1Dexamethasone inhibits hypoxia-induced epithelial-mesenchymal transition in colon cancer显示文摘AIM:To elucidate the effects of dexamethasone on hypoxia-induced epithelial-to-mesenchymal transition(EMT) in colon cancer.METHODS:Human colon cancer HCT116 and HT29 cells were exposed to normoxic(21%) and hypoxic(1%) conditions. First,the effect of dexamethasone on cell viability was examined by MTT cell proliferation assay. In order to measure the expression levels of EMT markers(Snail,Slug,Twist,E-cadherin,and integrin αVβ6) and hypoxia-related genes [Hypoxia-inducible factor-1α(HIF-1α) and vascular endothelial growth factor(VEGF)] by dexamethasone,quantitative real-time polymerase chain reaction and western blot analysis were performed. Furthermore,the morphological changes of colon cancer cells and the expression pattern of E-cadherin by dexamethasone were detected through immunocytochemistry. Finally,the effects of dexamethasone on the invasiveness and migration of colon cancer cells were elucidated using matrigel invasion,migration,and wound healing migration assays.RESULTS:Under hypoxia,dexamethasone treatment inhibited HIF-1α protein level and its downstream gene,VEGF m RNA level in the colon cancer cell lines,HCT116 and HT29. In addition,the presence of dexamethasone down-regulated the m RNA levels of hypoxia-induced Snail,Slug,and Twist,all transcriptional factors of EMT,as well as hypoxia-induced integrin αVβ6 protein level,a well-known EMT marker for colon cancer cells. Furthermore,reduced E-cadherin in hypoxic condition was found to be recoverable by treating with dexamethasone in both colon cancer cell lines. Similarly,under hypoxic conditions,dexamethasone restored the growth pattern and morphological phenotype reminiscent of colon cancer cells grown under normoxic conditions; dexamethasone blocked the migration and invasion of both colorectal cancer cell lines in hypoxia.CONCLUSION:Our study suggested that dexamethasone has inhibitory effects on cell migration and invasion by suppressing EMT of colon cancer cell lines in hypoxic condition.Jung Ho Kim You-Jin Hwang Sang Hoon Han Young Eun Lee Saerom Kim Yoon Jae Kim Jae Hee Cho Kwang An Kwon Ju Hyun Kim Se-Hee Kim 2015World Journal of Gastroenterology2015,21,34:5

5Highly pixelated,untethered tactile interfaces for an ultra-flexible on-skin telehaptic system显示文摘Realizing highly immersive tactile interactions requires a skin-integrated,untethered,high-definition tactile transducer devices that can record and generate tactile stimuli.However,the rigid and bulky form factor,and insufficient resolution of existing actuators are hindering the reproduction of sophisticated tactile sensations and immersive user experiences.Here,we demonstrate an ultraflexible tactile interface with high spatial resolution of 1.8 mm for telehaptic communication on human skin.Dual mechanism sensors and sub-mm scale piezoceramic actuators are designed to record and generate the static and dynamic pressures in a wide frequency range(1 Hz to 1 kHz).Moreover,actuators are integrated on ultra-flexible substrate with chessboard pattern to minimize stress during mechanical deformations.Finally,remote transmissions of various tactile stimuli,such as shapes,textures,and vibration patterns were demonstrated by the telehaptic system with low latency(<1.55 ms)and high fidelity as proven by the shorttime Fourier-transform analysis.Hanbit Jin Yunjeong Kim Wooseup Youm Yulim Min Saerom Seo Chaehyun Lim Chan-Hwa Hong Seyoung Kwon Gyeongseok Park Steve Park Hye Jin Kim 2022npj Flexible Electronics2022,6,1:0
6Effect of open-field experimental warming on the leaf phenology of oriental oak (Quercus variabilis) seedlings显示文摘Aims An open-field warming experiment enables us to test the effects of projected temperature increase on change in plant phenology with fewer confounding factors and to study phenological response to temperature ranges beyond natural variability.This study aims to(i)examine the effect of temperature increase on leaf unfold-ing and senescence of oriental oak(Quercus variabilis Blume)under experimental warming and(ii)measure temperature-related parameters used in estimating phenological response to tempera-ture elevation.Methods Using an open-field warming system with infrared heaters,we increased the air temperature by~3℃ in the warmed plots compared with that of the control plots consistently for 2 years.Leaf unfolding and senescence dates of Q.variabilis seedlings were recorded and temperature-related phenological parameters were analysed.Important Findings The timing of leaf unfolding was advanced by 3-8 days(1.1-3.0 days/℃)and the date of leaf senescence was delayed by 14-19 days(5.0-7.3 days/℃)under elevated air temperatures.However,the cumulative degree days(CDD)of leaf unfolding were not significantly changed by experimental warming,which suggest the applicability of a constant CDD value to estimate the change in spring leaf phe-nology under 3℃ warming.Consistent ranges of advancement and temperature sensitivity in spring phenology and delayed autumn phe-nology and proposed temperature parameters from this study might be applied to predict future phenological change.Saerom Han Haegeun Chung Nam Jin Noh Sun Jeoung Lee Wooyong Jo Tae Kyung Yoon Koong Yi Chan-woo Park Suin Ko Yowhan Son 2014Journal of Plant Ecology2014,7,6:0
