| 1 | Technology-Driven and Evidence-Based Genomic Analysis for Integrated Pediatric and Prenatal Genetics Evaluation显示文摘The first decade since the completion of the Human Genome Project has been marked with rapid development of genomic technologies and their immediate clinical applications.Genomic analysis using oligonucleotide array comparative genomic hybridization(aCGH) or single nucleotide polymorphism(SNP) chips has been applied to pediatric patients with developmental and intellectual disabilities(DD/ ID),multiple congenital anomalies(MCA) and autistic spectrum disorders(ASD).Evaluation of analytical and clinical validities of aCGH showed>99%sensitivity and specificity and increased analytical resolution by higher density probe coverage.Reviews of case series, multi-center comparison and large patient-control studies demonstrated a diagnostic yield of 12%—20%;approximately 60%of these abnormalities were recurrent genomic disorders.This pediatric experience has been extended toward prenatal diagnosis.A series of reports indicated approximately 10%of pregnancies with ultrasound-detected structural anomalies and normal cytogenetic findings had genomic abnormalities,and 30%of these abnormalities were syndromic genomic disorders.Evidence-based practice guidelines and standards for implementing genomic analysis and web-delivered knowledge resources for interpreting genomic findings have been established.The progress from this technology-driven and evidence-based genomic analysis provides not only opportunities to dissect disease-causing mechanisms and develop rational therapeutic interventions but also important lessons for integrating genomic sequencing into pediatric and prenatal genetic evaluation. | Yuan Wei Fang Xu Peining Li | 2013 | Journal of Genetics and Genomics2013,40,1: | 4 |
| 2 | Cgr1,a ripe rot resistance QTL in Vitis amurensis‘Shuang Hong’grapevine显示文摘Ripe rot is a serious grapevine disease in Vitis L.and Muscadinia(Planch.)Small.However,resistance to this disease has been reported in some oriental Vitis species.To identify resistance-related Quantitative Trait Loci(QTLs)from the Chinese grape species V.amurensis,an F1 population of V.vinifera‘Cabernet Sauvignon’×V.amurensis‘Shuang Hong’was used to map the ripe rot resistance loci expected in‘Shuang Hong’grape.A total of 7598 single nucleotide polymorphisms(SNPs)between the parents were identified in our previous study,and 934 SNPs were selected for genetic map construction.These SNPs are distributed across the 19 chromosomes covering a total of 1665.31 cM in length,with an average of 1.81 cM between markers.Ripe rot resistance phenotypes among the hybrids were evaluated in vitro using excised leaves for three consecutive years from 2016 to 2018;a continuous variation was found among the F1 hybrids,and the Pearson correlation coefficients of the phenotypes scored in all three years were significant at the 0.01 level.Notably,the first QTL reported for resistance to grape ripe rot disease,named Cgr1,was identified on chromosome 14 of‘Shuang Hong’grapevine.Cgr1 could explain up to 19.90%of the phenotypic variance.In addition,a SNP named‘np19345’was identified as a molecular marker closely linked to the peak of Cgr1 and has the potential to be developed as a marker for the Cgr1 resistance haplotype. | Peining Fu Quanyou Tian Gongti Lai Rongfang Li Shiren Song Jiang Lu | 2019 | Horticulture Research2019,6,1: | 3 |
| 3 | Spectrum of Cytogenomic Abnormalities Revealed by Array Comparative Genomic Hybridization on Products of Conception Culture Failure and Normal Karyotype Samples显示文摘Approximately 30% of pregnancies after implantation end up in spontaneous abortions, and 50% of them are caused by chromosomal abnormalities. However, the spectrum of genomic copy number variants(CNVs) in products of conception(POC) and the underlying genedosage-sensitive mechanisms causing spontaneous abortions remain largely unknown. In this study, array comparative genomic hybridization(a CGH) analysis was performed as a salvage procedure for 128 POC culture failure(POC-CF) samples and as a supplemental procedure for106 POC normal karyotype(POC-NK) samples. Chromosomal abnormalities were detected in 10% of POC-CF and pathogenic CNVs were detected in 3.9% of POC-CF and 5.7% of POC-NK samples. Compiled results from this study and relevant case series through a literature review demonstrated an abnormality detection rate(ADR) of 35% for chromosomal abnormalities in POC-CF samples, 3.7% for pathogenic CNVs in POC-CF samples, and 4.6% for pathogenic CNVs in POC-NK samples. Ingenuity Pathway Analysis(IPA) was performed on the genes from pathogenic CNVs found in POC samples. The denoted primary gene networks suggested that apoptosis and cell proliferation pathways are involved in miscarriage. In summary, a similar spectrum of cytogenomic abnormalities was observed in POC culture success and POC-CF samples. A threshold effect correlating the number of dosage-sensitive genes in a chromosome with the observed frequency of autosomal trisomy is proposed. | 13 | Modeling the pressure drop of wet gas in horizontal pipe显示文摘A model for gas–liquid annular and stratified flow through a horizontal pipe is investigated, using the two-phase hydrokinetics theory. Taking into consideration the flow factors including the void fraction, the friction between the two phases and the entrainment in the gas core, the one-dimensional momentum equation for gas has been solved. The differential pressure of the wet gas between the two tapings in the straight pipe has been modeled in the pressure range of 0.1–0.8 MPa. In addition a more objective iteration approach to determine the local void fraction is proposed. Compared with the experimental data, more than 83% deviation of the test data distributed evenly within the band of ± 10%.
