| 3 | Diffuse intestinal ganglioneuromatosis an uncommon manifestation of Cowden syndrome显示文摘Diffuse intestinal ganglioneuromatosis is a hamartomatous polyposis characterized by a disseminated, intramural or transmural proliferation of neural elements involving the enteric plexuses. It has been associated with MEN Ⅱ, neurofibromatosis type 1 and hamartomatous polyposis associated with phosphatase and tensin homolog mutation. We report the case of a female patient with a history of a breast and endometrial tumor who presented in a colonoscopy performed for rectal bleeding diffuse ganglioneuromatosis, which oriented the search for other characteristic findings of Cowden syndrome given the personal history of the patient. The presence of an esophagogastric polyposis was also noted. Cowden syndrome is characterized by skin lesions, but it is rarely diagnosed by these lesions, because they are usually overlooked. Intestinal polyposis is not a major diagnostic criterion but it is very useful for early diagnosis. | 4 | Magnetic resonance imaging for prostate cancer before radical and salvage radiotherapy: What radiation oncologists need to know显示文摘External beam radiotherapy(EBRT) is one of the principal curative treatments for patients with prostate cancer(PCa). Risk group classification is based on prostate-specific antigen(PSA) level, Gleason score, and T-stage. After risk group determination, the treatment volume and dose are defined and androgen deprivation therapy is prescribed, if appropriate. Traditionally, imaging has played only a minor role in T-staging due to the low diagnostic accuracy of conventional imaging strategies such as transrectal ultrasound, computed tomography, and morphologic magnetic resonance imaging(MRI). As a result, a notable percentage of tumours are understaged, leading to inappropriate and imprecise EBRT. The development of multiparametric MRI(mp MRI), an imaging technique that combines morphologic studies with functional diffusion-weighted sequences and dynamic contrastenhanced imaging, has revolutionized the diagnosis and management of PCa. As a result, mpM RI is now used in staging PCa prior to EBRT, with possible implications for both risk group classification and treatment decisionmaking for EBRT. mpM RI is also being used in salvageradiotherapy(SRT), the treatment of choice for patients who develop biochemical recurrence after radical prostatectomy. In the clinical context of biochemical relapse, it is essential to accurately determine the site of recurrence-pelvic(local, nodal, or bone) or distant-in order to select the optimal therapeutic management approach. Studies have demonstrated the value of mpM RI in detecting local recurrences-even in patients with low PSA levels(0.3-0.5 ng/m L)-and in diagnosing bone and nodal metastasis. The main objective of this review is to update the role of mpM RI prior to radical EBRT or SRT. | 5 | European cooperation to tackle the legacies of hexachlorocyclohexane(HCH)and lindane显示文摘Hexachlorocyclohexane(HCH)waste isomers from lindane production are the largest single POPs legacy,with an estimated 4.8 to 7.4 million tonnes of disposed waste.The largest part of this waste e 1.8 to 3 million tonnes e was disposed in Europe,where most producers were located.This paper provides a short overview of projects supported by the European Union(EU)to address this waste legacy and to implement the Stockholm Convention for this group of POPs with associated protection of soil,ecosystems and human health.We report here particularly on the results of a project financed by the EU called the“HCH in EU project”,which aimed to develop a systematic inventory of sites where HCH was handled and potentially resulted in contamination.The compiled information provide guidance for competent authorities to further assess their national HCH inventory and to further develop a strategy to address this large POP legacy in future.The systematic inventory revealed that there were at least 299 sites where HCH was handled.These sites include 54 former production sites,76 pesticide processing plants that used lindane,59 uncontrolled HCH waste isomer deposits,29 landfills with HCH waste,34 former or current storage sites for stocks of obsolete pesticides including technical HCH or lindane,and 16 HCH treatment or disposal sites.Additionally,at 31 of the sites lindane/technical HCH was used in applications with significant risk of soil pollution,such as wood treatment.The number of sites in this latter category is likely higher and will need further assessment.In addition to this inventory,the“HCH in EU project”produced detailed country reports,a guidance document for how to find potentially HCHimpacted sites,and a strategy document for implementing the sustainable management of these sites EU-wide,with proposed actions at the EU,country,and site level.Furthermore,the