1Methylation profile of the promoter CpG islands of 31 genes that may contribute to colorectal carcinogenesis显示文摘AIM: To establish the methylation profile of the promoter CpG islands of 31 genes that might play etiological roles in colon carcinogenesis.METHODS: The methylation specific PCR in conjunction of sequendng verification was used to establish the methylationprofile of the promoter CpG islands of 31 genes in colorectal cancer (n = 65), the neighboring non-cancerous tissues (n = 5), colorectal adenoma (n = 8), and normal mucosa (n = 1). Immunohistochemically, expression of 10 genes was assessed on the home-made tissue microarrays of tissues from 58 patients. The correlation of tumor specific changes with each of clinical-pathologic features was scrutinized with relevant statistic tools.RESULTS: In comparison with the normal mucosa of the non-cancer patients, the following 14 genes displayed no tumor associated changes: breast cancer 1, early onset (BRCA1), cadherin 1, type 1, E-cadherin (epithelial) (CDH1),death-associated protein kinase 1 (DAPK1), DNA (cytosine-5-)-methyltransferase 1 (DNMT1), melanoma antigen, family A, 1 (directs expression of antigen MZ2-E) (MAGEA1), tumor suppressor candidate 3 (N33), cyclin-dependent kinase inhibitor 1A (p21, Cipl) (p21^WAF1), cyclin-dependent kinase inhibitor 1B (p27, 10pl) (p27^WAF1), phosphatase and tensin hornolog (mutated in multiple advanced cancers 1) (PTEN), retinoic acid receptor, beta (RAR-, Ras association (RaIGDS/AF-6) domain family 1 C (RASSFIC), secreted frizzled-related protein 1 (SFRP1), tissue inhibitor of metalloproteinase 3 (Sorsby fundus dystrophy, pseudoinfiammatory) (TIMP3),and von HippeI-Lindau syndrome (VHL). The rest 17 targets exhibited to various extents the tumor associated changes.As changes in methylation of the following genes occurred marginally, their impact on the formation of colorectal cancer were trivial: adenomatous polyposis coli (APC) (8%, 5165),Ras association (RaIGDS/AF-6) domain family 1A (RASSFIA) (3%, 2/65) and cyclin-dependent kinase inhibitor 2A,alternated reading frame Co14~) (6%, 4/65). The following genes exhibited moderate changes in rnethylation: O-6rnethylguanine-DNA rnethyltransferase (MGMT) (20%, 13/65),rnutL hornolog 1, colon cancer, nonpolyposis type 2 (E. coli) (hMLH1) (18%, 12/65), cyclin-dependent kinase inhibitor 2A (melanoma, p16, inhibits CDK4) P16^NK4a) (10%, 10/65),rnethylated in tumor 1 (MINT1) (15%, 10/65), methylated in tumor 31 (MINT31) (11%, 7/65). The rest changed greatly in the rnethylation pattern in colorectal cancer (CRC): cyclin A1 (cyclin al) (100%, 65/65), caudal type homeobox transcriptdon factor 1 (CDX1) (100%, 65/65), RAR(85%, 55/65), myogenic factor 3 (MYOD1) (69%, 45/65),cyclin-dependent kinase inhibitor 2B (p15, inhibits CDK4)(p15^INK4b) (68%, 44/65), prostaglandin-endoperoxide synthase 2 (prostaglandin G/H synthase and cyclooxygenase) (COX2) (72%, 47/65), cadherin 13, H-cadherin (heart) (CDH13) (65%, 42/65), CAAX box 1 (OO~/) (58%, 38/65),tumor protein p73 (p73) (63%, 41/65) and Wilrns tumor 1 (WT/) (58%, 38/65). However, no significant correlation of changes in rnethylation with any given clinical-pathological features was detected. Furthermore, the frequent changes in rnethylation appeared to be an early phase event of colon carcinogenesis. The in situ expression of 10 genes was assessed by the irnrnunohistochernical approach at the protein level: CDH1, CDH13, COX2, cyclin A1, hMLH1,MGMT, p14^ARF, p73, RAR-, and TIMP3 genes in the context of the rnethylation status in colorectal cancer. 