| 1 | Atorvastatin protects cardiac progenitor cells from hypoxia-induced cell growth inhibition via MEG3/miR-22/HMGB1 pathway显示文摘Heart failure (HF)induced by ischemia myocardial infarction (MI)is one of the major causes of morbidity and mortality all around the world.Atorvastatin,a hydroxyrnethylglutaryl coenzyme A reductase inhibitor,has been demonstrated to benefit patients with ischemic or non-ischemicinduced HF,but the mechanism is still poorly understood.Increasing evidence indicates that IncRNAs play important role in variety of human disease.However,the role and underlying molecular mechanisms remain largely unclear.In our work,we applied 0.5%02 to generate a hypoxia cardiac progenitor cell (CPC)model.Then,CCK8 and EdU assays were employed to investigate the role of atorvastatin in hypoxia CPC cell model.We found that hypoxia inhibits CPC viability and proliferation through modulating MEG3 expression,while atorvastatin application can protect CPCs from hypoxia-induced injury through inhibiting MEG3 expression.Then,we demonstrated that repression of MEG3 inhibited the hypoxia-induced injury of CPCs and overexpression of MEG3 inhibited the protective effect of atorvastatin in the hypoxia-induced injury of CPCs.Furthermore,our study illustrated that atorvastatin played its role in CPC viability and proliferation by modulating the expression of HMGB1 through the MEG3/miR-22 pathway.Our study, for the first time,uncovered the molecular mechanism of atorvastatin's protective role in cardiomyocytes under hypoxia condition,which may provide an exploitable target in developing effective therapy drugs for MI patients. | Jinwen Su Ming Fang Bei Tian Jun Luo Can Jin Xuejun Wang Zhongping Ning Xinming Li | 2018 | Acta Biochimica et Biophysica Sinica2018,50,12: | 6 |
| 2 | Relationship between P-glycoprotein and CD44 expression in esophageal carcinoma显示文摘Objective: To investigate the relationship between P-glycoprotein (P-gp) and adhesion molecule CD44 expression as well as their clinical significance in esophageal carcinoma. Methods: To examine the expressed level of P-gp and CD44 by flow cytometry (FCM) in the operated samples of 70 cases with esophageal carcinoma and their normal mucosa of esophageal incision, and to evaluate their relationship with clinicopathological factors. Results: Among the 70 cases with esophageal car- cinoma, the expression of P-gp in the 27 cases (38.6%) was negative (positive cells <25%); 11 cases (15.7%) were 25%–40% expression of P-gp positive cells; 14 cases (20%) were 41%–60% expression of P-gp positive cells; 18 cases (25.7%) were the high expression (positive cells >60%) of P-gp. Of the cases with the tumor sizes being more than 4 cm, the expression of CD44 showed a significant difference (P<0.05) in 25 cases with P-gp positive, compared with 19 cases with P-gp negative. Of the cases with high-mild differentiated esophageal carcinoma, the expression of CD44 showed a significant difference (P<0.05) in 22 cases with P-gp positive, compared with 17 cases with P-gp negative. Of the cases with clinical III--IV stage, the expression of CD44 showed a significant difference (P<0.05) in 26 cases with P-gp positive, compared with 10 cases with P-gp negative. Of the cases with lymph node metastasis, the CD44 expression showed a significant difference (P=0.050) in 27 cases with P-gp positive, compared with 11 cases with P-gp negative. Of the cases of the patients’ age being more than 56 years, the expression of CD44 showed a significant difference (P<0.01) in 27 cases with P-gp positive, compared with 12 cases with P-gp negative. When the P-gp and CD44 expression were positive, the clinical II stage and III–IV stage in esophageal carcinoma was showed a significant difference (P<0.05). | 4 | Preparation of SiC Porous Ceramics by Crystalline Silicon Cutting Waste显示文摘SiC porous ceramics were prepared at 1 400 ℃ for4 h with crystalline silicon cutting waste and activated carbon as main starting materials and NH_4HCO_3 as the pore-forming agent. Effects of NH_4HCO_3 additions( 0,20%,30%,40%,by mass) on the phase composition,microstructure,sintering properties,cold compressive strength and thermal shock resistance of as-prepared Si C porous ceramics were investigated. The results show that:( 1) addition of NH_4HCO_3 remarkably influences the apparent porosity and cold compressive strength of specimens. The apparent porosity achieves its maximum value( 63. 