| 1 | Autumn Photoproduction of Carbon Monoxide in Jiaozhou Bay, China显示文摘Carbon monoxide(CO) plays a significant role in global warming and atmospheric chemistry. Global oceans are net natural sources of atmospheric CO. CO at surface ocean is primarily produced from the photochemical degradation of chromophoric dissolved organic matter(CDOM). In this study, the effects of photobleaching, temperature and the origin(terrestrial or marine) of CDOM on the apparent quantum yields(AQY) of CO were studied for seawater samples collected from Jiaozhou Bay. Our results demonstrat that photobleaching, temperature and the origin of CDOM strongly affected the efficiency of CO photoproduction. The concentration, absorbance and fluorescence of CDOM exponentially decreased with increasing light dose. Terrestrial riverine organic matter could be more prone to photodegradation than the marine algae-derived one. The relationships between CO AQY and the dissolved organic carbon-specific absorption coefficient at 254 nm for the photobleaching study were nonlinear, whereas those of the original samples were strongly linear. This suggests that: 1) terrestrial riverine CDOM was more efficient than marine algae-derived CDOM for CO photoproduction; 2) aromatic and olefinic moieties of the CDOM pool were affected more strongly by degradation processes than by aliphatic ones. Water temperature and the origin of CDOM strongly affected the efficiency of CO photoproduction. The photoproduction rate of CO in autumn was estimated to be 31.98 μmol m-2 d-1 and the total DOC photomineralization was equivalent to 3.25%- 6.35% of primary production in Jiaozhou Bay. | 2 | Theory and methods of global stability analysis for high arch dam显示文摘The global stability of high arch dam is one of the key problems in the safety study of arch dams,but no feasible method with theoretical basis is available.In this paper,based on the stability theory of mechanical system,it is demonstrated that the global failure of high arch dams belongs to a physical instability starting from local strength failure,which is the extreme point instability according to the characteristics of load-displacement curve obtained from the failure process of dam-foundation system. | 3 | Depression and stress levels increase risk of liver cancer through epigenetic downregulation of hypocretin显示文摘Recent studies suggest that Hypocretin (HCRT, Orexin) are involved in stress regulation of depression through the hypothalamic-pituitary-adrenal (HPA) axis. However, the molecular mechanism by which Hypocretin regulate neurobiological responses is unknown. Herein, the effects of chronic stress on the epigenetic modification of HCRT and its association with depression were explored with regard to a potential role in cancer progression. In the study, Sprague Dawley (SD) rats were used to establish an animal model of cancer with depression by administrating n-nitrosodiethylamine (DEN) and chronic unpredictable mild stress (CUMS). RNA-sequencing was used to detect differentially expressed genes in the hippocampus of rats and quantitative real-time polymerase chain reaction (qRT-PCR) was used to validate the results of RNA-sequencing. The status of HCRT promoter methylation was assessed by methylation specific polymerase chain reaction. Behavioral tests showed that rats exposed to CUMS had significant depressive-like behaviors. The number of liver tumors and tumor load in depressed rats exposed to CUMS was higher than in SD rats without CUMS. RNA-sequencing revealed that HCRT was one of the most siginificantly downregulated gene in the hippocampus of SD rats with CUMS compared to non-stressed group, which was validated by qRT-PCR. HCRT mRNA expression was downregulated and the promoter for HCRT was hyper-methylated in those with depression. These results identified a critical role for chronic psychological stressors in tumorigenesis and cancer progression, via epigenetic HCRT downregulation. | 4 | Temporal and spatial analysis of changes in snow cover in western Sichuan based on MODIS images显示文摘We developed a method for analyzing the change in snow cover using MODIS imagery.The method was applied to images of western Sichuan Province,China taken between 2002 and 2008.The model for extracting data on snow cover from MODIS images was created by spectral analysis.The multi-temporal snow layers were used to evaluate the temporal and spatial change in the area under snow cover between 2002 and 2008 using overlay and statistical analysis in ARCGIS.The majority(60.4%) of western Sichuan was rarely covered by snow and only 0.3% was covered by perennial snow in 2002.Snow cover was pri-marily distributed in Garzê and Aba.The area under snow cover was