1Release of endogenous hydrogen sulfide in enteric nerve cells suppresses intestinal motility during severe acute pancreatitis显示文摘Previous studies have shown that during severe acute pancreatitis(SAP)attacks,hydrogen sulfide(H2S)is released in the colon.However,the roles played byH2S in regulating enteric nerves remain unclear.In this study,we examined the association between SAP-induced H2S release and loss of intestinal motility,and also explored the relevant mechanism in enteric nerve cells.A rat SAP model was constructed and enteric nerve cells were prepared.Intestinal mobility was evaluated by measuring the number of bowel movements at indicated time points and by performing intestinal propulsion tests.The production of inflammatory cytokines during a SAP attack was quantified by ELISA,and the levels of cystathionine-y-lyase(CSE)and cystathionine-p-synthase(CBS)were examined by immunohistochemistry and western blot analysis.In vivo studies showed that PI3K/Akt/Sp1 signaling in enteric nerve cells was blocked,confirming the mechanism of endogenous H2S formation by western blot analysis and immunofluorescence.Our results also showed that rats with SAP symptoms had reduced intestinal motility.Furthermore,PI3K/Akt/Sp1 signaling was triggered and CSE expression was up-regulated,and these changes were associated with H2S formation in the colon.In addition,propargylglycine reduced the levels of inflammatory cytokines and suppressed the release of H2S.Enteric nerve cells that were incubated with LY294002 and transfected with a Sp7-knockdown vector displayed decreased levels of CSE production,which led to a decrease in H2S production.These results suggest that SAP symptoms suppressed the intestinal motility of rats via the release of H2S in enteric nerve cells,which was dependent on the inflammation-induced PI3K/Akt/Sp1 signaling pathway.Ribin Liao Liwei Xue Zhanrong Qiang Cheng Zhang Ying Liu 2020Acta Biochimica et Biophysica Sinica2020,52,1:2
2Study on Formula Optimization of Huangjing Zhima Pills显示文摘[Objectives]With Polygonati Rhizoma steamed and sundried for 9 times as the main raw material, a type of Huangjing Zhima Pills was developed and its optimal formula was studied. [Methods] The Huangjing Zhima Pills was developed with nine-steamed nine-sundried Polygonati Rhizoma as the main raw material by adding black sesame, black beans, and honey. Taking sensory score and texture properties as evaluation indexes, the optimal formula parameters of Huangjing Zhima Pills were determined by single factor experiments and an orthogonal test combined with a formula scoring method. [Results] The best formula of Huangjing Zhima Pills was addition amount of nine-steamed nine-sundried Polygonati Rhizoma 25%, addition amount of black bean powder 15%, addition amount of honey added 17%, and addition amount of black sesame powder 30%. The moisture content in the finished product was(13.367±0.056) g/100 g, and the total colony count, Escherichia coli, mold and yeast and other microbial indexes all met the standards. The Huangjing Zhima Pills prepared under these conditions had pleasant sesame flavor and Polygonati Rhizoma flavor, delicate and sweet tasted, and a slight aftertaste of sweetness. Meanwhile, Polygonatum, black sesame and black beans are all medicinal and food homologous plants with certain health care effects, and the Huangjing Zhima Pills prepared from them are healthy snacks that people can carry and eat easily.