40%) when 40% NH_4HCO_3 is added,while the minimum cold compressive strength is 4. 77 MPa;( 2) the specimen with 40% NH_4HCO_3 has the best thermal shock resistance. | 5 | Effect of Starch Addition on Properties of Corundum-mullite Porous Ceramics显示文摘Corundum-mullite porous ceramics(CMPCs)were fabricated using fly ash and aluminum ash as the raw materials,adding different amounts of starch(0,10%,20%,and 30%,by mass)as the pore-forming agent,molding and firing at 1200℃ for 1 h.The apparent porosity,the bulk density,the cold compressive strength,the thermal shock resistance and the thermal conductivity of the CMPCs were tested.The phase composition and the microstructure of the CMPCs were characterized by XRD and SEM.The effect of the starch addition on the properties of the CMPCs was studied.The results show that:(1)the CMPCs with high porosity can be successfully prepared by adding starch as the pore-forming agent and firing at 1200℃ for 1 h;(2)with the starch addition increasing,the porosity increases;the cold compressive strength and the number of the quenching cycles of the CMPCs decrease,but they remain at high levels;meanwhile,the thermal conductivity decreases;(3)generally,when adding 20 mass%starch,the CMPC has the apparent porosity of 48.6%,the cold compressive strength of 52.1 MPa,the quenching cycles of 5 and the thermal conductivity of 1.63 W·m-1·K-1 and 1.52 W·m-1·K-1,respectively,at 25℃ and 500℃,showing good performance. | SU Chang MA Beiyue REN Xinming LIU Guoqiang ZHU Qiang | 2020 | China's Refractories2020,29,4: | 2 |

| 8 | Effect of Main Material Ratio on Properties of SiC-Al_2O_3 Porous Ceramics via Polymeric Replication Method显示文摘In order to fabricate porous ceramics with good properties and proper production cost,SiC-Al2 O3 porous ceramics were prepared at 1 450 ℃ for 2 h from the powders of commercial silicon carbide and white fused corundum via a polymeric replication method. Effects of the mass ratio of SiC powder to white fused corundum powder( 1 ∶ 3,1 ∶ 1 and 3 ∶ 1) on the appearance,phase composition,sintering properties and thermal shock resistance were investigated. The research results indicate that the as-prepared Si C-Al2 O3 porous ceramics have uniform pores,and their linear shrinkage ratio,apparent porosity and bulk density reach 4. 70%,67. 17%and 0. 83 g·cm-3,respectively. The thermal shock cycles from 1 400 ℃ to room temperature reach 23 including 15 cycles in air cooling condition and then 8 cycles in water cooling condition. Their main phases areα-Al2 O3 and Al6 Si2 O13 as well as a small amount of SiC and free SiO2. The volume of electronic components, especially the height, is very important for the structural and thermal design of electronic devices. This paper presents a design scheme of an online height measurement device based on laser triangulation and commercial charge coupled device(CCD). It analyzes the principles of electronic component height measurement, and expounds the composition and working principle of the laser measurement system. In addition, the commonly used methods to determine the center position are compared and analysed. These methods include circle fitting, gray centroid and extension method or Gaussian fitting. These methods usually lead to different results. The experimental results show that the fitting speed of the gray centroid is faster. | 12 | Hypoxia induces hypomethylation of the HMGB1 promoter via the MAPK/DNMT1/ HMGB1 pathway in cardiac progenitor cells显示文摘Apoptosis is involved in the death of cardiac progenitor cells (CPCs)after myocardial infarction (MI) in the heart.The loss of CPCs results in infarct scar and further deterioration of the heart function. Though stem cell-based therapy provides an effective approach for heart function recovery after MI,the retention of CPCs in the infarcted area of the heart is the main barrier that limits its promising therapy.Therefore,the underlying mechanisms of CPC apoptosis in hypoxia are important for the development of new therapeutic targets for MI patients.In this work,we found that the expression of high-mobility group box 1(HMGB1)was upregulated in CPCs under hypoxia conditions. Further study demonstrated that HMGBI was regulated by DNA methyltransferases 1 (DNMT1)via changing the methylation state of CpGs in the promoter of HMGB1 in CPCs duringhypoxia process.
