1Host pathogen interactions in Helicobacter pylori related gastric cancer显示文摘Helicobacter pylori (H. pylori), discovered in 1982, is a microaerophilic, spiral-shaped gram-negative bacterium that is able to colonize the human stomach. Nearly half of the world's population is infected by this pathogen. Its ability to induce gastritis, peptic ulcers, gastric cancer and mucosa-associated lymphoid tissue lymphoma has been confirmed. The susceptibility of an individual to these clinical outcomes is multifactorial and depends on H. pylori virulence, environmental factors, the genetic susceptibility of the host and the reactivity of the host immune system. Despite the host immune response, H. pylori infection can be difficult to eradicate. H. pylori is categorized as a group?Ⅰ?carcinogen since this bacterium is responsible for the highest rate of cancerrelated deaths worldwide. Early detection of cancer can be lifesaving. The 5-year survival rate for gastric cancer patients diagnosed in the early stages is nearly 90%. Gastric cancer is asymptomatic in the early stages but always progresses over time and begins to cause symptoms when untreated. In 97% of stomach cancer cases, cancer cells metastasize to other organs. H. pylori infection is responsible for nearly 60% of the intestinaltype gastric cancer cases but also influences the development of diffuse gastric cancer. The host genetic susceptibility depends on polymorphisms of genes involved in H. pylori-related inflammation and the cytokine response of gastric epithelial and immune cells. H. pylori strains differ in their ability to induce a deleterious inflammatory response. H. pylori-driven cytokines accelerate the inflammatory response and promote malignancy. Chronic H. pylori infection induces genetic instability in gastric epithelial cells and affects the DNA damage repair systems. 3Mucosal bacterial microflora and mucus layer thickness in adolescents with inflammatory bowel disease显示文摘AIM:To assess the mucosa-associated bacterial microflora and mucus layer in adolescents with inflammatory bowel disease(IBD) .METHODS:Sixty-one adolescents(mean age 15 years,SD ± 4.13) were included in the study.Intestinal biopsies from inflamed and non-inflamed mucosa of IBD patients and from controls with functional abdominal pain were cultured under aerobic and anaerobic conditions.The number of microbes belonging to the same group was calculated per weight of collected tissue.The mucus thickness in frozen samples was measured under a fluorescent microscope.RESULTS:The ratios of different bacterial groups in inflamed and non-inflamed mucosa of IBD patients and controls were specific for particular diseases.Streptococcus spp.were predominant in the inflamed mucosa of Crohn's disease(CD) patients(80% of all bacteria) ,and Lactobacillus spp.were predominant in ulcerative colitis patients(90%) .The differences were statistically significant(P = 0.01-0.001) .Lower number of bifidobacteria was observed in the whole IBD group.A relation was also found between clinical and endoscopic severity and decreased numbers of Lactobacillus and,to a lesser extent,of Streptococcus in biopsies from CD patients.The mucus layer in the inflamed sites was significantly thinner as compared to controls(P = 0.0033) and to non-inflamed areas in IBD patients(P = 0.031) .CONCLUSION:The significantly thinner mucosa of IBD patients showed a predominance of some aerobes specific for particular diseases,their numbers decreased in relation to higher clinical and endoscopic activity of the disease.Krzysztof Fyderek Magdalena Strus Kinga Kowalska-Duplaga Tomasz Gosiewski Andrzej Wedrychowicz Urszula Jedynak-Wasowicz Malgorzata Sladek Stanislaw Pieczarkowski Pawel Adamski Piotr Kochan Piotr B Heczko 2009World Journal of Gastroenterology2009,15,42:14

7Endoscopic management of gastrointestinal perforations, leaks and fistulas显示文摘Gastrointestinal perforations, leaks and fistulas may be serious and life-threatening. The increasing number of endoscopic procedures with a high risk of perforation and the increasing incidence of leakage associated with bariatric operations call for a minimally invasive treatment for these complications. The therapeutic approach can vary greatly depending on the size,location, and timing of gastrointestinal wall defect recognition. Some asymptomatic patients can be treated conservatively, while patients with septic symptoms or cardio-pulmonary insufficiency may require intensive care and urgent