| 1 | Neuroprotective effect of Longshengzhi capsule following permanent middle cerebral artery occlusion in rats显示文摘Objective:Longshengzhi capsule(LSZC)is an optimized preparation based on the traditional Chinese Medicine formula Buyanghuanwu Decoction(BYHWD),and is approved by the China Food and Drug Administration for treating stroke-induced disability and vascular diseases.Herein,we examined the pharmacodynamics,anti-apoptotic and anti-oxidant actions,and potential mechanisms of action of LSZC following stroke in rats.Methods:Permanent middle cerebral artery occlusion(MCAO)was used as an ischemic stroke model.LSZC was administered intragastrically.We examined the survival rate,bodyweight,and neurological deficits in stroke rats.Brain infarct size was determined by 2,3,5-triphenyltetrazolium chloride staining.Brain pathology was examined using hematoxylin and eosin staining,Nissl staining,and TdT-mediated dUTP Nick-End Labeling staining.Malondialdehyde,catalase,superoxide dismutase,and glutathione levels were examined by commercial kits.Expression of Nrf2,heme oxygenase-1,Bax,Bcl-2,cleaved caspase-3,and caspase-3 proteins in brain tissue was measured by Western blot.Results:LSZC markedly improved the survival rate and bodyweight,and reduced infarct volume and neurological deficit scores,in MCAO stroke rats.LSZC also significantly attenuated oxidative stress,as indicated by decreased expression of malondialdehyde,and upregulation of Nrf2,heme oxygenase-1,catalase,superoxide dismutase,and glutathione.Moreover,LSZC significantly decreased apoptosis,including a decrease in Bax and cleaved caspase-3 expression,and an increase in Bcl-2,as well as a reduction in numbers of apoptotic neurons.Conclusion:LSZC treatment is neuroprotective against ischemic stroke,potentially via reducing oxidative stress and apoptosis.The Nrf2 and apoptotic signaling pathways may play important roles in the antioxidant and anti-apoptotic actions of LSZC. | Xiaoli Sun Shuzhen Guo Yong Wang Bangze Fu Junjie Liu Yili Zhang Ruocong Yang Chun Li Jian Gao Li Gao Wei Wang | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 4 |
| 2 | Network pharmacology and in vitro study of Fengreqing oral liquid(风热清口服液) in the intervention of wind-heat pattern显示文摘OBJECTIVE:To investigate the underlying mechanism of the effect of Fengreqing oral liquid(风热清口服液,FOL)on wind-heat pattern(WHP).METHODS:In this study,we predicted the potential targets of FOL via the approach of network pharmacology and verified it by in vitro inflammation model.In the network pharmacology part,two strategies,namely the direct target search and the indirect one,were used to collect the target sets of FOL in WHP treatment.The enrichment analysis was carried out by David database and Clue Go plug-in in Cytoscape.Furthermore,the potential targets were mapped in the candidate pathways.In the verification experiment section,in vitro model of lipopolysaccharide(LPS)induced RAW 264.7 was used to confirm the predictive results in the network pharmacology part.RESULTS:Through the two screening strategies,a total of 141 non-repetitive intervention targets of FOL on WHP were obtained.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis showed that the intervention effect was mainly focused on the anti-inflammatory effect,and the Toll-like receptor signaling pathway was one of the most critical regulatory pathways.Further mapping analysis showed that phosphatidylinositol 3-kinase(PI3 K)-protein kinase B(AKT)signaling transfer might be the key part of regulating the concentration of inflammation mediators of FOL in the Toll-like receptor signaling pathway.In vitro experiment showed that FOL significantly reduced the levels of NO,IL-1,IL-6,and TNF-αproduced by RAW264.7 induced by LPS.Further immunofluorescence found that this effect is related to the regulation of PI3 K-AKT pathway activity by FOL.CONCLUSION:FOL can intervene in WHP by regulating the content of inflammatory mediators via the PI3 K-AKT pathway. | YANG Ruocong RAO Zhili LI Xiangyu WANG Dejian PENG Xi ZENG Nan | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 1 |
| 3 | Pathophysiological changes of muscle after ischemic stroke:a secondary consequence of stroke injury显示文摘Sufficient clinical evidence suggests that the damage caused by ischemic stroke to the body occurs not only in the acute phase but also during the recovery period,and that the latter has a greater impact on the long-term prognosis of the patient.However,current stroke studies have typically focused only on lesions in the central nervous system,ignoring secondary damage caused by this disease.Such a phenomenon arises from the slow progress of pathophysiological studies examining the central nervous system.Further,the appropriate therapeutic time window and benefits of thrombolytic therapy are still controversial,leading scholars to explore more pragmatic intervention strategies.As treatment measures targeting limb symptoms can greatly improve a patient’s quality of life,they have become a critical intervention strategy.As the most vital component of the limbs,skeletal muscles have become potential points of concern.Despite this,to the best of our knowledge,there are no comprehensive reviews of pathophysiological changes and potential treatments for post-stroke skeletal muscle.The current review seeks to fill a gap in the current understanding of the pathological processes and mechanisms of muscle wasting atrophy,inflammation,neuroregeneration,mitochondrial changes,and nutritional dysregulation in stroke survivors.In addition,the challenges,as well as the optional solutions for individualized rehabilitation programs for stroke patients based on motor function are discussed. | Hu Qi Dan Tian Fei Luan Ruocong Yang Nan Zeng | 2024 | Neural Regeneration Research2024,19,4: | 0 |
