| 1 | X线照射后肺癌细胞肿瘤坏死因子mRNA的表达显示文摘目的探讨X线诱导和调节肺癌细胞株NCI-H596和A549肿瘤坏死因子(TNF-α)mRNA的表达作用及其临床意义。方法应用实时荧光定量RT-PCR技术,定量检测NCI-H596和A549细胞株照射前和接受不同剂量(2,5,10,20,30和40Gy)的X线照射后,以及受照30Gy后不同时间TNF-αmRNA的表达。同时进行集落形成试验,检测NCI-H596和A549细胞株的放射敏感性。结果集落形成试验结果显示,未照射的NCI-H596细胞株集落形成率为0.12~0.24;A549细胞株集落形成率为0.37~0.45。照射后,A549细胞的存活分数(SF)在2Gy时为47.3%±9.0%,4Gy时为18.0%±3.0%,6Gy时为6.0%±2.0%,8Gy时为1.4%±0.3%;NCI-H596细胞的SF在2Gy时为55.2%±51.0%,4Gy时为15.9%±9.2%,6Gy时为3.5%±1.7%;8Gy时为0.9%±0.6%;二者SF差异无统计学意义,二者的D0值和Dq值差异亦无统计学意义(P>0.05)。实时荧光定量RT-PCR检测显示,NCI2H596细胞受照后TNF-αmRNA表达逐渐升高,照射剂量达40Gy时,TNF-αmRNA表达水平达高峰,为对照组的83倍;受照后,1hTNF-αmRNA表达逐渐升高,6h达高峰,为对照组的568倍。A549细胞受照后,1hTNF-αmRNA表达即达高峰,为对照组的136倍。结论X线可诱导肺癌细胞株A549和NCI2H596产生TNF-α,且呈剂量、时间依赖性,可提高肺癌放射治疗的疗效,也可对正常组织和? | 刘莉 陆海 CE Ruebe CH Ruebe | 2005 | 中华肿瘤杂志2005,27,6: | 7 |
| 2 | TGF-β1转基因小鼠肺组织中基质金属蛋白酶和基质金属蛋白酶组织抑制物的表达显示文摘目的探讨肺组织内高水平的转化生长因子-β1(TGF-β1)蛋白对基质金属蛋白酶(MMPs)和基质金属蛋白酶组织抑制物(TIMPs)表达的调节,以及MMPs/TIMPs表达紊乱与肺组织纤维化的关系。方法8只TGF-β1转基因小鼠和8只正常对照小鼠在出生后8周经断头术处死,收集血浆及肺组织标本,测定血浆和肺组织中TGF-β1蛋白浓度,以及MMP-2、MMP-9、MMP-3、MMP-13、TIMP-1、TIMP-2和TIMP-3蛋白的表达,并通过Masson-Goldner染色法检测肺组织中胶原的沉积。结果在转基因小鼠肺组织中,MMP-2的表达显著降低,TIMP-1、TIMP-2和TIMP-3的表达显著增加,并且转基因小鼠肺组织呈现明显纤维化。结论转基因小鼠肺组织中高浓度的TGF-β1可能通过抑制MMP-2表达,以及增加TIMP-1、TIMP-2、TIMP-3表达,减少细胞外基质的降解,增加细胞外基质的沉积,促进肺纤维化的产生。 | 杨坤禹 胡豫 刘莉 张涛 伍钢 Claudia Ruebe Christian Ruebe | 2006 | 华中科技大学学报(医学版)2006,35,6: | 5 |
| 3 | 肺部X射线照射后炎性细胞因子在支气管上皮细胞中的表达显示文摘目的研究C57BL/6J小鼠双肺照射12Gy后,炎性细胞因子肿瘤坏死因子-α(TNF-α)、白介素-1α(IL-1α)和白介素-6(IL-6)蛋白在肺组织中的表达。方法实验组小鼠给予双肺单次照射12Gy,对照组小鼠在相同条件下双肺假照射0Gy。分别于照射后0·5、1、3、6、12、24、48和72h,1、2、4、8、16和24周处死,通过免疫组织化学方法和图像分析定量检测肺组织中TNF-α、IL-1α和IL-6蛋白的表达。结果胸部照射后数小时,上述炎性因子蛋白表达即比对照组显著增加。在随后的急性放射性肺炎阶段,支气管上皮以及肺间质的炎性细胞产生了大量的TNF-α(最大值出现在全肺照射后第4周,9·74%±1·78%)、IL-1α(最大值出现在全肺照射后第8周,14·76%±7·77%)和IL-6(最大值出现在全肺照射后第8周,4·28%±1·33%)。结论肺部照射以后,在胸部照射的最初数小时以内,支气管上皮是上述炎性因子最重要的来源,这些炎性细胞因子进一步通过征集和激活炎症细胞来促进和放大放射性肺炎的产生和发展。 | 杨坤禹 胡豫 Ruebe Claudia Ruebe Christian | 2006 | 中华放射医学与防护杂志2006,26,5: | 4 |

| 13 | TGF-beta1 Transgenic Mouse Model of Thoracic Irradiation: Modulation of MMP-2 and MMP-9 in the Lung Tissue显示文摘To investigate the effects of TGF-β1 on the two gelatinases (MMP-2 and MMP-9), and their roles in lung remodeling after irradiation-induced lung injury. Expressions of TGF-β1 were measured with western blot, and expressions of MMP-2 and MMP-9 were analyzed with zymography in a TGF-β1 transgenic mouse model after thoracic irradiation with 12 Gy. We found expressions of TGF-β1 in the lung from the transgenic mice were three folds as compared to those from control mice. With densitometrical analysis, we found a significant decrease in MMP-9 activity in lung homogenates from the transgenic mice as compared with those from non-transgenic control mice 8 weeks after sham-irradiation (relative MMP-9 activity: C: 1.000±0.1091; TG: 0.4772±0.470 (n=8, P<0.05). But MMP-2 was constitutively expressed in the lung homogenates from the transgenic mice as compared to those from control mice 8 weeks after sham-irradiation (relative MMP-2 activity 8 weeks after sham-irradiation: C: 1.000±0.1556, TG: 1.0075±0.1472). Eight weeks after thoracic irradiation with 12 Gy, we observed a significant increase of MMP-2 and MMP-9 activity in lung homogenates from both transgenic and normal mice. In TGF-β1 transgenic mice relative MMP-9 activity was increased to 1.5321±0.2217 folds 8 weeks after thoracic irradiation with 12 Gy as compared to those after sham-irradiation (1.000±0.2153), and relative MMP-2 activity was increased to 1.7142±0.4231 folds. Our results show that TGF-β1 itself down-regulates activity of MMP-9, thereby decreases ECM degradation in lungs of TGF-β1 transgenic mice. Also we find that ionizing irradiation upregulates both MMP-2 and MMP-9 activity.
