2Development of Plant Prime-Editing Systems for Precise Genome Editing显示文摘Prime-editing systems have the capability to perform efficient and precise genome editing in human cells.In this study,we first developed a plant prime editor 2(pPE2)system and test its activity by generating a targeted mutation on an HPT^(-ATG) reporter in rice.Our results showed that the pPE2 system could induce programmable editing at different genome sites.In transgenic T0 plants,pPE2-generated mutants occurred with 0%–31.3%frequency,suggesting that the efficiency of pPE2 varied greatly at different genomic sites and with prime-editing guide RNAs of diverse structures.To optimize editing efficiency,guide RNAs were introduced into the pPE2 system following the PE3 and PE3b strategy in human cells.However,at the genomic sites tested in this study,pPE3 systems generated only comparable or even lower editing frequencies.Furthemore,we developed a surrogate pPE2 system by incorporating the HPT^(-ATG) reporter to enrich the prime-edited cells.The nucleotide editing was easily detected in the resistant calli transformed with the surrogate pPE2 system,presumably due to the enhanced screening efficiency of edited cells.Taken together,our results indicate that plant prime-editing systems we developed could provide versatile and flexible editing in rice genome.Rongfang Xu Juan Li Xiaoshuang Liu Tiaofeng Shan Ruiying Qin Pengcheng Wei 2020Plant Communications2020,1,3:10

5Biodegradation of geosmin in drinking water by novel bacteria isolated from biologically active carbon显示文摘Three strains of Gram-negative bacteria capable of removing geosmin from drinking water were isolated from biologically active carbon and identified to be Chryseobacterium sp., Sinorhizobium sp. and Stenotrophomonas sp. based on physio-biochemistry analysis and 16S rRNA gene sequence analysis. Removal efficiencies of 2 mg/L geosmin in mineral salts medium were 84.0%, 80.2% and 74.4% for Chryseobacterium sp., Sinorhizobium sp. and Stenotrophomonas sp., respectively, while removal efficiencies of 560 ng/L geosmin in filter influent were 84.8%, 82.3% and 82.5%, respectively. The biodegradation of geosmin was determined to be a pseudo first-order reaction, with rate constants at 2 mg/L and 560 ng/L being 0.097 and 0.086 day 1 , 0.089 and 0.084 day 1 , 0.074 and 0.098 day 1 for the above mentioned degraders, respectively. 6Direct reduction of hematite powders in a fluidized bed reactor显示文摘Ultrafine hematite powder was reduced to produce ultrafine iron powder in a 50%Ar-50%H_2 atmosphere at 450-550℃ in a fluidized bed reactor.The ultrafine hematite powder shows the typical agglomerating fluidization behavior with large agglomerates fluidized at the bottom of the bed and small agglomerates fluidized at the upper part of the bed.It was found that defluidization occurred even at the low temperature of 450℃ with low metallization rate.Defluidization was attributed mainly to the sintering of the newly formed iron particles.Granuation was employed to improve the fluidization quality and to tackle the defluidization problem,where granules fluidized like a Geldart's group A powder.Granulation was found to effectively reduce defluidization during reduction,without however sacrificing reduction speed.The asreduced iron powders from both the ultrafine and the granulated hematite exhibited excellent sintering activity,that is,fast sintering at temperature of as low as^580σ_(RR)~2),which is much superior as compared to that of nano/ultrafine iron powders made by other processes.Qingshan Zhu Rongfang Wua Hongzhong Li 2013Particuology2013,11,3:8

