1Expression of miR-15/107 Family MicroRNAs in Human Tissues and Cultured Rat Brain Cells显示文摘The miR-15/107 family comprises a group of 10 paralogous microRNAs(miRNAs),sharing a 50AGCAGC sequence.These miRNAs have overlapping targets.In order to characterize the expression of miR-15/107 family miRNAs,we employed customized TaqMan Low-Density micro-fluid PCR-array to investigate the expression of miR-15/107 family members,and other selected miRNAs,in 11 human tissues obtained at autopsy including the cerebral cortex,frontal cortex,primary visual cortex,thalamus,heart,lung,liver,kidney,spleen,stomach and skeletal muscle.miR-103,miR-195 and miR-497 were expressed at similar levels across various tissues,whereas miR-107 is enriched in brain samples.We also examined the expression patterns of evolutionarily conserved miR-15/107 miRNAs in three distinct primary rat brain cell preparations(enriched for cortical neurons,astrocytes and microglia,respectively).In primary cultures of rat brain cells,several members of the miR-15/107 family are enriched in neurons compared to other cell types in the central nervous system(CNS).In addition to mature miRNAs,we also examined the expression of precursors(pri-miRNAs).Our data suggested a generally poor correlation between the expression of mature miRNAs and their precursors.In summary,we provide a detailed study of the tissue and cell type-specific expression profile of this highly expressed and phylogenetically conserved family of miRNA genes.Wang-Xia Wang Robert J.Danaher Craig S.Miller Joseph R.Berger Vega G.Nubia Bernard S.Wilfred Janna H.Neltner Christopher M.Norris Peter T.Nelson 2014Genomics, Proteomics & Bioinformatics2014,12,1:9

3Molecularly specific detection of bacterial lipoteichoic acid for diagnosis of prosthetic joint infection of the bone显示文摘Discriminating sterile inflammation from infection, especially in cases of aseptic loosening versus an actual prosthetic joint infection, is challenging and has significant treatment implications. Our goal was to evaluate a novel human monoclonal antibody(mAb) probe directed against the Gram-positive bacterial surface molecule lipoteichoic acid(LTA). Specificity and affinity were assessed in vitro. We then radiolabeled the anti-LTA mAb and evaluated its effectiveness as a diagnostic imaging tool for detecting infection via immuno PET imaging in an in vivo mouse model of prosthetic joint infection(PJI). In vitro and ex vivo binding of the anti-LTA mAb to pathogenic bacteria was measured with Octet, ELISA, and flow cytometry. The in vivo PJI mouse model was assessed using traditional imaging modalities, including positron emission tomography(PET) with [^(18)F]FDG and [^(18)F]Na F as well as X-ray computed tomography(CT), before being evaluated with the zirconium-89-labeled antibody specific for LTA([^(89)Zr]SAC55).The anti-LTA mAb exhibited specific binding in vitro to LTA-expressing bacteria. Results from imaging showed that our model could reliably simulate infection at the surgical site by bioluminescent imaging, conventional PET tracer imaging, and bone morphological changes by CT. One day following injection of both the radiolabeled anti-LTA and isotype control antibodies, the anti-LTA antibody demonstrated significantly greater(P < 0.05) uptake at S. aureus-infected prosthesis sites over either the same antibody at sterile prosthesis sites or of control non-specific antibody at infected prosthesis sites. Taken together, the radiolabeled anti-LTA mAb, [^(89)Zr]SAC55, may serve as a valuable diagnostic molecular imaging probe to help distinguish between sterile inflammation and infection in the setting of PJI. 4Geodynamics of oceanic plateau and plume head accretion and their role in Phanerozoic orogenic systems of China显示文摘We present three 3D numerical models of deep subduction where buoyant material from an oceanic plateau and a plume interact with the overriding plate to assess the influence on subduction dynamics,trench geometry,and mechanisms for plateau accretion and continental growth.Transient instabilities of the convergent margin are produced,resulting in:contorted trench geometry;trench migration parallel with the plate margin;folding of the subducting slab and orocline development at the convergent margin;and transfer of the plateau to the overriding plate.The presence of plume material beneath the oceanic plateau causes flat subduction above the plume,resulting in a 'bowed' shaped subducting slab.In plateau-only models,plateau accretion at the edge of the overriding plate results in trench migration around the edge of the plateau before subduction is re-established directly behind the trailing edge of the plateau.The plateau shortens and some plateau material subducts.The presence of buoyant plume material beneath the oceanic plateau has a profound influence on the behaviour of the convergent margin.In the plateau + plume model,plateau accretion causes rapid trench advance.Plate convergence is accommodated by shearing at the base of the plateau and shortening in the overriding plate.The trench migrates around the edge of the plateau and subduction is re-established well behind the trailing edge of the plateau,effectively embedding the plateau into the overriding plate.A slab window forms beneath the accreted plateau and plume material is transferred from the subducting plate to the overriding plate through the window.In all of the models,the subduction zone maintains a relatively stable configuration away from the buoyancy anomalies within the downgoing plate.The models provide a dynamic context for plateau and plume accretion in Phanerozoic accretionary orogenic systems such as the East China Orogen and the Central Asian Orogen(Altiads),which are characterised by accreted ophiolite complexes with diverse geochemical affinities,and a protracted evolution of accretion of exotic terranes including oceanic plateau and terranes with plume origins.Peter G.Betts Louis Moresi Meghan S.Miller David Willis 2015Geoscience Frontiers2015,6,1:2

