1CRISPR-Cas adaptive immune systems in Sulfolobales:genetic studies and molecular mechanisms显示文摘CRISPR-Cas systems provide the small RNA-based adaptive immunity to defend against invasive genetic elements in archaea and bacteria.Organisms of Sulfolobales,an order of thermophilic acidophiles belonging to the Crenarchaeotal Phylum,usually contain both type I and typeⅢCRISPR-Cas systems.Two species,Saccharolobus solfataricus and Sulfolobus islandicus,have been important models for CRISPR study in archaea,and knowledge obtained from these studies has greatly expanded our understanding of molecular mechanisms of antiviral defense in all three steps:adaptation,expression and crRNA processing,and interference.Four subtypes of CRISPR-Cas systems are common in these organisms,including I-A,I-D,Ⅲ-B,andⅢ-D.These cas genes form functional modules,e.g.,all genes required for adaptation and for interference in the I-A immune system are clustered together to form aCas and i Cas modules.Genetic assays have been developed to study mechanisms of adaptation and interference by different CRISPR-Cas systems in these model archaea,and these methodologies are useful in demonstration of the protospacer-adjacent motif(PAM)-dependent DNA interference by I-A interference modules and multiple interference activities byⅢ-B Cmr systems.Ribonucleoprotein effector complexes have been isolated for SulfolobalesⅢ-B andⅢ-D systems,and their biochemical characterization has greatly enriched the knowledge of molecular mechanisms of these novel antiviral immune responses.Zhenxiao Yu Suping Jiang Yuan Wang Xuhui Tian Pengpeng Zhao Jianan Xu Mingxia Feng Qunxin She 2021Science China(Life Sciences)2021,64,5:2
2Rapid response of brain metastasis to crizotinib in a patient with KLC1-ALK fusion and MET gene amplification positive non-small cell lung cancer:a case report显示文摘Non-small cell lung cancer(NSCLC) ranks as the leading cause of cancer-related death in the world. Brain metastasis(BM) is a common complication of NSCLC, with 25%–40% of patients developing BM during the course of the disease. A significant strategy of local disease control in the central nervous system is radiation therapy. With the development of precision medicine,the concept of treating lung cancer BM has gradually changed. In this case, we performed a surgical procedure to obtain enough tumor tissue for the detection of the target gene and other related experiments after the patient was informed. Finally, we found that the patient had both hepatocyte growth factor receptor(MET) gene amplification and kinesin light chain 1-anaplastic lymphoma kinase fusion(KLC1-ALK) through next-generation sequencing and showed sensitivity to the targeted therapy of crizotinib. The patient exhibited good response. 3Study on the erosion characteristics of copper-carbon electrode pairs by DC air arc显示文摘As common electric contact materials,copper-carbon electrode pairs are widely used in the field of electric contact such as in high-speed railways,motor collector rings and grounding brushes.They play a key role in energy transmission in the above-mentioned systems.Arc erosion has become a key issue threatening system safety.In this work,a dedicated arc erosion platform was built to study the polarity effect of copper-carbon electrode pairs.It was found that arc dynamics and electrodes erosion behaviours were significantly different under different polarities,which could be understood from the mechanism of heat transfer,cooling and electron emission of the electrode.Then,when the carbon material was used as an anode,the arc caused carbon transfer:the carbon material sublimated quickly due to electron condensation and thermal conduction and quickly condensed on the low-temperature metal electrode,while the carbon material transfer could partly inhibit the oxidation of the cathode metal electrode.In addition,the rapid sublimation of the carbon material produced high-temperature airflow,which would make the arc impedance fluctuate and cause the arc voltage to jump.Finally,the anode heat transfer and cathode cooling mechanism directly affected the material state,and the erosion of the anode material was more serious。Wenhan Xie Guangning Wu Zefeng Yang Pengpeng She Hong Wang Haozi Zuo Wenfu Wei Guoqiang Gao Chuanjun Tu 2021High Voltage2021,6,4:1
