| 1 | Arenobufagin is a novel isoform-specific probe for sensing human sulfotransferase 2A1显示文摘Human cytosolic sulfotransferase 2 A1(SULT2A1) is an important phase II metabolic enzyme. The detection of SULT2A1 is helpful for the functional characterization of SULT2A1 and diagnosis of its related diseases. However, due to the overlapping substrate specificity among members of the sulfotransferase family, it is difficult to develop a probe substrate for selective detection of SULT2A1. In the present study, through characterization of the sulfation of series of bufadienolides, arenobufagin(AB) was proved as a potential probe substrate for SULT2A1 with high sensitivity and specificity. Subsequently, the sulfation of AB was characterized by experimental and molecular docking studies. The sulfate-conjugated metabolite was identified as AB-3-sulfate.The sulfation of AB displayed a high selectivity for SULT2A1 which was confirmed by in vitro reaction phenotyping assays. The sulfation of AB by human liver cytosols and recombinant SULT2A1 both obeyed Michaelis-Menten kinetics, with similar kinetic parameters. Molecular docking was performed to understand the interaction between AB and SULT2A1, in which the lack of interaction with Met-137 and Tyr-238 of SULT2A1 made it possible to eliminate substrate inhibition of AB sulfation. Herein, we design the bis(pyrene)-Lys-Leu-Val-Phe-PheGly-poly ethylene glycol(BP-KLVFFGPEG) based nanoparticles, which capture Aβ_(42) through recognition and co-assembly of KLVFF. The resulting co-assemblies show different morphologies, such as large aggregates, nanoparticles and nanofibers corresponding to the different length chain of PEG, leading to different capture efficiencies. | 6 | Sustained co-delivery of 5-fluorouracil and cis-platinum via biodegradable thermo-sensitive hydrogel for intraoperative synergistic combination chemotherapy of gastric cancer显示文摘Gastric cancer is the fifth most common cancer and the third leading cause of cancer death worldwide,posing a severe threat to human health.Surgical resection remains the most preferred option for gastric cancer treatment.However,for advanced gastric cancer,the curative effect of surgical resection is usually limited by the local recurrence,peritoneal carcinomatosis,or distal metastasis.Intraoperative chemotherapy is an attractive in situ adjuvant treatment strategy to reduce the recurrence and metastasis after surgical resection.Here,we designed a 5-fluorouracil(5-FU)and cis-platinum(DDP)co-delivery system based on a biodegradable temperature-sensitive hydrogel(PDLLA-PEG-PDLLA,PLEL)for intraoperative adjuvant combination chemotherapy of gastric cancer.This 5-FU+DDP/PLEL hydrogel system characterized by a special sol-gel phase transition in response to physiological temperature and presented sustained drug release in vitro and in vivo.A strong synergistic cell proliferation inhibition and apoptosis promotion of 5-FU+DDP/PLEL were observed against gastric cancer MKN45-luc cells.After intraperitoneal injection,the dual-drug loaded hydrogel formulation showed superior anti-tumor effects than the single-drug carrying hydrogels and combination of free 5-FU and DDP on the gastric cancer peritoneal carcinomatosis model.The use of hydrogel for dual-drug delivery had benefited to fewer side effects as well.What’s more,we established a mouse model for postsurgical residual tumors and peritoneal carcinomatosis of gastric cancer,in which the intraoperative administration of 5-FU+DDP/PLEL also remarkably inhibited the local recurrence of the orthotopic tumors and the growth of the abdominal metastatic tumors,resulting in an extended lifetime.Hence,this developed dual-drug loaded hydrogel system has great potential in the intraoperative chemotherapy of gastric cancer,that suggests a clinically-relevant and valuable option for postsurgical management of gastric cancer. | Wen Chen Kun Shi Jie Liu Peipei Yang Ruxia Han Meng Pan Liping Yuan Chao Fang Yongyang Yu Zhiyong Qian | 2023 | Bioactive Materials2023,,5: | 0 |

| 8 | Needle-like cobalt phosphide arrays grown on carbon fiber cloth as a binder-free electrode with enhanced lithium storage performance显示文摘Cobalt phosphide(CoP) is a promising anode candidate for lithium-ion batteries(LIBs) due to its high specific capacity and low working potential.However,the poor cycling stability and rate performance,caused by low electrical conductivity and huge volume variation,impede the further practical application of CoP anode materials.Herein,we report an integrated binder-free electrode featuring needle-like CoP arrays grown on carbon fiber cloth(CC) for efficient lithium storage.The as-prepared CoP/CC electrode integrates the advantages of 1 D needle-like CoP arrays for efficient electrolyte wettability and fast cha rge transpo rtation,and 3 D CC substrate for superior mechanical stability,flexibility and high conductivity.As a result,the CoP/CC electrode delivers an initial specific capacity of 1283 mAh/g and initial Coulombic effeciencies of 85.4%,which are much higher than that of conventional CoP electrode.Notably,the Co P/CC electrode shows outstanding cycling performance up to 400 cycles at 0.5 A/cm^(2) and excellent rate performance with a discharge capacity of 549 mAh/g even at 5 A/cm^(2).This work demonstrates the great potential of integrated CoP/CC hybrid as efficient bind-free and freestanding electrode for LIBs and future flexible electronic devices. | Ze Zhang Peipei Zhu Chao Li Ji Yu Jianxin Cai Zhenyu Yang | 2021 | Chinese Chemical Letters2021,32,1: | 0 |