10Mortality of Amur tigers:The more things change,the more they stay the same显示文摘Poaching as well as loss of habitat and prey are identified as causes of tiger population declines.Although some studies have examined habitat requirements and prey availability,few studies have quantified cause-specific mortality of tigers.We used cumulative incidence functions(CIFs)to quantify cause-specific mortality rates of tigers,expanding and refining earlier studies to assess the potential impact of a newly emerging disease.To quantify changes in tiger mortality over time,we re-examined data first collected by Goodrich et al.(2008;study period 1:1992–2004)as well as new telemetry data collected since January 2005(study period 2:2005–2012)using a total of 57 tigers(27 males and 30 females)monitored for an average of 747 days(range 26–4718 days).Across the entire study period(1992 to 2012)we found an estimated average annual survival rate of 0.75 for all tigers combined.Poaching was the primary cause of mortality during both study periods,followed by suspected poaching,distemper and natural/unknown causes.Since 2005,poaching mortality has remained relatively constant and,if combined with suspected poaching,may account for a loss of 17–19%of the population each year.Canine distemper virus(CDV)may be an additive form of mortality to the population,currently accounting for an additional 5%.Despite this relatively new source of mortality,poaching remains the main threat to Amur tiger survival and,therefore,population growth.Hugh S.ROBINSON John M.GOODRICH Dale G.MIQUELLE Clayton S.MILLER Ivan V.SERYODKIN 2015Integrative Zoology2015,10,4:1

14Review of research methodologies for tigers:Telemetry显示文摘Over the past half century,wildlife research has relied on technological advances to gain additional insight into the secretive lives of animals.This revolution started in the 1960s with the development of radio telemetry and continues today with the use of Global Positioning System(GPS)-based research techniques.In the present paper we review the history of radio telemetry from its origins with grizzly bears in Yellowstone to its early applications in tiger research and conservation in Asia.We address the different types of data that are available using radio telemetry as opposed to using other research techniques,such as behavioral observations,camera trapping,DNA analysis and scat analysis.In the late 1990s,the rapid development of GPS collar technology revolutionized wildlife research.This new technology has enabled researchers to dramatically improve their ability to gather data on animal movements and ecology.Despite the ecological and conservation benefits of radio telemetry,there have been few telemetry studies of tigers in the wild,and most have been on the Bengal or Amur subspecies.We close with an assessment of the current tiger conservation efforts using GPS technology and discuss how this new information can help to preserve tigers for future generations.Clayton S.MILLER Mark HEBBLEWHITE John M.GOODRICH Dale G.MIQUELLE 2010Integrative Zoology2010,5,4:0
