2Immunization with chlamydial plasmid protein pORF5 DNA vaccine induces protective immunity against genital chlamydial infection in mice显示文摘To validate the immune protective efficacy of pORF5 DNA vaccine and to analyze potential mechanisms related to this protection. In this study, pORF5 DNA vaccine was constructed and evaluated for its protective immunity in a mouse model of genital chlamydial infection. Groups of BALB/c mice were immunized intranasally with pORF5 DNA vaccine. Humoral and cell mediated immune responses were evaluated. The clearance ability of chlamydial challenge from the genital tract and the chlamy- dia-induced upper genital tract gross pathology and histopathological characterization were also de- tected. The results showed that the total and the IgG2a anti-pORF5 antibody levels in serum were sig- nificantly elevated after pcDNA3.1-pORF5 vaccination, as were the total antibody and IgA levels in vaginal fluids. pcDNA3.1-pORF5 induced a significantly high level of Th1 response as measured by robust gamma interferon (IFN-γ). Minimal IL-4 was produced by immune T cells in response to the re-stimulation with pORF5 protein or the inactive elementary body in vitro. pcDNA3.1-pORF5-vacci- nated mice displayed significantly reduced bacterial shedding upon a chlamydial challenge and an accelerated resolution of infection. 100% of pcDNA3.1-pORF5 vaccinated mice successfully resolved the infection by day 24. pcDNA3.1-pORF5-immunized mice also exhibited protection against patho- logical consequences of chlamydial infection. The stimulated index was significantly higher than that of mice immunized with pcDNA3.1 and PBS (P<0.05). 5Association of a SLC30A8 Genetic Variant with Monotherapy of Repaglinide and Rosiglitazone Effect in Newly Diagnosed Type 2 Diabetes Patients in China显示文摘Objective To investigate a potential relationship between Solute carrier family 30 (zinc transporter) member 8 (SLC30A8) rs13266634 variant and efficacy of rosiglitazone or repaglinide in treating newly diagnosed Chinese type 2 diabetes patients.Methods A total of 209 diabetic patients without any antihyperglycemic history were recruited and treated with repaglinide or rosiglitazone randomly for 48 weeks (104 and 105 patients,respectively).Anthropometric measurements and clinical laboratory tests were carried out before and after the treatment.An non-synonymous variant rs13266634 was genotyped by matrix-assisted laser desorption ionization-time of flight mass spectroscopy.Results Ninety-one patients in repaglinide group and ninety-three patients in rosiglitazone group completed the study.Δ value of homeostasis model assessment of beta cell function (HOMA-B)and Δ value of fasting proinsulin levels were statistically significant between three genotype groups (P=0.0149 and 0.0246,respectively) after rosiglitazone treatment.However,no genotype association was observed in the repaglinide or rosiglitazone group with other parameters.Conclusion The SLC30A8 variant was associated with the efficacy of insulin sensitizer monotherapy on insulin secretion in patients with newly diagnosed type 2 diabetes mellitus in Shanghai,China.JIANG Feng LI Qing HU Cheng ZHANG Rong WANG Cong Rong YU Wei Hui LU Jing Yi TANG Shan Shan BAO Yu Qian XIANG Kun San JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:8
6Effect of matrine and carvedilol on collagen and MMPs activity of hypertrophy myocardium induced by pressure overload显示文摘Objective: To explore the effect and mechanism of matrine (Mt.) on myocardial interstitial fibrosis induced by pressure overload. Methods: Pressure overloaded myocardial hypertrophy was produced by banding of aorta abdominalis in 67 male Sprague-Dawley rats weighing (200±15) g. The rats were assigned into one of the following groups: sham-operation control, operation control, operation group treated with matrine (15 mg/(kgd)) and treated with carvedilol (Car.) (3.6 mg/(kgd)) group. The rats were given drugs one day after operation. Five weeks after treatment, the left ventricular weight (LVW) was measured and the volume of myocardial cells was detected with Hematoxylin-Eosin (H-E) stain and Masson stain was used to assess the level of fibrosis of the myocardial matrix. Myocardial metalloproteinase activity was quantified with zymography, and survival rate was calculated. Results: Survival rate significantly decreased (P<0.05), LVW/BW (body weight), MMP-2 (matrix metalloproteinase-2) activity (P<0.05), size of cardiomyocytes and interstitial fibrosis obviously increased in the operation group compared with sham control group. Mt. and Car. treatment can significantly increase survival rate (P<0.05), decrease LVW/BW (P<0.05) and MMP-2 activity (P<0.05), decrease size of cardiomyocytes and interstitial fibrosis compared with operation group. But there was differ-ence compared with sham group. 