) can be obtained.It was found that the atomic ratios of Ti,V and Mo in(Ti,V,Mo)C carbides were changed as the CT increased.The precipitates with the size of \ 10 nm were the V-rich particles at higher CT of 600 and 650 °C,while the Ti-rich particles were observed at lower CT of 500 and 550 °C.Theoretical calculations indicated that the maximum nucleation rate of(Ti,V,Mo)C in ferrite matrix occurred around 630 °C,which was consistent with the 625 °C obtained from experiment results. | Ke Zhang Hui Wang Xin-Jun Sun Feng-Li Sui Zhao-Dong Li En-Xiang Pu Zheng-Hai Zhu Zhen-Yi Huang Hong-Bo Pan Qi-Long Yong | 2018 | Acta Metallurgica Sinica(English Letters)2018,31,9: | 4 |

| 20 | Strip line beam position monitor for HLS LINAC显示文摘The parameters of beam transverse positions at HLS 200 MeV LINAC are very important to injection ef-ficiency. We have designed a new non-interceptive strip line beam position monitor (BPM) as a substitution for the original interceptive fluorescent target. This paper gives out the theoretical analysis and bench test result of the strip line BPM. The BPM has a characteristic impedance of (50±5)?, a reflection coefficient of less than –4 dB, a band-width of 400 MHz, a coupling coefficient of less than -15 dB, and a sensitivity of 1.16 dB/mm.