surgical treatment.However, most gastrointestinal wall defects can be satisfactorily treated by endoscopy. Although the initial endoscopic closure rates of chronic fistulas is very high, the long-term results of these treatments remain a clinical problem. The efficacy of endoscopic therapy depends on several factors and the best mode of treatment will depend on a precise localization of the site, the extent of the leak and the endoscopic appearance of the lesion. Many endoscopic tools for effective closure of gastrointestinal wall defects are currently available. In this review, we summarized the basic principles of the management of acute iatrogenic perforations, as well as of postoperative leaks and chronic fistulas of the gastrointestinal tract. 8Modeling of particle transport and combustion phenomena in a large-scale circulating fluidized bed boiler using a hybrid Euler-Lagrange approach显示文摘The constantly developing fluidized combustion technology has become competitive with a conventional pulverized coal(PC) combustion.Circulating fluidized bed(CFB) boilers can be a good alternative to PC boilers due to their robustness and lower sensitivity to the fuel quality.However,appropriate engineering tools that can be used to model and optimize the construction and operating parameters of a CFB boiler still require development.This paper presents the application of a relatively novel hybrid Euler-Lagrange approach to model the dense gas-solid flow combined with a combustion process in a large-scale industrial CFB boiler.In this work,this complex flow has been resolved by applying the ANSYS FLUENT 14.0commercial computational fluid dynamics(CFD) code.To accurately resolve the multiphase flow,the original CFD code has been extended by additional user-defined functions.These functions were used to control the boiler mass load,particle recirculation process(simplified boiler geometry),and interphase hydrodynamic properties.This work was split into two parts.In the first part,which is referred to as pseudo combustion,the combustion process was not directly simulated.Instead,the effect of the chemical reactions was simulated by modifying the density of the continuous phase so that it corresponded to the mean temperature and composition of the flue gases.In this stage,the particle transport was simulated using the standard Euler-Euler and novel hybrid Euler-Lagrange approaches.The obtained results were compared against measured data,and both models were compared to each other.In the second part,the numerical model was enhanced by including the chemistry and physics of combustion.To the best of the authors' knowledge,the use of the hybrid Euler-Lagrange approach to model combustion is a new engineering application of this model.In this work,the combustion process was modeled for air-fuel combustion.The simulation results were compared with experimental data.The performed numerical simulations showed the applicability of the hybrid dense discrete phase model approach to model the combustion process in large-scale industrial CFB boilers.Wojciech P.Adamczyk Gabriel Wecel Marcin Klajny Pawel Kozolub Adam Klimanek Ryszard A.Bialecki 2014Particuology2014,12,5:7
9Emerging blood-based biomarkers for detection of gastric cancer显示文摘Early detection and efficient monitoring of tumor dynamics are prerequisites for reducing disease burden and mortality, and for improving the management of patients with gastric cancer(GC). Blood-based biomarker assays for the detection of early-stage GC could be of great relevance both for population-wide or risk groupbased screening programs, while circulating biomarkers that reflect the genetic make-up and dynamics of the tumor would allow monitoring of treatment efficacy, predict recurrences and assess the genetic heterogeneity of the tumor. Recent research to identify blood-based biomarkers of GC has resulted in the identification of a wide variety of cancer-associated molecules, including various proteins, autoantibodies against tumor associated antigens, cell-free DNA fragments, m RNAs and various non-coding RNAs, circulating tumor cells and cancer-derived extracellular vesicles. Each type of these biomarkers provides different information on the disease status, has different advantages and disadvantages, and distinct clinical usefulness. 