| 10 | Closed rhinoplasty:effects and changes on voice-a preliminary report显示文摘Aim:Effects of rhinoplasty were already studied from many points of view:otherwise poor is scientific production focused on changes of voice after rhinoplasty.This preliminary study analyzed objectively and subjectively these potential effects on 19 patients who underwent exclusively closed rhinoplasty.Methods:This preliminary evaluation was conducted from September 2012 to May 2013 and 19 patients have undergone primary rhinoplasty with exclusively closed approach(7 males,12 females).All patients were evaluated before and 6 months after surgery.Each of them answered to a questionnaire(Voice Handicap Index Score)and the voice was recorded for spectrographic analysis:this system allowed to perform the measurement of the intensity and frequency of vowels(“A”and“E”)and nasal consonants(“N”and“M”)before and after surgery.Data were analysed with the Mann-Whitney test.Results:Sixteen patients showed statistically significant differences after surgery.It was detected in 69%of cases an increased frequency of emission of the consonant sounds(P=0.046),while in 74%of cases the same phenomenon was noticed for vowel sounds(P=0.048).Conclusion:Many patients who undergo rhinoplasty think that the intervention only leads to anatomical changes and improvement of respiratory function.The surgeon should instead accurately inform patients about the potential effects on the voice.This preliminary study reveals the significant effects of closed rhinoplasty on the human voice. | Giuseppe Guarro Romano Maffia Barbara Rasile Carmine Alfano | 2016 | Plastic and Aesthetic Research2016，3，1： | 0 |
| 11 | Epithelial-to-mesenchymal transition in pancreatic ductal adenocarcinoma: Characterization in a 3D-cell culture model显示文摘AIM: To analyze the effect of three-dimensional(3D)-arrangement on the expression of epithelial-tomesenchymal transition markers in pancreatic adenocarcinoma(PDAC) cells.METHODS: HPAF-Ⅱ, HPAC, and PL45 PDAC cells were cultured in either 2D-monolayers or 3D-spheroids. Ultrastructure was analyzed by transmission electron microscopy. The expression of E-cadherin, β-catenin, N-cadherin, collagen type Ⅰ(COL-I), vimentin, α-smooth muscle actin(αSMA), and podoplanin was assayed by confocal microscopy in cells cultured on 12-mm diameter round coverslips and in 3D-spheroids. Gene expression for E-cadherin, Snail, Slug, Twist, Zeb1, and Zeb2 was quantified by real-time PCR. E-cadherin protein level and its electrophoretic pattern were studied by Western blot in cell lysates obtained from cells grown in 2D-monolayers and 3D-spheroids. RESULTS: The E-cadherin/β-catenin complex was expressed in a similar way in plasma membrane cell boundaries in both 2D-monolayers and 3D-spheroids. E-cadherin increased in lysates obtained from 3D-spheroids, while cleavage fragments were more evident in 2D-monolayers. N-cadherin expression was observed in very few PDAC cells grown in 2D-monolayers, but was more evident in 3D-spheroids. Some cells expressing COL-I were observed in 3D-spheroids. Podoplanin, expressed in collectively migrating cells, and αSMA were similarly expressed in both experimental conditions.
