2Blood flow restriction in human skeletal muscle during rest periods after high-load resistance training down-regulates miR-206 and induces Pax7显示文摘Backgroud:Blood flow re striction(BFR) with low-intensity re sistance training has been shown to result in hypertrophy of skeletal muscle.In this study,we tested the hypothesis that BFR during the rest periods between acute,high-intensity resistance exercise sessions(70% of 1 repetition maximum,7 sets with 10 repetitions) enhances the effects of the resistance training.Methods:A total of 7 healthy young men performed squats,and between sets BFR was carried out on one leg while the other leg served as a control.Because BFR was applied during rest periods,even severe occlusion pressure(approximately 230 mmHg),which almost completely blocked blood flow,was well-tolerated by the participants.Five muscle-specific microRNAs were measured from the biopsy samples,which were taken 2 h after the acute training.Results:Doppler data showed that the pattern of blood flow recovery changed significantly between the first and last BFR.microRNA-206 levels significantly decreased in the BFR leg compared to the control.The mRNA levels of RAC-β serine/threonine-protein kinase v22,nuclear re spiratory factor 1,vascular endothelial growth factor,lupus Ku autoantigen protein p70 genes(p <0.05),and paired box 7(p <0.01) increased in the BFR leg.The protein levels of paired box 7,nuclear respiratory factor 1,and peroxisome proliferator-activated receptor y coactivator 1α did not differ between the BFR leg and the control leg.Conclusion:BFR,during the rest periods of high-load resistance training,could lead to mRNA elevation of those proteins that regulate angiogenesis,mitochondrial biogenesis,and muscle hypertrophy and repair.However,BFR also can cause DNA damage,judging from the increase in mRNA levels of lupus Ku autoantigen protein p70.Ferenc Torma Zoltan Gombos Marcell Fridvalszki Gergely Langmar Zsofia Tarcza Bela Merkely Hisashi Naito Noriko Ichinoseki-Sekine Masaki Takeda Zsolt Murlasits Peter Osvath Zsolt Radak 2021Journal of Sport and Health Science2021,10,4:3

5Reduced expression of Ca^(2+)-regulating proteins in the upper gastrointestinal tract of patients with achalasia显示文摘AIM: To compare expression of Ca2+-regulating proteins in upper gastrointestinal (GI) tract of achalasia patients and healthy volunteers and to elucidate their role in achalasia. METHODS: Sarcoplasmic reticulum Ca2+ ATPase (SERCA) isoforms 2a and 2b, phospholamban (PLB), calsequestrin (CSQ), and calreticulin (CRT) were assessed by quantitative Western blotting in esophagus and heart of rats, rabbits, and humans. Furthermore, expression profi les of these proteins in biopsies of lower esophageal sphincter and esophagus from patients with achalasia and healthy volunteers were analyzed. RESULTS: SERCA 2a protein expression was much higher in human heart (cardiac ventricle) compared to esophagus. However, SERCA 2b was expressed predominantly in the esophagus. The highest CRT expression was noted in the human esophagus, while PLB, although highly expressed in the heart, was below our detection limit in upper GI tissue. Compared to healthy controls, CSQ and CRT expression in lower esophageal sphincter and distal esophageal body were signif icantly reduced in patients with achalasia (P < 0.05). CONCLUSION: PLB in the human esophagus mightbe of lesser importance for regulation of SERCA than in heart. 13Blood flow restriction during the resting periods of high-intensity resistance training does not alter performance but decreases MIR-1 and MIR-133A levels in human skeletal muscle显示文摘Blood flow restriction(BFR)during exercise bouts has been used to induce hypertrophy of skeletal muscle,even with low loads.However,the effects of BFR during the rest periods between sets are not known.We have tested the hypothesis that BFR during rest periods between sets of high-intensity resistance training would enhance performance.Twenty-two young adult male university students were recruited for the current study,with n=11 assigned to BFR and n=11 to a control group.The results revealed that four weeks training at 70%of 1 RM,five sets and 10 repetitions,three times a week with and without BFR,resulted in similar progress in maximal strength and in the number of maximal repetitions.The miR-1 and miR-133a decreased significantly in the vastus lateralis muscle of BFR group compared to the group without BFR,while no significant differences in the levels of miR133b,miR206,miR486,and miR499 were found between groups.In conclusion,it seems that BFR restrictions during rest periods of high-intensity resistance training,do not provide benefit for enhanced performance after a four-week training program.However,BFR-induced downregulation of miR-1 and miR-133a might cause different adaptive responses of skeletal muscle to high intensity resistance training.Ferenc Torma Peter Bakonyi Zsolt Regdon Zoltan Gombos Matyas Jokai Gergely Babszki Marcell Fridvalszki Laszl Virg Hisashi Naito SyedRehan Iftikhar Bukhari Zsolt Radak 2021Sports Medicine and Health Science2021,3,1:0