2Association of cyclooxygenase-2 expression with Hp-cagA infection in gastric cancer显示文摘AIM: To observe the expression of cyclooxygenase-2 (COX-2) and to investigate the association between COX-2expression and infection with cytotoxic-associated gene A( cagA) positive strair Helicobacter pylori ( Hp) in humangastric cancer, and subsequently to provide fresh ideas forthe early prevention of gastric cancer.METHODS: 32 Specimens of gastric cancer andcorresponding adjacent normal gastric mucosa were obtainedfrom patients who had undergone surgical operations ofgastric cancer. All the samples including 1 case of stomachmalignant lymphoma and 31 cases of gastric adenocarcinomawere confirmed by pathology diagnosis. The expression ofCOX-2 in 32 specimens of gastric cancer and correspondingadjacent normal gastric mucosa was quantitativelydetermined and analyzed with Flow Cytometry, and the levelsof COX-2 protein were compared between specimens withcagA+ Hp infection and those without cagA+ Hp infection.The cagA gene in 32 specimens of gastric cancer wasdetected bypolymerase chain reaction (PCR) method.RESULTS: Twenty-seven of 32 (84 %) specimens of gastriccancer showed over-expression of COX-2, compared withthe adjacent normal gastric mucosa. cagA+ gene weredetected from 19 specimens of gastric cancer, but not fromthe other 13 specimens. The levels of COX-2 protein in 19specimens of gastric cancer with cagA+ Hp infection (thenumber of positive cells was 73.82±18.2) were significantlyhigher than those in the 13 specimens without cagA+ Hpinfection (the number of positive cells was 35.92±22.1).CONCLUSION: COX-2 is overexpressed in gastric cancerand cagA+Hp infection could up-regulate the expression ofCOX-2 in gastric cancer in human. 3Methylation profile of the promoter CpG islands of 14 “drug-resistance”genes in hepatocellular carcinoma显示文摘AIM: To establish the DNA methylation patterns of the promoter CpG islands of 14 'drug-resistance' genes in hepatocellular carcinoma (HCC).METHODS: The methylation specific polymerase chain reaction in conjunction with sequencing verification was used to establish the methylation patterns of the 14 genes in the liver tissues of four healthy liver donors, as well as tumor and the paired non-cancerous tissues of 30 HCC patients.RESULTS: While 11 genes (ATP-binding cassette, sub-family G (WHITE), member 2(ABCG2), activating transcription factor (ATF2), beta-2-microglobulin (B2M), deoxycytidine kinase(DCK, occludin (OCLN, v-raf-1 murine leukemia viral oncogene homolog (RAF/), ralA binding protein 1 (RALBP1),splicing factor (45 kD) (SPF45), S-phase kinase-associated protein 2 (p45) (SKP2), tumor protein p53 (Li-Fraumeni syndrome) (TP53) and topoisomerase (DNA) II beta (TOP2B) maintained the unmethylated patterns, three genes displayed to various extents the hypermethylation state in tumor tissues in comparison with the normal counterparts. The catalase (CAT) was hypermethylated in tumor and the neighboring non-cancerous tissue of one case (3.3%). Both glutathione S-transferase pi (GSTp/) (80%, 24/30 in tumor and 56.7%,17/30 in the paired non-cancerous tissues) and cystic fibrosis transmembrane conductance regulator, ATP-binding cassette (sub-family C, member 7) (CF-FR) (77%, 23/30 in tumor and 50%, 15/30 in the paired non-cancerous tissues) genes were prevalently hypermethylated in HCC as well as their neighboring non-cancerous tissues. No significant difference in the hypermethylation occurrence was observed between the HCC and its neighboring non-cancerous tissues.CONCLUSION: Hypermethylation of promoter CpG islands of both CF-FR and GSTpigenes occurs prevalently in HCC,which may correlate with the low expression of these two genes at the mRNA level and has the profound etiological and clinical implications. 4Current preventive treatment for recurrence after curative hepatectomy for liver metastases of colorectal carcinoma: A literature review of randomized control trials显示文摘To review the preventive approaches for recurrence after curative resection of hepatic metastases from coloreclal carcinoma, we have summarized all available publications reporting randomized control trials (RCTs) covered in PubMed. The treatment approaches presented above include adjuvant intrahepatic arterial infusion chemotherapy,systemic chemotherapy, neoadjuvant chemotherapy, and immunotherapy. Although no standard treatment has been established, several approaches present promising results, which are both effective and tolerable