| 1 | Correlation of p53 over-expression and alteration in p53 gene detected by polymerase chain reaction-single strand conformation polymorphism in adenocarcinoma of gastric cancer patients from India显示文摘AIM:To study the alterations in p53 gene among Indian gastric cancer patients and to correlate them with the various clinicopathological parameters. METHODS:A total of 103 gastric cancer patients were included in this study.The p53 alterations were studied by both immunohistochemical method as well as polymerase chain reaction(PCR)-single strand conformation polymorphism(SSCP)analysis.We only studied four(exon 5,6,7,and 8)of the 11 p53 exons.The alterations in p53 were also correlated with respect to various clinicopathological parameters. RESULTS:Among 103 cases,p53 over-expression and alteration were detected in 37(35.92%)and 19(18.44%)cases,respectively.Most of the p53 alterations were found at exon 5(31.54%),followed by exon 6(26.31%),exon 7(21.04%)and exon 8(21.04%).A significant correlation of p53 over- expression was found with p53 alteration(P=0.000). Concordance between p53 alteration(as detected by SSCP)and over-expression[as detected by immunohistochemistry(IHC)]was found in 75%cases. We found that IHC-positive/SSCP-negative cases accounted for 21%of cases and IHC-negative/SSCP- positive cases accounted for remaining 4%cases. CONCLUSION:Our results show that p53 genemutations are significantly correlated with p53 protein overxpression,with 75%concordance in overexpression and alteration in the p53 gene,but 25% disconcordance also cautions against the assumption that p53 over-expression is always associated with a gene mutation.There may be other mechanisms responsible for stabilization and accumulation of p53 protein with no evidence of gene mutation that reflect an accumulation of a non-mutated protein,or a false negative SSCP result. | Sajjad Karim Arif Ali | 2009 | World Journal of Gastroenterology2009,15,11: | 28 |

| 3 | Using Path Analysis to Identify the Influence of Climatic Factors on Spring Peak Flow Dominated by Snowmelt in an Alpine Watershed显示文摘Spring snowmelt peak flow(SSPF) can cause serious damage. Precipitation as rainfall directly contributes to the SSPF and influences the characteristics of the SSPF, while temperature indirectly impacts the SSPF by shaping snowmelt rate and determining the soil frozen state which partitions snowmelt water into surface runoff and soil infiltration water in spring. It is necessary to identify the important and significant paths of climatic factors influencing the SSPF and provide estimates of the magnitude and significance of hypothesized causal connections between climatic factors and the SSPF.This study used path analysis with a selection of five factors – the antecedent precipitation index(API),spring precipitation(SP), winter precipitation as snowfall(WS), <0°C temperature accumulation in winter(|ATN|), and average >0°C temperature accumulation in spring(AT) – to analyze their influences on the SSPF in the Kaidu River in Xinjiang,China. The results show that |ATN|, AT and WS have a significant correlation with the SSPF, while API and SP do not show a significant correlation. AT and WS directly influence the SSPF, while as the influence of |ATN| on SSPF is indirect through WS and AT. The indirect influence of |ATN| on SSPF through WS accounts for 69% of the total influence of |ATN| on SSPF. Compared to the multiple linear regression method, path analysis provides additional valuable information, including influencing paths from independent variables to the dependent variable as well as direct and indirect impacts of external variables on the internal variable. | 7 | An analytical model for estimating rock strength parameters from small-scale drilling data显示文摘The small-scale drilling technique can be a fast and reliable method to estimate rock strength parameters. It needs to link the operational drilling parameters and strength properties of rock. The parameters such as bit geometry, bit movement, contact frictions and crushed zone affect the estimated parameters.An analytical