9Genome editing mediated by SpCas9 variants with broad non-canonical PAM compatibility in plants显示文摘Streptococcus pyogenes Cas9(SpCas9)is the most widely used genome editing tool in plants.The editing induced by SpCas9 strictly requires a canonical NGG protospacer-adjacent motif(PAM),significantly limiting its scope of application.Recently,five SpCas9 variants,SpCas9-NRRH,SpCas9-NRCH,SpCas9-NRTH,SpG,and SPRY,were developed to recognize non-canonical PAMs in human cells.In this study,these variants were engineered for plant genome editing,and their targeted mutagenesis capabilities were comprehensively examined at various canonical and non-canonical PAM sites in rice(Oryza sativa)by stable transformation.Moreover,both cytosine base editors using a rat APOBEC1 or a human APO-BEC3a and adenine base editors using a directly evolved highly compatible TadA*-8e deaminase were developed from these SpCas9 variants.Our results demonstrated that the developed SpCas9 variantsbased base editors readily generated conversions between C.G and T.A in the target sites with noncanonical PAMs in transgenic rice lines.Collectively,the toolbox developed in this study substantially expands the scope of SpCas9-mediated genome editing and will greatly facilitate gene disruption and precise editing in plants.Juan Li Rongfang Xu Ruiying Qin Xiaoshuang Liu Fanna Kong Pengcheng Wei 2021Molecular Plant2021,14,2:6
10Engineered porous Ni2P-nanoparticle/Ni2P-nanosheet arrays via the Kirkendall effect and Ostwald ripening towards efficient overall water splitting显示文摘Transitional metal phosphides with array-like structure grown on conductive support materials are promising bifunctional catalysts for the oxygen evolution reaction(OER)and hydrogen evolution reaction(HER).In this study,a method was developed to synthesize directly porous Ni2P nanosheet arrays and Ni2P nanoparticles onto nickel foam via a hydrothermal reaction followed by a phosphorization process.Mechanistic studies revealed that the allomorphs of Ni2P nanosheets and Ni2P nanoparticles in the array-like structure were formed via the Kirkendall effect and Ostwald ripening.A fully functional water electrolyzer containing Ni2P as electrodes for the OER and HER exhibited promising activity and stability.At 10 mA·cm^−2,a Ni2P cell voltage of 1.63 V was obtained,which was only 0.05 V smaller than that found for Pt/C/NF||RuO2/NF cell.The enhanced electrocatalytic performance resulted from the favorable porosity of the Ni2P arrays and the synergistic effect between Ni2P nanosheets and Ni2P nanoparticles.Yutai Wu Hui Wang Shan Ji Bruno G.Pollet Xuyun Wang Rongfang Wang 2020Nano Research2020,13,8:6

12Properties of Reactive Magnetron Sputtered ITO Films without in-situ Substrate Heating and Post-deposition Annealing显示文摘Indium tin oxide (ITO) films were prepared on polyester, Si and glass substrate with relatively high deposition rate of above 0.9 nm/s by DC reactive magnetron sputtering technique at the sputtering pressure of 0.06 Pa system, respectively. The dependence of resistivity on deposition parameters, such as deposition rate, target-to-substrate distance (TSD), oxygen flow rate and sputtering time (thickness), has been investigated, together with the structural and the optical properties. It was revealed that all ITO films exhibited lattice expansion. The resistivity of ITO thin films shows significant substrate effect: much lower resistivity and broader process window have been reproducibly achieved for the deposition of ITO films onto polyester rather than those prepared on both Si and glass substrates. 14Proteomics study of Mycoplasma pneumoniae pneumonia reveals the Fc fragment of the IgG-binding protein as a serum biomarker and implicates potential therapeutic targets显示文摘Macrolide and corticosteroid resistance has been reported in patients with Mycoplasma pneumoniae(MP)pneumonia(MPP).MP clearance is difficult to achieve through antibiotic treatment in sensitive patients with severe MPP(SMPP).SMPP in children might progress to airway remodeling and even bronchiolitis/bronchitis obliterans.Therefore,identifying serum biomarkers that indicate MPP progression and exploring new targeted drugs for SMPP treatment require urgency.In this study,serum samples were collected from patients with general MPP(GMPP)and SMPP to conduct proteomics profiling.The Fc fragment of the IgG-binding protein(FCGBP)was identified as the most promising indicator of SMPP.Biological enrichment analysis indicated uncontrolled inflammation