10Impact of IL28B and OAS gene family polymorphisms on interferon treatment response in Caucasian children chronically infected with hepatitis B virus显示文摘AIM To investigate the impact of IL28 B and OAS gene polymorphisms on interferon treatment responses in children with chronic hepatitis B.METHODS We enrolled 52 children(between the ages of 4 and 18) with hepatitis B e antigen-negative chronic hepatitis B(CHB), who were treated with pegylated interferon alfa for 48 wk. Single nucleotide polymorphisms in the OAS1(rs1131476), OAS2(rs1293747),OAS3(rs2072136), OASL(rs10849829) and IL28B(rs12979860, rs12980275 and rs8099917) genes were studied to examine their associations with responses to IFN treatment in paediatric patients. We adopted two criteria for the therapeutic response, achieving an hepatitis B virus(HBV) DNA level < 2000 IU/m L and normalization of ALT activity(< 40 IU/L). To perform the analyses, we compared the patients in terms of achieving a partial response(PR) and a complete response(CR) upon measurement at the 24-wk posttreatment follow-up. RESULTS The PR and CR rates were 80.8% and 42.3%, respectively. Factors such as age, gender and liver histology had no impact on the type of response(partial or complete). A statistically significant relationship between higher baseline HBV DNA and ALT activity levels and lower rates of PR and CR was shown(P < 0.05). The allele association analysis revealed that only the IL-28 B rs12979860(C vs T) and IL28 B rs12980275(A vs G) markers significantly affected the achievement of PR(P = 0.021, OR = 3.3, 95%CI: 1.2-9.2 and P = 0.014, OR = 3.7, 95%CI: 1.3-10.1, respectively). However, in the genotype analysis, only IL-28 B rs12980275 was significantly associated with PR(AA vs AG-GG, P = 0.014, OR = 10.9, 95%CI: 1.3-93.9). The association analysis for CR showed that the TT genotype of IL28 B rs12979860 was present only in the no-CR group(P = 0.033) and the AA genotype of OASL rs10849829 was significantly more frequent in the noCR group(P = 0.044, OR = 0.26, 95%CI: 0.07-0.88). 11Comprehensive Approach for Porous Materials Analysis Using a Dedicated Preprocessing Tool for Mass and Heat Transfer Modeling显示文摘The paper presents a comprehensive, newly developed software – poROSE(poROus materials examination SoftwarE) for the qualitative and quantitative assessment of porous materials and analysis methodologies developed by the authors as a solution for emerging challenges. A low porosity rock sample was analyzed and thanks to the developed and implemented methodologies in poROSE software, the main geometrical properties were calculated. A tool was also used in preprocessing part of the computational analysis to prepare a geometrical representation of the porous material. The basic functions as elimination of blind pores in the geometrical model were completed and the geometrical model was exported for CFD software. As a result, it was possible to carry out calculations of the basic properties of the analyzed porous material sample. The developed tool allows to carry out quantitative and qualitative analysis to determine the most important properties characterized porous materials. In presented tool the input data can be images from X-ray computed tomography(CT), scanning electron microscope(SEM) or focused ion beam with scanning electron microscope(FIB-SEM) in grey level. A geometric model developed in the proper format can be used as an input to modeling mass, momentum and heat transfer, as well as, in strength or thermo-strength analysis of any porous materials. In this example, thermal analysis was carried out on the skeleton of rock sample. 15趋化因子CCL5在食管癌组织中的表达及意义显示文摘目的 探讨趋化因子CCL5在食管癌组织中的表达及意义.方法 采用逆转录聚合酶链反应(RT-PCR)检测食管癌组织和癌旁组织中CCL5、CD8和CD8^+T细胞杀伤功能相关细胞因子穿孔素(perforin)和颗粒酶B(granzyme B)的表达水平.采用流式细胞术检测食管癌患者外周血单核细胞(PBMC)和肿瘤浸润性淋巴细胞(TIL)中CD8^+T细胞和CCR5^+CD8^+T细胞比例.采用Transwell实验检测CCL5对T细胞运动的影响.结果 食管癌组织中CCL5和perforin mRNA的表达水平分别为0.348 2±0.300 1和0.181 9±0.118 6,癌旁组织中CCL5和perforin mRNA的表达水平分别为0.279 6±0.138 0和0.118 0±0.1098,但差异均无统计学意义(均P>0.05).食管癌组织中CD8和granzyme B mRNA的表达水平分别为0.4649±0.3008和0.6487±0.5160,癌旁组织中CD8和granzyme B mRNA的表达水平分别为0.279 0±0.173 4和0.469 7±0.259 1,差异有统计学意义(均P <0.05).食管癌患者中CCL5与CD8、perforin和granzyme B表达呈正相关(rCD8=0.272,P=0.034;rerforin=0.305,P=0.026;rgranzymeB=0.108,P=0.012).流式细胞术结果显示,TIL和PBMC中CD8^+T细胞比例分别为(45.86±16.09)%和(34.05±15.07)%,差异有统计学意义(P =0.022);TIL和PBMC中CCR5^+ CD8^+T细胞比例分别为(48.12±26.75)%和(19.53±13.67)%,差异有统计学意义(P<0.001).Transwell实验结果显示,CCL5显著增强T细胞趋化运动.CCL5表达与患者性别、年龄和淋巴结转移无关,但CCL5在早期食管癌组织中的相对表达水平为0.319 9±0.161 7,在晚期食管癌组织中的相对表达水平为0.232 8±0.121 0,差异有统计学意义(P=0.008).早期食管癌患者TIL中CD8^+T细胞和CCR5^+ CD8^+T细胞比例分别为(48.86±15.87)%和(56.23±26.23)%,而在晚期食管癌患者TIL中CD8^+T细胞和CCR5^+ CD8^+T细胞比例分别为(33.25±16.49)%和(33.53±21.03)%,差异均有统计学意义(PCD8 =0.007,PccR5=0.010).结论 CD8^+T细胞能够在CCL5的诱导下向肿瘤部位运动,并影响患者疾病进展,CCL5可作为食管癌治疗的新靶点。刘金燕 李峰 陈新峰 王丽萍 岳冬丽 赵松 胡伟 Pawel Kalinski Stephen Thorne Hou Jingzhou 张毅 2014中华肿瘤杂志2014,36,11:5