project has facilitated information exchange and e together with other related EU projects e has led to sharing information and best practices among member states and to establishing a network of authorities and other stakeholders working on the lindane/HCH waste legacy.This collaboration will facilitate a more systematic and better coordinated process to further assess,secure,and remediate the large HCH waste legacy and reduce and control lindane/HCH releases in the EU and possibly beyond.Such a coordinated effort and exchange of information for inventorying and managing contaminated sites might also be useful for other POPs such as PFOS/PFOA or dioxins. | John Vijgen Boudewijn Fokke Guido van de Coterlet Katja Amstaetter Javier Sancho Carlo Bensaïah Roland Weber | 2022 | Emerging Contaminants2022,8,1: | 4 |

| 8 | Glutathione metabolism is essential for self-renewal and chemoresistance of pancreatic cancer stem cells显示文摘BACKGROUND Cellular metabolism regulates stemness in health and disease.A reduced redox state is essential for self-renewal of normal and cancer stem cells(CSCs).However,while stem cells rely on glycolysis,different CSCs,including pancreatic CSCs,favor mitochondrial metabolism as their dominant energy-producing pathway.This suggests that powerful antioxidant networks must be in place to detoxify mitochondrial reactive oxygen species(ROS)and maintain stemness in oxidative CSCs.Since glutathione metabolism is critical for normal stem cell function and CSCs from breast,liver and gastric cancer show increased glutathione content,we hypothesized that pancreatic CSCs also rely on this pathway for ROS detoxification.AIM To investigate the role of glutathione metabolism in pancreatic CSCs.METHODS Primary pancreatic cancer cells of patient-derived xenografts(PDXs)were cultured in adherent or CSC-enriching sphere conditions to determine the role of glutathione metabolism in stemness.Real-time polymerase chain reaction(PCR)was used to validate RNAseq results involving glutathione metabolism genes in adherent vs spheres,as well as the expression of pluripotency-related genes following treatment.Public TCGA and GTEx RNAseq data from pancreatic cancer vs normal tissue samples were analyzed using the webserver GEPIA2.The glutathione-sensitive fluorescent probe monochlorobimane was used to determine glutathione content by fluorimetry or flow cytometry.Pharmacological inhibitors of glutathione synthesis and recycling[buthionine-sulfoximine(BSO)and 6-Aminonicotinamide(6-AN),respectively]were used to investigate the impact of glutathione depletion on CSC-enriched cultures.Staining with propidium iodide(cell cycle),Annexin-V(apoptosis)and CD133(CSC content)were determined by flow cytometry.Self-renewal was assessed by sphere formation assay and response to gemcitabine treatment was used as a readout for chemoresistance.RESULTS Analysis of our previously published RNAseq dataset E-MTAB-3808 revealed upregulation of genes involved in the KEGG(Kyoto Encyclopedia of Genes and Genomes)Pathway Glutathione Metabolism in CSC-enriched cultures compared to their differentiated counterparts.Consistently,in pancreatic cancer patient samples the expression of most of these up-regulated genes positively correlated with a stemness signature defined by NANOG,KLF4,SOX2 and OCT4 expression(P<10-5).Moreover,3 of the upregulated genes(MGST1,GPX8,GCCT)were associated with reduced disease-free survival in patients[Hazard ratio(HR)2.2-2.5;P=0.03-0.0054],suggesting a critical role for this pathway in pancreatic cancer progression.CSC-enriched sphere cultures also showed increased expression of different glutathione metabolism-related genes,as well as enhanced glutathione content in its reduced form(GSH).Glutathione depletion with BSO induced cell cycle arrest and apoptosis in spheres,and diminished the expression of stemness genes.Moreover,treatment with either BSO or the glutathione recycling inhibitor 6-AN inhibited self-renewal and the expression of the CSC marker CD133.GSH content in spheres positively correlated with intrinsic resistance to gemcitabine treatment in different PDXs r=0.96,P=5.8×1011).Additionally,CD133+cells accumulated GSH in response to gemcitabine,which was abrogated by BSO treatment(P<0.05).Combined treatment with BSO and gemcitabine-induced apoptosis in CD133+cells to levels comparable to CD133-cells and significantly diminished self-renewal(P<0.05),suggesting that chemoresistance of CSCs is partially dependent on GSH metabolism.CONCLUSION Our data suggest that pancreatic CSCs depend on glutathione metabolism.Pharmacological targeting of this pathway showed that high GSH content is essential to maintain CSC functionality in terms of self-renewal and chemoresistance. | Petra Jagust Sonia Alcala Bruno Sainz Jr Christopher Heeschen Patricia Sancho | 2020 | World Journal of Stem Cells2020,12,11: | 2 |
