| 1 | Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. | 2 | Stress-induced CsPbBr3 nanocrystallization on glass surface:Unexpected mechanoluminescence and applications显示文摘In this work,we discovered an unexpected mechanoluminescence (ML) phenomena occurring when transforming amorphous into crystalline,due to the stress-induced precipitation of CsPbBr3 perovskite nanocrystals on glass surface.It is revealed that,unlike the conventional thermal-induced phase transformation mechanism,the breakage of bonding of glass network provides the energy for nucleation and growth,and the shear stress avoids the long-range migration of structural units for crystallization.Such unique ML phenomenon enables the visualization of dynamical force that is inaccessible by common strategy,and so,opens up some novel applications,such as the pressure-sensitive 'glassy pencil' to learn people's writing habits,and the pb^2+-detection with good sensitivity and selectivity.These findings not only demonstrate an effective route for the preparation of perovskite materials in a green,time-saving,low cost,and scalable way,enrich the knowledge of glass crystallization mechanism,but also exploit a useful avenue to quantitatively visualize the dynamical force. | Xiaoqiang Xiang Hang Lin Renfu Li Yao Cheng Qingming Huang Ju Xu Congyong Wang Xueyuan Chen Yuansheng Wang | 2019 | Nano Research2019,12,5: | 8 |
| 3 | Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery. | Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang | 2020 | Light(Science & Applications)2020,9,1: | 6 |

| 5 | Metal-Organic Frameworks Offering Tunable Binary Active Sites toward HighlyEfficient Urea Oxidation Electrolysis显示文摘Electrocatalytic urea oxidation reaction(UOR)is regarded as an effective yet challenging approach for the degradation of urea in wastewater into harmless N2 and CO_(2).To overcome the sluggish kinetics,catalytically active sites should be rationally designed to maneuver the multiple key steps of intermediate adsorption and desorption.Herein,we demonstrate that metal-organic frameworks(MOFs)can provide an ideal platform for tailoring binary active sites to facilitate the rate-determining steps,achieving remarkable electrocatalytic activity toward UOR.Specifically,the MOF(namely,NiMn_(0.14)-BDC)based on Ni/Mn sites and terephthalic acid(BDC)ligands exhibits a low voltage of 1.317 V to deliver a current density of 10 mA cm^(-2).As a result,a high turnover frequency(TOF)of 0.15 s^(-1) is achieved at a voltage of 1.4 V,which enables a urea degradation rate of 81.87%in 0.33 M urea solution.The combination of experimental characterization with theoretical calculation reveals that the Ni and Mn sites play synergistic roles in maneuvering the evolution of urea molecules and key reaction intermediates during the UOR,while the binary Ni/Mn sites in MOF offer the tunability for electronic structure and d-band center impacting on the intermediate evolution.This work provides important insights into active site design by leveraging MOF platform and represents a solid step toward highly efficient UOR with MOF-based electrocatalysts. | Xuefei Xu Qingming Deng Hsiao-Chien Chen Muhammad Humayun Delong Duan Xia Zhang Huachuan Sun Xiang Ao Xinying Xue Anton Nikiforov Kaifu Huo Chundong Wang Yujie Xiong | 2022 | Research2022,,4: | 0 |
| 6 | Identification and Analysis on True or False Hybrids from Hybridization Populations of Litchi( Litchi chinensis Sonn.) Using EST-SSR Markers显示文摘In order to better distinguish true hybrids in litchi crossbreeding,F1generations of two litchi hybridized combinations namely that‘Xuehuaizi’בGuiwei’and‘Xuehuaizi’בJiaohesanyuehong’were used as materials to identify true hybrids,to construct mapping populations and to study the genetic diversities of the two populations with EST-SSR markers.The results showed that F1generations of the two hybrid populations could be identified with an identification rate of100%by the combination of four pair primers,respectively,and the 159 individual plants of the two populations were true hybrids.In addition,variations were exited in leaf morphology of the progenies of the two populations and bands of parents absented were occurred on genotype.The clustering analysis showed that 113 F1plants from the hybridized combination of‘Xuehuaizi’בGuiwei’were clustered into six categories(similarity coefficient was 0.68),and 63.72%(72 plants)of which clustered into one group with male parent.The genetic distances between 32 plants(28.3%)and their parents were far which indicated that larger variation or recombinant appeared in these plants.Forty six hybrid progenies of the combination of‘Xuehuaizi’בJiaohesanyuehong’were divided into two categories when the similarity coefficient was 0.642 and most individual plants(60.87%)showed closer genetic relationship with female parent and partial maternal genetic tendency.It is concluded that EST-SSR markers are suitable to identify true hybrids of litchi.The construction of the two F1mapping populations has established basis for further genetic linkage mapping,meanwhile,has accumulated materials for cultivar improvement of litchi. | Qingming SUN Shuaipeng MA We LIU Nonhui JIANG Jing FANG Xiaoyan YANG Xu XIANG | 2015 | Agricultural Biotechnology2015,4,2: | 0 |
| 7 | Characteristics of homologous recombination repair pathway genes mutation in ovarian cancers显示文摘Background:Few studies have investigated the characteristics of non‐BRCA homologous recombination repair(HRR)pathway somatic mutations,and the impact of these mutations on efficacy of treatment in ovarian cancer patients is not clear.Therefore,we conducted this study to analyze the frequency and spectrum of somatic mutations in HRR pathway genes in patients with ovarian cancer and to examine the relationships between somatic mutations in HRR pathway genes and their effects on the efficacy of platinum‐based chemotherapy.Methods:We performed targeted sequencing of 688 genes related to the occurrence,development,treatment,and prognosis of solid tumors.Somatic mutations were identified by paired analysis of tumor tissue and germline DNA in blood cells.Results:A total of 38 patients with ovarian cancer were included in the study,and 35(92.1%)patients were diagnosed with high‐grade serous carcinoma.All patients exhibited somatic mutations in the tumor tissue samples.The commonly mutated genes were TP53(73.7%),BRCA2(55.3%),NF1(52.6%),BRCA1(47.4%),and CDH1(47.4%).Overall,71.1%of the patients exhibited mutation in at least one HRR pathway gene.The most frequently altered HRR genes were BRCA2(55.3%),followed by BRCA1(47.4%),ATM(44.7%),BARD1(42.1%),and CHEK1(36.8%).The median progression‐free survival(PFS)in patients with HRR pathway mutation was 36.0 months compared with 13.6 months in patients with no HRR pathway mutation(hazard ratio[HR],0.25;95%confidence interval[CI],0.08–0.77;p=0.016).Patients harboring BRCA1/2 and/or CDK12 mutations displayed a longer PFS(median,36.0 months)compared with patients with no BRCA1/2 or CDK12 mutation(median,13.6 months;HR,0.21;95%CI,0.07–0.61;p=0.004).In multivariate analysis Cox proportional hazards models,after adjustment for tumor stage at diagnosis and histology of initial diagnosis,patients with HRR pathway mutation had a longer PFS than patients with HRR wild‐type genes(p=0.006).Conclusions:HRR pathway somatic mutations are common in Chinese patients with ovarian cancer.HRR pathway somatic mutations were associated with improved sensitivity to platinum-based chemotherapy.Large-scale prospective studies are needed to verify our findings. | Zongbi Yi Min Chen Shaoxing Sun Chunxu Yang Zijie Mei Hui Yang Qingming Xiang Hui Qiu | 2022 | Cancer Innovation2022,1,3: | 0 |