in posthepatectomy patients. Intrahepatic arterial infusion chemotherapy should be regarded as effective and tolerable and it increases overall survival (OS) and diseasefree survival (DFS) of patients, while 5-fluorouracil-based systemic chemotherapy has not shown any significant survival benefit. Fortunately chemotherapy combined with hepatic arterial infusion and intravenous infusion has shown OS and DFS benefit in many researches. Few neoadjuvant RCT studies have been conduced to evaluate its effect on prolonging survivals although many retrospective studies and case reports are published in which unresectable colorectal liver metastases are downstaged and made resectable with neoadjuvant chemotherapy.Liver resection supplemented with immunotherapy is associated with optimal results; however, it is also questioned by others. In conclusion, several adjuvant approaches have been studied for their efficacy on recurrence after hepatectomy for liver metastases from colorectal cancer (CRC), but multi-centric RCT is still needed for further evaluation on their efficacy and systemic or local toxicities. 6The Arabidopsis phosphatase PP2C49 negatively regulates salt tolerance through inhibition of At HKT1;1显示文摘Type 2C protein phosphatases(PP2Cs)are the largest protein phosphatase family.PP2Cs dephosphorylate substrates for signaling in Arabidopsis,but the functions of most PP2 Cs remain unknown.Here,we characterized PP2 C49(AT3G62260,a Group G PP2C),which regulates Na^(+)distribution under salt stress and is localized to the cytoplasm and nucleus.PP2C49 was highly expressed in root vascular tissues and its disruption enhanced plant tolerance to salt stress.Compared with wild type,the pp2c49 mutant contained more Na^(+)in roots but less Na^(+)in shoots and xylem sap,suggesting that PP2C49 regulates shoot Na^(+)extrusion.Reciprocal grafting revealed a root-based mechanism underlying the salt tolerance of pp2 c49.Systemic Na+distribution largely depends on AtHKT1;1 and loss of function of AtHKT1;1 in the pp2c49 background overrode the salt tolerance of pp2c49,resulting in salt sensitivity.Furthermore,compared with plants overexpressing PP2C49 in the wild-type background,plants overexpressing PP2C49 in the athtk1;1 mutant background were sensitive to salt,like the athtk1;1 mutants.Moreover,protein-protein interaction and two-voltage clamping assays demonstrated that PP2C49 physically interacts with AtHKT1;1 and inhibits the Na^(+)permeability of AtHKT1;1.This study reveals that PP2C49 negatively regulates AtHKT1;1 activity and thus determines systemic Na^(+)allocation during salt stress.Moli Chu Pengwang Chen Sufang Meng Peng Xu Wenzhi Lan 2021Journal of Integrative Plant Biology2021,63,3:5

8A novel protein-DNA interaction involved with the CpG dinucleotide at -30 upstream is linked to the DNA methylation mediated transcription silencing of the MAGE-A1 gene显示文摘To understand the DNA-methylation mediated gene silencing mechanisms, we analyzed in cell culture of the promoter function of the MAGE-A1 gene, which is frequently demethylated and over-expressed in human hepatocellular carcinoma. We have established the correlation of the DNA methylation of the promoter CpG island with expression status of this gene in a panel of the established liver cancer cell lines. The crucial CpG dinucleotide(s) within the minimal promoter subjected to the control mediated by DNA methylation with profound biological functions was also delineated.Furthermore, a novel sequence-specific DNA-protein interaction at the -30 CpG dinucleotide upstream of the gene was found having a vital part to play in the DNA methylation mediated transcription silencing of the MAGE-A1 gene. 9Construction of a novel Shigella live-vector strain co-expressing CS3 and LTB/STm of enterotoxigenic E.coli显示文摘AIM: To construct and evaluate a polyvalent recombinant vaccinie strain Shigella flexneri2a T32 against enterotoxigenic E.coli(ETEC).METHODS: By using a host-plasmid balanced lethal system based on asdgene, a polyvalent recombinant