7The effects of the guide field on the structures of electron density depletions in collisionless magnetic reconnection显示文摘Two-dimensional particle-in-cell simulations are performed to investigate the formation of electron density depletions in collisionless magnetic reconnection.In anti-parallel reconnection,the quadrupole structures of the out-of-plane magnetic field are formed,and four symmetric electron density depletion layers can be found along the separatrices due to the effects of magetic mirror.With the increase of the initial guide field,the symmetry of both the out-of-plane magnetic field and electron density depletion layers is distorted.When the initial guide field is sufficiently large,the electron density depletion layers along the lower left and upper right separatrices disappear.The parallel electric field in guide field reconnection is found to play an important role in forming such structures of the electron density depletion layers.The structures of the out-of-plane magnetic field By and electron depletion layers in anti-parallel and guide field reconnection are found to be related to electron flow or in-plane currents in the separatrix regions.In anti-parallel reconnection,electrons flow towards the X line along the separatrices,and are directed away from the X line along the magnetic field lines just inside the separatrices.In guide field reconnection,electrons can only flow towards the X line along the upper left and lower right separatrices due to the existence of the parallel electric field in these regions.LU San LU QuanMing CAO Yang HUANG Can XIE JinLin WANG Shui 2011Chinese Science Bulletin2011,56,1:8
8Electron dynamics in collisionless magnetic reconnection显示文摘Magnetic reconnection provides a physical mechanism for fast energy conversion from magnetic energy to plasma kinetic energy. It is closely associated with many explosive phenomena in space plasma, usually collisionless in character. For this reason, researchers have become more interested in collisionless magnetic reconnection. In this paper, the various roles of electron dynamics in collisionless magnetic reconnection are reviewed. First, at the ion inertial length scale, ions and electrons are decoupled. The resulting Hall effect determines the reconnection electric field. Moreover, electron motions determine the current system inside the reconnection plane and the electron density cavity along the separatrices. The current system in this plane produces an out-of-plane magnetic field. Second, at the electron inertial length scale, the anisotropy of electron pressure determines the magnitude of the reconnection electric field in this region. The production of energetic electrons, which is an important characteristic during magnetic reconnection, is accelerated by the reconnection electric field. In addition, the different topologies, temporal evolution and spatial distribution of the magnetic field affect the accelerating process of electrons and determine the final energy of the accelerated electrons. Third, we discuss results from simulations and spacecraft observations on the secondary magnetic islands produced due to secondary instabilities around the X point, and the associated energetic electrons. Furthermore, progress in laboratory plasma studies is also discussed in regard to electron dynamics during magnetic reconnection. 10Human adenovirus(HAdV)infection in children with acute respiratory tract infections in Guangzhou,China,2010–2021:a molecular epidemiology study显示文摘Background Human adenovirus(HAdV)infection can cause a variety of diseases.It is a major pathogen of pediatric acute respiratory tract infections(ARIs)and can be life-threatening in younger children.We described the epidemiology and subtypes shifting of HAdV among children with ARI in Guangzhou,China.Methods We conducted a retrospective study of 161,079 children diagnosed with acute respiratory illness at the Guangzhou Women and Children’s Medical Center between 2010 and 2021.HAdV specimens were detected by real-time PCR and the hexon gene was used for phylogenetic analysis.Results Before the COVID-19 outbreak in Guangzhou,the annual frequency of adenovirus infection detected during this period ranged from 3.92%to 13.58%,with an epidemic peak every four to fve years.HAdV demonstrated a clear seasonal distribution,with the lowest positivity in March and peaking during