16Thermal Analysis of Underground Power Cable System显示文摘The paper presents the application of Finite Element Method in thermal analysis of underground power cable system. The computations were performed for power cables buried in-line in the ground at a depth of 2 meters. The developed mathematical model allows determining the two-dimensional temperature distribution in the soil, thermal backfill and power cables. The simulations studied the effect of soil and cable backfill thermal conductivity on the maximum temperature of the cable conductor. Also, the effect of cable diameter on the temperature of cable core was studied. Scarce natural resources and our choices to protect them or exploit them jointly determine the economic and environmental systems. In this paper we describe a model with a particular focus on the relationship among income, pollution, and non-renewable resources. We want to combine both economic and environmental sectors. The system dynamics approach is used in analyzing these complex relationships. This paper gives an insight into the possibilities for replacing non-renewable resources with more renewable ones. Next, we present the simulation runs of the model that are conducted with the help of existing system dynamics modeling tools. 18Dolomite fracture modeling using the Johnson-Holmquist concrete material model:Parameter determination and validation显示文摘In this paper,the Johnson-Holmquist concrete(JHC)constitutive model is adopted for modeling and simulating the fracture of dolomite.A detailed step-by-step procedure for determining all required parameters,based on a series of experiments under quasi-static and dynamic regimes,is proposed.Strain rate coefficients,failure surfaces,equations of state and damage/failure constants are acquired based on the experimental data and finite element analyses.The JHC model with the obtained parameters for dolomite is subsequently validated using quasi-static uniaxial and triaxial compression tests as well as dynamic split Hopkinson pressure bar(SHPB)tests.The influence of mesh size is also analyzed.It shows that the simulated fracture behavior and waveform data are in good agreement with the experimental data for all tests under both quasi-static and dynamic loading conditions.Future studies will implement the validated JHC model in small-and large-scale blasting simulations.Michal Kucewicz Pawel Baranowski Jerzy Maachowski 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,2:4
19Role of cyclooxygenase-2 gene polymorphisms in pancreatic carcinogenesis显示文摘AIM:To evaluate the clinical significance of-765G/C and-1195G/A cyclooxygenase-2 (COX-2) gene polymorphisms in patients with pancreatic cancer (PC).METHODS:The study included 201 patients:85 with PC and 116 healthy controls.-765G/C and-1195G/A COX-2 gene polymorphisms were studied in DNA isolated from blood samples.The associations of the analyzed genotypes and clinical data at diagnosis were evaluated.RESULTS:We found an increased frequency of the homozygous-1195AA COX-2 genotype in patients with PC (53.7%) compared with the control group (21%) (P < 0.01).In contrast,the distribution of genotypeand allele frequencies of the-765G/C COX-2 polymorphism in the PC patients were not different from those in control groups.A correlation between presence of homozygous-1195AA COX-2 genotype and tumor size > 3 cm was observed (P < 0.05).Analyzed polymorphisms were unrelated to the patients' sex and age,nor to the presence of regional or distant metastases.CONCLUSION:These preliminary results indicate that the-1195G/A COX-2 polymorphism may play an important role in PC prognosis and carcinogenesis.Renata Talar-Wojnarowska Anita Gasiorowska Marek Olakowski Pawel Lampe Beata Smolarz Hanna Romanowicz-Makowska Ewa Malecka-Panas 2011World Journal of Gastroenterology2011,17,36:4
20Generation of sub-100 fs pulses tunable from 1700 to 2100 nm from a compact frequency-shifted Er-fiber laser显示文摘We report generation of sub-100 fs pulses tunable from 1700 to 2100 nm via Raman soliton self-frequency shift.The nonlinear shift occurs in a highly nonlinear fiber, which is pumped by an Er-doped fiber laser. The whole system is fully fiberized, without the use of any free-space optics.