in SMPP.ELISA results proved that the FCGBP level in patients with SMPP was substantially higher than that in patients with GMPP.Furthermore,the FCGBP levels showed a decreasing trend in patients with GMPP but the opposite trend in patients with SMPP during disease progression.Connectivity map analyses identified 25 possible targeted drugs for SMPP treatment.Among them,a mechanistic target of rapamycin kinase(mTOR)inhibitor,which is a macrolide compound and a cell proliferation inhibitor,was the most promising candidate for targeting SMPP.To our knowledge,this study was the first proteomics-based characterization of patients with SMPP and GMPP.Jinrong Liu Rongfang Shen Lin Feng Shujun Cheng Jun Chen Ting Xiao Shunying Zhao 2022Frontiers of Medicine2022,16,3:3
15Cgr1,a ripe rot resistance QTL in Vitis amurensis‘Shuang Hong’grapevine显示文摘Ripe rot is a serious grapevine disease in Vitis L.and Muscadinia(Planch.)Small.However,resistance to this disease has been reported in some oriental Vitis species.To identify resistance-related Quantitative Trait Loci(QTLs)from the Chinese grape species V.amurensis,an F1 population of V.vinifera‘Cabernet Sauvignon’×V.amurensis‘Shuang Hong’was used to map the ripe rot resistance loci expected in‘Shuang Hong’grape.A total of 7598 single nucleotide polymorphisms(SNPs)between the parents were identified in our previous study,and 934 SNPs were selected for genetic map construction.These SNPs are distributed across the 19 chromosomes covering a total of 1665.31 cM in length,with an average of 1.81 cM between markers.Ripe rot resistance phenotypes among the hybrids were evaluated in vitro using excised leaves for three consecutive years from 2016 to 2018;a continuous variation was found among the F1 hybrids,and the Pearson correlation coefficients of the phenotypes scored in all three years were significant at the 0.01 level.Notably,the first QTL reported for resistance to grape ripe rot disease,named Cgr1,was identified on chromosome 14 of‘Shuang Hong’grapevine.Cgr1 could explain up to 19.90%of the phenotypic variance.In addition,a SNP named‘np19345’was identified as a molecular marker closely linked to the peak of Cgr1 and has the potential to be developed as a marker for the Cgr1 resistance haplotype.Peining Fu Quanyou Tian Gongti Lai Rongfang Li Shiren Song Jiang Lu 2019Horticulture Research2019,6,1:3
16A Low-Complexity Resource Allocation Scheme for OFDMA Multicast Systems with Proportional Fairness显示文摘A low-complexity optimization scheme is proposed to balance the tradeoff between system capacity and proportional fairness in orthogonal frequency division multiple access(OFDMA) based multicast systems. The major challenge is to solve the non-convexity optimization problem with strict proportional fairness. Constrained team progress algorithm(CTPA) solves this non-convexity problem by allocating sub-channels to each group based on sub-channel gains and proportional fairness constraint. Mapping power algorithm(MPA) guarantees strict proportional fairness with efficient power allocation which utilizes the mapping relation between power and throughput. CTPA-MPA is analyzed in three aspects: complexity, fairness and efficiency. We numerically show that when the system capacity is slightly increased in lower power region compared with several previous approaches, CTPA-MPA improves the proportional fairness in a typical scenario with 4 groups over 16 sub-channels, while reducing the complexity from exponential to linear in the number of sub-channels. 