model considering operational drilling data and effective parameters can be used for these purposes. In this research, an analytical model was developed based on limit equilibrium of forces in a Tshaped drag bit considering the effective parameters such as bit geometry, crushed zone and contact frictions in drilling process. Based on the model, a method was used to estimate rock strength parameters such as cohesion, internal friction angle and uniaxial compressive strength of different rock types from operational drilling data. Some drilling tests were conducted by a portable and powerful drilling machine which was developed for this work. The obtained results for strength properties of different rock types from the drilling experiments based on the proposed model are in good agreement with the results of standard tests. Experimental results show that the contact friction between the cutting face and rock is close to that between bit end wearing face and rock due to the same bit material. | 12 | Evaluating effectiveness of frequency ratio, fuzzy logic and logistic regression models in assessing landslide susceptibility: a case from Rudraprayag district, India显示文摘Rudraprayag in Garhwal Himalayan division is one of the most vulnerable districts to landslides in India. Heavy rainfall, steep slope and developmental activities are important factors for the occurrence of landslides in the district. Therefore, specific assessment of landslide susceptibility and its accuracy at regional level is essential for disaster management and proper land use planning. The article evaluates effectiveness of frequency ratio, fuzzy logic and logistic regression models for assessing landslide susceptibility in Rudraprayag district of Uttarakhand state, India. A landslide inventory map was prepared and verified by field data. Fourteen landslide parameters and generated inventory map were utilized to prepare landslide susceptibility maps through frequency ratio, fuzzy logic and logistic regression models. Landslide susceptibility maps generated through these models were classified into very high, high, medium, low and very low categories using natural breaks classification. Receiver operating characteristics(ROC) curve, spatially agreed area approach and seed cell area index(SCAI) method were used to validate the landslide models. Validation results revealed that fuzzy logic model was found to be more effective in assessing landslide susceptibility in the study area. | 15 | Differential gene expression of chemokines in KRAS and BRAF mutated colorectal cell lines:Role of cytokines显示文摘AIM:To study KRAS/BRAF mutations in colorectalcancer(CRC)that influences the efficacy of treatment.To develop strategies for overcoming combination of treatment.METHODS:Five colonic cell-lines were investigated:DLD-1 with KRAS(G13D)mutation,HT 29 and Colo205 with BRAF(V600E)mutation as well as the wild type(Wt)cell-lines Caco2 and Colo-320.DLD-1(KRAS),HT-29(BRAF)and Caco2(Wt)cell lines were treated with cytokines(TNFα50 ng,IL-1β1 ng and IFNγ50ng)and harvested at different time points(1-24 h).KRAS inhibition was performed by the siRNA-approach.Two colorectal cancer cells DLD-1 and Caco2 were used for KRAS inhibition.About 70%confluency were confirmed before transfection with small interferring RNA(siRNA)oligonucleotides.All the synthetic siRNA sequences were designed in our laboratory.Total RNA and protein was isolated from the cells for RT-PCR and Western blotting.Densitometry of the Western blotting was analyzed with the Image J software(NIH).Results are shown as mean±SD.RESULTS:RT-PCR analysis in non-stimulated cells showed a low basal expression of TNFαand IL-1βin the DLD-1 KRAS-mutated cell-line,compared to Caco2wild type.No detection was found for IL-6 and IFNγin any of the studied cell lines.In contrast,pro-angiogenic chemokines(CXCL1,CXCL8)showed a high constitutive expression in the mutated cell-lines DLD-1(KRAS),HT-29 and Colo205(BRAF),compared to wild type(Caco2).The anti-angiogenic chemokine(CXCL10)showed a high basal expression in wild-type,compared to mutated cell-lines.KRAS