strain was constructed to highly express CS3 and regularly express fusion enterotoxin of LTB subunit and mutant ST (LTB/STm) in a vaccine strain Shigella flexneri 2a T32with specific deletion of asd gene. Fimbria CS3 was observed by immunofluorescence and electron microscopy assay. The security of LTB/STm was examined by ileal loop assay and suckling mouse assay. To evaluate this new candidate vaccine, it was compared with a previous vaccine strain in plasmid and protein level, growth assay and immunogenicity in Balb/c mice.RESULTS: The newly constructed vaccine expressed CS3 and grew better than the previously constructed vaccine except for the lower expression of LTB/STm. Serum IgG and mucosal IgA against CS3, LTB, ST, and host lipopolysaccharide (LPS) were produced after immunization of Balb/c mice by oral route with the new strain. 13Mercury(Ⅱ) detection by water-soluble photoluminescent ultra-small carbon dots synthesized from cherry tomatoes显示文摘Mercury ions have been considered highly toxic to human health. What would be great is to develop the ionic probes without any toxicities themselves. Here, we report a friendly, highly sensitive mercury(II) ionic probe, watersoluble photoluminescence carbon dots which were synthesized by simply hydrothermal treatment of fresh cherry tomatoes without adding any other reagents. The ultra-small(\1 nm) carbon dots show robust excitation-depended photoluminescence under a wide p H range(4–10) or a strong ionic strength of up to 1 M, and the detection limit of mercury(II) has been determined as low as 18 n M. 18Identifying critical factors that affect the application of information technology in construction management: A case study of China显示文摘The new mode for managing construction projects with information technology(IT)has attracted worldwide attention because it can help managers and workers perform tasks and bring potential benefits,such as high-quality products and accident-free production,effectively.However,the application of IT in site have not achieved expected results because it is faced with many constraints caused by internal factors from enterprises and projects and external factors from the government and environment.Although many relevant studies have discussed the constraints of implementing different IT and devices in the construction industry or site,few articles have specifically focused on identifying and analyzing the indicator system.In this work,we took China as the background,scientifically identified 23 influential factors that affect the implementation of IT in construction management through literature review and expert interviews.Subsequently,questionnaires were issued,and Delphi method was used to obtain empirical data that aimed at four different management fields.Then,an efficient and convenient method called DEMATEL was used to deal with these data.Afterward,the factors were divided into four categories,namely,core,diving,independent,and impact factors.Finally,the similarities and differences of the analysis results from the four fields were compared,and the key factors were identified.Results show that the cross-domain talent ability,concept and value cognition,and organization structure are core factors in all management fields that should be managed first along with the IT innovation ability in the enterprise.The formulation of technical standards and related device and training input are also critical in specific fields.Strategic planning plays a role in macro control and promotion.Data management and application,platform construction,solution,and collaboration have direct impacts on information management.The research results provide suggestions not only for the government to formulate effective policies for IT application and promotion in construction industry,but also for enterprises to take measures in improving management efficiency in the construction site and realizing its substantial benefits.Meishan JIA Youquan XU Pengwang HE Lingmin ZHAO 2022Frontiers of Engineering Management2022,9,2:1