summer(July or August)every year.A signifcant increase in HAdV cases was recorded for 2018 and 2019,which coincided with a shift in the dominant HAdV subtype from HAdV-3 to HAdV-7.The latter was associated with a more severe disease compared to HAdV-3.The average mortality proportion for children infected with HAdV from 2016 to 2019 was 0.38%but increased to 20%in severe cases.After COVID-19 emerged,HAdV cases dropped to 2.68%,suggesting that non-pharmaceutical interventions probably reduced the transmission of HAdV in the community.Conclusion Our study provides the foundation for the understanding of the epidemiology of HAdV and its associated risks in children in Southern China.Yi Chen Tao Lin Chang‑Bing Wang Wan‑Li Liang Guang‑Wan Lian Mark Zanin Sook‑San Wong Xin‑Gui Tian Jia‑Yu Zhong Ying‑Ying Zhang Jia‑Hui Xie Ling‑Ling Zheng Fei‑Yan Chen Run Dang Ming‑Qi Zhao Yi‑Yu Yang Rong Zhou Bing Zhu 2022World Journal of Pediatrics2022,18,8:5

12Magnetotail dipolarization fronts and particle acceleration:A review显示文摘In this paper, the particle acceleration processes around magnetotail dipolarization fronts(DFs) were reviewed. We summarize the spacecraft observations(including Cluster, THEMIS, MMS) and numerical simulations(including MHD, testparticle, hybrid, LSK, PIC) of these processes. Specifically, we(1) introduce the properties of DFs at MHD scale, ion scale, and electron scale,(2) review the properties of suprathermal electrons with particular focus on the pitch-angle distributions,(3)define the particle-acceleration process and distinguish it from the particle-heating process,(4) identify the particle-acceleration process from spacecraft measurements of energy fluxes, and(5) quantify the acceleration efficiency and compare it with other processes in the magnetosphere(e.g., magnetic reconnection and radiation-belt acceleration processes). We focus on both the acceleration of electrons and ions(including light ions and heavy ions). Regarding electron acceleration, we introduce Fermi,betatron, and non-adiabatic acceleration mechanisms;regarding ion acceleration, we present Fermi, betatron, reflection, resonance, and non-adiabatic acceleration mechanisms. 13Costimulation of resting B lymphocytes alters the IL-4-activated IRS2 signaling pathway in a STAT6 independent manner: implications for cell survival and proliferation显示文摘IL-4 is an important B cell survival and growth factor. IL-4 induced the tyrosine phosphorylation of IRS2 in resting B lymphocytes and in LPS- or CD40L-activated blasts. Phosphorylated IRS2 coprecipitated with the p85 subunit of PI 3’ kinase in both resting and activated cells. By contrast, association of phosphorylated IRS2 with GRB2 was not detected in resting B cells after IL-4 treatment although both proteins were expressed. However, IL-4 induced association of IRS2 with GRB2 in B cell blasts. The pattern of IL-4- induced recruitment of p85 and GRB2 to IRS2 observed in B cells derived from STAT6 null mice was identical to that observed for normal mice. While IL-4 alone does not induce activation of MEK, a MEKI inhibitor suppressed the IL-4-induced proliferative response of LPS-activated B cell blasts. These results demonstrate that costimulation of splenic B cells alters IL-4-induced signal transduction independent of STAT6 leading to proliferation. 15Free-standing ZnO nanorod arrays modified with single-walled carbon nanotubes for betavoltaics and photovoltaics显示文摘A full-duplex radiant energy converter based on both betavoltaic and photovoltaic effects in an easyto-implement way is an attractive alternative for the autonomous wireless sensor microsystem.Here,we report a novel beta/photovoltaic cell based on free-standing Zn O nanorod arrays(ZNRAs)modified with metallic single-walled carbon nanotubes(m-SWCNTs),using radioisotope63 Ni as beta-emitting source.The ZNRAs were grown on Al-doped Zn O(AZO)conductive glass using hydrothermal method.The optimum length and diameter of Zn O nanorods were determined by Monte Carlo simulation for beta energy deposition in ZNRAs.The m-SWCNTs were anchored into the ZNRAs to form a three-dimensional(3-D)Schottky junction structure for effectively separating the beta/photo-excited electron-hole pairs.Experimentally,the betavoltaic and photovoltaic effects were confirmed through the I-V measurements of beta/photovoltaic cells under beta/UV/Vis irradiations,respectively.It is suggested that the m-SWCNTs play key role for the enhancement of beta/photovoltaic performance through the formation of extensive3-D Schottky junction,the conductive network for hole transport,and the surface plasmon resonance exciton absorption for visible light.Changsong Chen Jiang Chen Zhen Wang Jian Zhang Haisheng San Shichao Liu Chunyu Wu Werner Hofmann 2020Journal of Materials Science & Technology2020,54,19:3