17Integrating ecosystem services evaluation and landscape pattern analysis into urban planning based on scenario prediction and regression model显示文摘Urban planning has become a widely concern for minimizing the negative effects of urban expansion on terrestrial ecosystems. We developed an interdisciplinary modeling framework to evaluate the effectiveness and shortcomings of urban expansion management strategies. A three-step method was applied to Yinchuan Plain in the northwestern of China, including(1)analyzing the relationship between landscape pattern and ecosystem service values through mathematical statistics;(2) predicting landscape pattern and ecosystem services change under different scenarios based on cellular automaton model(SLEUTH-3r model); and(3) designing and validating optimized scenario through integrating historical analysis experiments and future multi-comparison suggestions. Results have suggested that landscape composition and configuration can significantly affect regional ecosystem service values, especially the connectivity and shape of landscape. Compact urban growth policy and medium environment protection policy are the appropriate setting for urban expansion plan. Optimization validation of the combined designed scenario implied the reliability of this method. 18Effects of 2, 4-Dichlorophenoxy Acetic Acid andLight on Growth of Gerbera (Gerbera jamesoniicv. Daxueju) Callus显示文摘This study was conducted to determine effects of 2,4-dichlorophenoxy acetic acid(2,4-D) and light on growth of gerbera(Gerbera jamesonii cv. Daxueju) callus. Callus was induced from both petiole and leaf explants of gerbera on Murashige and Skoog medium supplemented with 3% sucrose and various concentrations of 2,4-D and placed under light and dark. Callus induction percentage, callus size and callus fresh and dry weights were efficiently higher when using petiole as explant. MS medium supplemented with 1.5 mg/L 2,4-D showed the highest callus induction percentage of 96.70%. Callus induced under light had larger weight mass. 20AIMP1 promotes multiple myeloma malignancy through interacting with ANP32A to mediate histone H3 acetylation显示文摘Background:Multiple myeloma(MM)is the second most common hematological malignancy.An overwhelming majority of patients with MM progress to serious osteolytic bone disease.Aminoacyl-tRNA synthetase-interacting multifunctional protein 1(AIMP1)participates in several steps during cancer development and osteoclast differentiation.This study aimed to explore its role in MM.Methods:The gene expression profiling cohorts of MM were applied to determine the expression of AIMP1 and its association with MM patient prognosis.Enzyme-linked immunosorbent assay,immunohistochemistry,and Western blotting were used to detect AIMP1 expression.Protein chip analysis,RNA-sequencing,and chromatin immunoprecipitation and next-generation sequencing were employed to screen the interacting proteins and key downstream targets of AIMP1.The impact of AIMP1 on cellular proliferation was determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay in vitro and a xenograft model in vivo.Bone lesions were evaluated using tartrate-resistant acid phosphatase staining in vitro.A NOD/SCID-TIBIA mouse model was used to evaluate the effect of siAIMP1-loaded exosomes on bone lesion formation in vivo.Results:AIMP1 expression was increased in MM patients and strongly associated with unfavorable outcomes.Increased AIMP1 expression promoted MM cell proliferation in vitro and in vivo via activation of the mitogen-activated protein kinase(MAPK)signaling pathway.Protein chip assays and subsequent experiments revealed that AIMP1 interacted with acidic leucine-rich nuclear phosphoprotein 32 family member A(ANP32A)to regulate histone H3 acetylation.In addition,AIMP1 increased histone H3 acetylation enrichment function of GRB2-associated and regulator of MAPK protein 2(GAREM2)to increase the phosphorylation of extracellular-regulated kinase 1/2(p-ERK1/2).Furthermore,AIMP1 promoted osteoclast differentiation by activating nuclear factor of activated T cells c1(NFATc1)in vitro.In contrast,exosome-coated small interfering RNA of AIMP1 effectively suppressed MM progression and osteoclast differentiation in vitro and in vivo.Conclusions:Our data demonstrate that AIMP1 is a novel regulator of histone H3 acetylation interacting with ANP32A in MM,which accelerates MM malignancy via activation of the MAPK signaling pathway.Rongfang Wei Yan Zhu Yuanjiao Zhang Wene Zhao Xichao Yu Ling Wang Chunyan Gu Xiaosong Gu Ye Yang 2022Cancer Communications2022,42,11:2