down-regulation by siRNA showed a significant decrease in CXCL1 and CXCL10gene expression in the DLD-1(KRAS)cell-line in comparison to wild type(Caco2)at 72 h after KRAS silencing.In contrast,the specific KRAS inhibition resulted in an up-regulation of CXCL1 and CXCL10.The results of our study show a higher expression of pro-angiogenic chemokines at basal level in mutated cell-lines,which was further increased by cytokine treatment.CONCLUSION:To summarize,basal chemokine gene expression for pro-angiogenic chemokines was high in mutated as compared to wild type cell-lines.This reflects the likely existence of a different microenvironment in tumours consistent of wild type or mutated cells.This may help to rationalize the choice of molecular targets for suitable therapeutic investigation in clinical studies. | Sajjad Khan Silke Cameron Martina Blaschke Federico Moriconi Naila Naz Ahmad Amanzada Giuliano Ramadori Ihtzaz Ahmed Malik | 2014 | World Journal of Gastroenterology2014,20,11: | 6 |

Around 18 million people are estimated to be infected with HDV. An important trend in HDV infection is global decline. HDV prevalence has decreased significantly in Europe since the 1970s and 1980s when it was f irst reported. The Asia-Pacif ic region now seems to be where HDV is a major health concern. There is a lack of available data from most of the countries from this region; hence, the true status of HDV cannot be determined. In South Asia, most of the countries have conditions that are favorable for the spread of hepatitis B and other related infections. Countries like Pakistan and Iran have shown an increase in HDV prevalence over a period of time. Other countries and region like China, Turkey, Australia, Japan, India and Taiwan, some of which had very high HDV prevalence in the past, have shown a decline in the incidence, but high prevalence persists in some. | 18 | Eh耦合的稻田土壤镉、砷释放机制显示文摘镉(Cd)、砷(As)是稻田土壤的主要重(类)金属污染物,稻田土壤Cd、As的释放与其氧化还原电位(Eh)密切相关,但Eh耦合的稻田土壤砷、镉释放机制仍不清楚.本实验利用高精度土壤微反应器模拟Eh梯度变化过程(-250、-50、170 mV),研究其耦合的土壤Cd、As及相关元素(Fe、Al、Ca、S、P、DOC)的释放特征,并结合同步辐射微区X射线荧光光谱技术(μ⁃XRF)探讨了不同Eh(-250、-50、170 mV)条件下稻田土壤As释放机制.结果表明:随着供试体系Eh从-250 mV升高至170 mV,土水混合液中DOC和pH值逐步下降,但Cd、As及相关Al、S、P、Fe元素浓度则同时升高.其中,Cd与S、Al增长均呈先缓后快特征,而As与P、Fe增长则呈先快后缓趋势,说明在Eh升高过程中Cd与S,以及As与Fe间密切相关.同步辐射μ⁃XRF分析发现,在低Eh条件下,土壤颗粒上As、Fe和S的高含量点信号重合,As⁃S及As⁃Fe的可决系数分别在Eh为-250 mV和170 mV条件下最高,说明在低Eh条件下As很可能主要受硫化铁控制,但在高Eh条件下则受铁氧化物影响.综上可知,低Eh条件下,As、Cd主要受到硫化铁的控制.随着Eh的升高,土壤Cd的活性逐渐升高,As的活性则受到铁氧化物控制.本研究结果可为理解土壤Cd、As共同释放机制及调控土壤Cd、As活性提供重要理论依据. | 张良东 杨建军 夏星 Sajjad Hussain | 2020 | 环境科学学报2020,40,5: | 6 |

| 20 | CRISPR/Cas9-mediated PBP1 and PBP3 mutagenesis induced significant reduction in electrophysiological response to sex pheromones in male Chilo suppressalis显示文摘Pheromone-binding proteins (PBPs) are thought to bind and transport sex pheromones onto the olfactory receptors on the dendrite membrane of olfactory neurons, and thus play a vital role in sex pheromone perception. However, the function of PBPs has rarely been demonstrated in vivo.In this study, two PBPs (PBP1 and PBP3) of Chilo suppressalis, one of the most notorious pyralid pests, were in vivo functionally characterized using insects with the PBP gene knocked out by the CRISPR/Cas9 system. First, through direct injection of PBP-single guide RNA (sgRNA)/Cas9 messenger RNA into newly laid eggs, a high rate of target-gene editing (checked with polled eggs) was induced at 24 h after injection, 21.3% for PBPl-sgRNA injected eggs and 19.5% for PBP3-sgRNA injected eggs. Second, by an in-crossing strategy, insects with mutant PBP1 or PBP3 (both with a premature stop codon) were screened and homozygous mutants were obtained in the G3 generation. Third, the mutant insects were measured for electroantennogram (EAG) response to female sex pheromones. As a result, both PBP mutant males displayed significant reduction in EAG response, and this reduction in PBP1 mutants was higher than that in PBP3 mutants, indicating a more important role of PBP1. Finally, the relative importance of two PBPs and the possible off target effect induced by sgRNA-injection are discussed.