19Genome-wide association study and genomic prediction of Fusarium ear rot resistance in tropical maize germplasm显示文摘Fusarium ear rot(FER)is a destructive maize fungal disease worldwide.In this study,three tropical maize populations consisting of 874 inbred lines were used to perform genomewide association study(GWAS)and genomic prediction(GP)analyses of FER resistance.Broad phenotypic variation and high heritability for FER were observed,although it was highly influenced by large genotype-by-environment interactions.In the 874 inbred lines,GWAS with general linear model(GLM)identified 3034 single-nucleotide polymorphisms(SNPs)significantly associated with FER resistance at the P-value threshold of 1×10^(-5),the average phenotypic variation explained(PVE)by these associations was 3%with a range from 2.33%to 6.92%,and 49 of these associations had PVE values greater than 5%.The GWAS analysis with mixed linear model(MLM)identified 19 significantly associated SNPs at the P-value threshold of 1×10^(-4),the average PVE of these associations was 1.60%with a range from 1.39%to 2.04%.Within each of the three populations,the number of significantly associated SNPs identified by GLM and MLM ranged from 25 to 41,and from 5 to 22,respectively.Overlapping SNP associations across populations were rare.A few stable genomic regions conferring FER resistance were identified,which located in bins 3.04/05,7.02/04,9.00/01,9.04,9.06/07,and 10.03/04.The genomic regions in bins 9.00/01 and 9.04 are new.GP produced moderate accuracies with genome-wide markers,and relatively high accuracies with SNP associations detected from GWAS.Moderate prediction accuracies were observed when the training and validation sets were closely related.These results implied that FER resistance in maize is controlled by minor QTL with small effects,and highly influenced by the genetic background of the populations studied.Genomic selection(GS)by incorporating SNP associations detected from GWAS is a promising tool for improving FER resistance in maize.Yubo Liu Guanghui Hu Ao Zhang Alexander Loladze Yingxiong Hu Hui Wang Jingtao Qu Xuecai Zhang Michael Olsen Felix San Vicente Jose Crossa Feng Lin Boddupalli M.Prasanna 2021The Crop Journal2021,9,2:3
20Genomic prediction of the performance of hybrids and the combining abilities for line by tester trials in maize显示文摘The two most important activities in maize breeding are the development of inbred lines with high values of general combining ability(GCA)and specific combining ability(SCA),and the identification of hybrids with high yield potentials.Genomic selection(GS)is a promising genomic tool to perform selection on the untested breeding material based on the genomic estimated breeding values estimated from the genomic prediction(GP).In this study,GP analyses were carried out to estimate the performance of hybrids,GCA,and SCA for grain yield(GY)in three maize line-by-tester trials,where all the material was phenotyped in 10 to 11 multiple-location trials and genotyped with a mid-density molecular marker platform.Results showed that the prediction abilities for the performance of hybrids ranged from 0.59 to0.81 across all trials in the model including the additive effect of lines and testers.In the model including both additive and non-additive effects,the prediction abilities for the performance of hybrids were improved and ranged from 0.64 to 0.86 across all trials.The prediction abilities of the GCA for GY were low,ranging between-0.14 and 0.13 across all trials in the model including only inbred lines;the prediction abilities of the GCA for GY were improved and ranged from 0.49 to 0.55 across all trials in the model including both inbred lines and testers,while the prediction abilities of the SCA for GY were negative across all trials.The prediction abilities for GY between testers varied from-0.66 to 0.82;the performance of hybrids between testers is difficult to predict.GS offers the opportunity to predict the performance of new hybrids and the GCA of new inbred lines based on the molecular marker information,the total breeding cost could be reduced dramatically by phenotyping fewer multiple-location trials.Ao Zhang Paulino Pérez-Rodríguez Felix San Vicente Natalia Palacios-Rojas Thanda Dhliwayo Yubo Liu Zhenhai Cui Yuan Guan Hui Wang Hongjian Zheng Michael Olsen Boddupalli M.Prasanna Yanye Ruan Jose Crossa Xuecai Zhang 2022The Crop Journal2022,10,1:3
