| 1 | 东亚钳蝎昆虫毒素BmK IT的初级结构研究显示文摘从中国产东亚钳蝎(Buthus martensi Karsch)毒中纯化的一个昆虫毒素BmK IT经还原和羧甲基化处理后,分别用TPCK-胰蛋白酶和金黄色葡萄球菌蛋白酶进行用TLC-薄膜分离酶解的肽段并分别测定了它们的氨基酸顺序。将BmK IT及其酶解肽段的氨基酸顺序的测定结果与由非洲产蝎Androctonus australis Hector毒中纯化的昆虫毒素AaH IT的初级结构进行参照对比,我们发现两种昆虫毒素的分子结构中仅有16个残基部位发生了变异,其同源性高达75%以上。这一结果为探讨蝎昆虫毒素的分子结构特征以及结构与功能的相互关系提供了重要资料,也为这类毒素的开发利用,如作为神经生物科学研究中的工具物或探讨作为生物杀虫剂的可能性等提供了必要的分子基础。 | 吉永华 P. | 2 | Amino Acid Sequence of an Excitatory Insect-selective Toxin (BmK IT) From Venom of the Scorpion Buthus martensi Karsch显示文摘The insect-selective neurotoxin(BmK IT) of scorpion Buthus martensi Karsch was first reduced and S-alkylated, and then digested by TPCK-trypsin and Staphylococcus aureus V-8 Protease. The enzymatic peptides were purified on TLC-plastic sheet and submitted to determine their amino acid compositions and sequences. The sequence of the 70 amino acid residues of BmK IT was established with reference to the primary structure of AaH IT, another excitatory insect-selective toxin from the venom of North African scorpion Androctonus australis Hector. About 75% of the homologous sequence was found in the molecules of BmK IT and AaH IT. It is obvious that the results contribute toward better understanding of the molecular structure characteristics, structure/activity relationship of scorpion insect-selective toxins, and they can serve as the molecular basis for utilizing the toxins as a tool to clarify molecular mechanism involved in channel gating, and to infer the possibility of developing them as new selective | 吉永华 P. | 4 | bFGF诱导MEF生成的条件培养基对人胚胎干细胞生长的影响显示文摘为了探讨低浓度bFGF诱导小鼠胚胎成纤维细胞(MEF)生成的条件培养基对人胚胎干细胞(hESC)生长分化的影响,以系列浓度的bFGF作用于MEF上,收集条件培养基(bFGF-MCM),用于hES2细胞的无滋养层培养。以不添加bFGF而收集的MEF条件培养基(MCM)为阴性对照,同样浓度的bFGF添加于SR培养基(bFGF-SR)为空白对照。通过形态学特征和碱性磷酸酶染色法对hES2的生长分化状态进行评估。结果发现,培养一周内,未分化hES2克隆的比率,阴性对照为23%;空白对照组为13%-31%。当bFGF浓度为0.1,0.3,1,4ng/ml时,bFGF-MCM组未分化克隆的比率分别为44%,74%,77%和78%,与阴性和空白对照组相比,未分化克隆的比率均有不同程度提高,差异有统计学意义(P<0.01)。该结果揭示了经bFGF诱导的MEF细胞所产生的条件培养基具备了维持hES细胞正常生长而不分化的能力。对bFGF-MCM的深入分析,有望更好地了解hESC的生长与分化机制。 | 周轶平 ROCHAT Anne HATZFELD Antoinette PEIFFER Isabelle BARBET Romain HATZFELD Jacques 李玛琳 | 2009 | 分子细胞生物学报2009,42,3: | 5 |
| 5 | Disagreement between symptom-reflux association analysis parameters in pediatric gastroesophageal reflux disease investigation显示文摘AIM: To assess the agreement within 3 commonly used symptom-reflux association analysis (SAA) parameters investigating gastroesophageal reflux disease (GERD) in infants. METHODS: Twenty three infants with suspected GERD were included in this study. Symptom index (SI), Symptom sensitivity index (SSI) and symptom association probability (SAP) related to cough and irritability were calculated after 24 h combined pH/multiple intraluminal impedance (MII) monitoring. Through defined cutoff values, SI, SSI and SAP values are differentiated in normal and abnormal, whereas abnormal values point towards gastroesophageal reflux (GER) as the origin of symptoms. We analyzed the correlation and the concordance of the diagnostic classification of these 3 SAA parameters.RESULTS: Evaluating the GER-irritability association, SI, SSI and SAP showed non-identical classification of normal and abnormal cases in 39.2% of the infants. When irritability was taken as a symptom, there was only a poor inter-parameter association between SI and SSI, and between SI and SAP (Kendall’s tau b = 0.37, P < 0.05; Kendall’s tau b = 0.36, P < 0.05, respectively). Evaluating the GER-cough association, SI, SSI and SAP showed non-identical classification of normal and abnormal cases in 52.2% of the patients. When cough was taken as a symptom, only SI and SSI showed a poor inter-parameter association (Kendall’s tau b = 0.33, P < 0.05). CONCLUSION: In infants investigated for suspected GERD with pH/MII-monitoring, SI, SSI and SAP showed a poor inter-parameter association and important dis-agreements in diagnostic classification.