significantly and negatively correlated with the average monthly temperature and rainfall in 2002.The largest area under snow cover was measured in 2006 and the smallest was in 2007.Similarly,the area of snowmelt was the highest in 2006 and lowest in 2007.In general,the elevation of the snow line in-creased throughout the period 2002-2008;however,the elevation decreased in some years.Our results provide an important insight into the distribution of snow in this region,and may be useful for climate modeling and predicting the availability of water resources and the occurrence of floods and droughts. | YANG CunJian ZHAO ZiJian NI Jing REN XiaoLan WANG Qin | 2012 | Science China Earth Sciences2012,55,8: | 2 |

| 8 | Enrichment of health-promoting lutein and zeaxanthin in tomato fruit through metabolic engineering显示文摘Carotenoids constitute a large group of natural pigments widely distributed in nature.These compounds not only provide fruits and flowers with distinctive colors,but also have significant health benefits for humans.Lutein and zeaxanthin,both oxygen-containing carotenoids,are considered to play vital roles in promoting ocular development and maintaining eye health.However,humans and mammals cannot synthesize these carotenoid derivatives,which can only be taken from certain fruits or vegetables.Here,by introducing four endogenous synthetic genes,SlLCYE,SlLCYB,SlHYDB,and SlHYDE under fruit-specific promoters,we report the metabolic engineering of lutein/zeaxanthin biosynthesis in tomato fruit.Transgenic lines overexpression of one(SlLCYE),two(SlLCYE and SlLCYB;SlLCYB and SlHYDB),and all these four synthetic genes re-established the lutein/zeaxanthin biosynthetic pathways in the ripe tomato fruit and thus resulted in various types of carotenoid riched lines.Metabolic analyses of these engineered tomato fruits showed the strategy involved expression of SlLCYE tends to produceα-carotene and lutein,as well as a higher content of β-carotene and zeaxanthin was detected in lines overexpressing SlLCYB.In addition,the different combinations of engineered tomatoes with riched carotenoids showed higher antioxidant capacity and were associated with a significantly extended shelf life during postharvest storage.This work provides a successful example of accurate metabolic engineering in tomato fruit,suggesting the potential utility for synthetic biology to improve agronomic traits in crops.These biofortified tomato fruits could be also exploited as new research subjects for studying the health benefits of carotenoid derivatives. | Yanjun Wu Yong Yuan Wenqian Jiang Xin Zhang Siyan Ren Hsihua Wang Xiaolan Zhang Yang Zhang | 2022 | Synthetic and Systems Biotechnology2022,7,4: | 0 |

| 11 | Palladium nanoplates scotch breast cancer lung metastasis by constraining epithelial-mesenchymal transition显示文摘Metastasis accounts for the majority of cancer deaths in many tumor types including breast cancer.Epithelial-mesenchymal transition(EMT)is the driving force for the occurrence and progression of metastasis,however,no targeted strategies to block the EMT program are currently available to combat metastasis.Diverse engineered nanomaterials(ENMs)have been reported to exert promising anti-cancer effects,however,no ENMs have been designed to target EMT.Palladium(Pd)nanomaterials,a type of ENM,have received substantial attention in nanomedicine due to their favorable photothermal performance for cancer therapeutics.Herein,Pd nanoplates(PdPL)were found to be preferentially biodistributed to both primary tumors and metastatic tumors.Importantly,PdPL showed a significant inhibition of lung metastasis with and without near-infrared(NIR)irradiation.Mechanistic investigations revealed that EMT was significantly compromised in breast cancer cells upon the PdPL treatment,which was partially due to the inhibition of the transforming growth factor-beta(TGF-β)signaling.Strikingly,the PdPL was found to directly interact with TGF-βproteins to diminish TGF-βfunctions in activating its downstream signaling,as evidenced by the reduced phosphorylation of Smad2.Notably,TGF-β-independent pathways were also involved in undermining EMT and other important biological processes that are necessary for metastasis.Additionally,NIR irradiation elicited synergistic effects on PdPL-induced inhibition of primary tumors and metastasis.In summary,these results revealed that the PdPL remarkably curbed metastasis by inhibiting EMT signaling,thereby indicating the promising potential of PdPL as a therapeutic agent for treating breast cancer metastasis. | Shunhao Wang Jingchao Li Mei Chen Liting Ren Wenya Feng Lining Xu Xiaolan Chen Tian Xia Nanfeng Zheng Sijin Liu | 2021 | National Science Review2021,8,7: | 0 |