Characterization of the interactions between hepatic stellate cells and tumor cells during uveal melanoma metastatic progression显示文摘Background:Uveal melanoma(UM)is the most common primary eye tumor in adults,and the most frequent site of malignant transformation of the melanocytes after the skin.It spreads to the liver in half of the cases,and the death rate following the report of metastasis is 92%at 2 years.Hepatic tropism of UM cannot simply be explained by the blood circulatory system organization,and illustrates the“seed and soil”hypothesis that describes an interaction between tumor cells(seed)and a specific microenvironment(soil).We decided to focus our study on the synergic interaction between UM cells and hepatic stellate cells(HSC),whose role has been previously described in the metastatic progression of colon and pancreatic cancers.Furthermore,HSC have been found surrounding UM liver metastasis,and the UM secretome contains activating cytokines of hepatic stromal cells.Our hypothesis is that HSC provide a specific microenvironment in the liver enhancing the growth of UM cells and increasing their therapeutic resistance.Using an in vitro 3D model and an original xenograft mouse model,we aim to decipher the mechanisms of UM metastatic progression,in order to elaborate new therapeutic strategies.Methods:First,using an agar coating,spheroids were generated with UM cells and were allowed to grow for 72 h.These tumor spheroids were then embedded in Matrigel and the HSC conditioned medium was used to evaluate the impact of the HSC secretome on UM invasion.Next,an original in vivo xenograft mouse model was generated,in which metastatic UM cells were injected alone or with human HSC in the spleen of immunodeficient mice.This model allows us to evaluate in 3-6 weeks the metastatic potential of each cell population,and thus to determine the cooperation between HSC and UM cells in the liver.Results:The HSC conditioned medium increased the invasion of UM spheroids compared to non-conditioned medium in our in vitro model.In addition,UM cells inoculated in the mouse spleen alone or with human HSC were able to metastasize to the liver,and the host HSC were also recruited by UM metastases.Conclusions:Our preliminary results strongly suggest that the secretome of HSC provides a permissive microenvironment for UM metastatic progression.We now have to confirm these results by characterizing the secretome of HSC,in order to identify cytokines or growth factors that increase the invasion of the liver by UM.Our models can be used to test the efficacy of new therapeutic strategies targeting the UM microenvironment.Léo Piquet William Pelletier Peter Gerges Julie Bérubé Solange Landreville 2018Annals of Eye Science2018,,1:0
15Peeling off the surface: Pt-decoration of WSe 2 nanoflakes results in exceptional photoelectrochemical HER activity显示文摘Photoelectrochemical(PEC)hydrogen evolution reaction(HER)was studied on exfoliated,pristine and Pt-decorated tungsten diselenide(p-WSe_(2))nanoflake samples,using a previously developed microdroplet PEC microscopy approach.The WSe 2 nanoflakes had well-defined thicknesses as measured by atomic force microscopy,and the Pt nanoparticles(NPs)were deposited by a variable num-ber of atomic layer deposition(ALD)cycles.An exceptionally high photocurrent density of 49.6 mA cm^(−2)(under 220 mW cm^(−2) irradiation)and internal-photon-to-electron-conversion efficiency(∼90%at 550 nm)were demonstrated on these Pt-decorated WSe_(2)(WSe_(2)-Pt)photocathodes.The Pt NP loading and thickness of WSe_(2) nanoflakes(in the 24–235 nm range)were used to fine-tune their PEC activity for HER.We found similar charge transfer and surface recombination kinetics of pristine and WSe_(2)-Pt specimens(as assessed by intensity-modulated photocurrent spectroscopy),which indicated significant differences in their bulk properties.X-ray and ultraviolet photoelectron spectroscopies were performed to identify defect states and quantify the density of states around the valence band of WSe_(2).The elevated temperature of the ALD process and the evolving Pt NP phase conspired to passivate the sub-surface(i.e.,bulk)defects in the WSe_(2) nanoflakes,resulting in their vastly improved PEC performance.Peter S.Toth Gabor Szabo Gabor Bencsik Gergely F.Samu Krishnan Rajeshwar Csaba Janaky 2022SusMat2022,2,6:0

17On the origin of X-shaped radio galaxies显示文摘After a brief,critical review of the leading explanations proposed for the small but important subset of radio galaxies showing an X-shaped morphology (XRGs) we propose a generalized model,based on the jet-shell interaction and spinflip hypotheses.The most popular scenarios for this intriguing phenomenon invoke either hydrodynamical backflows and over-pressured cocoons or rapid jet reorientations,presumably from the spin-flips of central engines following the mergers of pairs of galaxies,each of which contains a supermassive black hole.We confront these models with a number of key observations,and thus argue that none of the models is capable of explaining the entire range of the salient observational properties of XRGs,although some of the arguments raised in the literature against the spin-flip scenario are probably not tenable.We then propose a new scenario which also involves galactic mergers but would allow the spin of the central engine to maintain its direction.Motivated by detailed multi-band observations of the nearest radio galaxy,Centaurus A,this new model emphasizes the role of the interactions between the jets and the shells of stars and gas that form and rotate around the merged galaxy and can cause temporary deflections of the jets,occasionally giving rise to an X-shaped radio structure.Although each model is likely to be relevant to a subset of XRGs,the bulk of the evidence indicates that most of them are best explained by the jet-shell interaction or spin-flip hypotheses.Gopal-Krishna Peter L.Biermann Lszló .Gergely Paul J.Wiita 2012Research in Astronomy and Astrophysics2012,12,2:0
