3Three-dimensional solution structure of ω-conotoxin SO_(3) determined by ^(1)H NMR显示文摘Cone snails (Conus) elaborate a series of conotoxin (CTX) peptides in their venoms to paralyze their prey. Among these toxins, w-CTX抯 specifically target to presynaptic voltage-gated calcium channel subsets, causing inhibition of neurotransmitter release. w-CTX SO3 was isolated from the venom of Conus striatus, which is the only available fish-hunting snail near the coast of the South China Sea. The three-dimensional solution structure of w-CTX SO3, a peptide which is the only w-conotoxin reported to show high homology with another w-CTX (MVIIA from C. Magus), has been determined by 1H NMR techniques. The molecular structure of w-CTX SO3 is stabilized by three disulfide bridges and a short triple-stranded antiparallel b-sheet with four turns. 4Screening and identification of Shigella flexneri 2a virulence-related genes induced after invasion of epithelial cells显示文摘An in vivo expression technology (IVET) was applied to screen s.flexneri 2a genes induced after invasion of epithelial cells, and virulence-related genes were further identified by mutational analysis. Thirteen intracellular induced genes were identified with a HeLa cell infection model. Of these, two were identified as alkylation-related genes; one was related to metabolism; one encoded a transcriptional regulator; three were identified as insertion elements; three ap- peared to be antisense to genes involved in the transmethylation,biosyntheseis, and phos- photransferase system;and three were predicted to encode polypeptides with unknown functions. Intracellular survival assavs showed that the mutants of alkA,citC and wcaJ genes had lower capability of intracellular replication or survival than the the wild-type strain.The results indicated that alkA, citC and wcaJ genes could take part in the intracellular survival or replication of S. flexneri 2a and the capability of intracellular survival or replication could be one of the major virulence elements. However, the yaiC mutant was able to survive in the murine infection assay but almost not in HeLa cell infection assay. Very possibly, yaiC gene was involved in the other mechanism of S. flexneri virulence. 5Correction of RNA aberrant splice increases foreign gene expression in transgenic mice显示文摘Transgenic mice with mammary gland secreting human granulocyte colony stimulating factor (G-CSF) were produced using mice whey acid protein gene promoter. It was found that there was very low expression level in mammary gland. Human G-CSF cDNA was obtained by RT-PCR from transgenic mice mammary gland. Sequence analysis showed that this G-CSF gene deleted the 4th exon, and compared with human G-CSF genomic DNA, there were donor and acceptor splice sites in the deletion fragment. It was considered that the 3rd and 4th introns also delete in G-CSF fragment. The transgenic construct was corrected by deleting the 3rd and 4th introns to construct the minigene, which was used to produce transgenic mice by microinjection. Northern blot showed that G-CSF expression using the new construct increased 5.4 times as that before in transgenic mice. 6Effect of monoclonal antibody to bull sperm sp18 on murine fertilization and embryos development显示文摘Western blot analysis revealed that one IgG1 monoclonal antibody (mAb) to sp18 family membrane proteins (Mr. 14, 16 and 18 ku) of bovine sperm reacted faintly with protein bands of 14,18, 22, 30 and 60 ku (reducing) in samples of mouse sperm. The mAb also reacted to protein of egg lysozyme. Using a laser confocal microscope, indirect immunofluorescence (IIF) showed that the sp18 antigens were present in the posterior head of murine sperm. In murine in vitro fertilization (IVF) and embryo development trails, a total of 426 oocytes from C57BL/6 and F1 hybrid strain (CD1×C57BL/6 cross) of 12, female mice were used in 3 independent trails. After preincubating capacitated sperm with 182 μg/mL of sp18 mAb in the modified TYH IVF medium for 15—20 min, cumulus-oocyte-complexes were introduced. The fertilization rate in sp18 mAb groups was 77.1%, which was not significantly (P>0.05) different from the nonspecific mouse IgG(79.2 %) and non-IgG(80.3%) control groups. 7Crystallization and preliminary crystallographic studies of insulin T_3R_3 states created in different conditions显示文摘In the presence of a series of small organic molecules including 1, 4 dioxane, DMF, isopropanol, rDNA human insulin, mutants A21 Asn→Ser and (A21 Asn→Ser, B27 Thr→Arg) human insulin have been crystallized in both Zn 2+ containing and Zn 2+ free system. Preliminary crystallographic analyses show that they all belong to T 3R 3 structural type. They are usetul in analgesia, neuroprotection, detection of some kinds of deseases, and receptor and ion channel study. in order to explore the conotoxin resourses of Chinese oceans, rapid amplification of 3’ cDNA ends (RACE) method was utilized to systemically analyze the O-superfamily conotoxin content of Conus striatus inhabited near Chinese Hainan Island.
