1Acupuncture's Role in Solving the Opioid Epidemic: Evidence, Cost-Effectiveness, and Care Availability for Acupuncture as a Primary, Non-Pharmacologic Method for Pain Relief and Management-White Paper 2017显示文摘The United States(U.S.) is facing a national opioid epidemic, and medical systems are in need of nonpharmacologic strategies that can be employed to decrease the public's opioid dependence. Acupuncture has emerged as a powerful, evidence-based, safe, cost-effective, and available treatment modality suitable to meeting this need. Acupuncture has been shown to be effective for the management of numerous types of pain conditions, and mechanisms of action for acupuncture have been described and are understandable from biomedical, physiologic perspectives. Further, acupuncture's cost-effectiveness can dramatically decrease health care expenditures, both from the standpoint of treating acute pain and through avoiding addiction to opioids that requires costly care, destroys quality of life, and can lead to fatal overdose. Numerous federal regulatory agencies have advised or mandated that healthcare systems and providers offer non-pharmacologic treatment options for pain. Acupuncture stands out as the most evidence-based, immediately available choice to fulfil these calls. Acupuncture can safely, easily, and cost-effectively be incorporated into hospital settings as diverse as the emergency department, labor and delivery suites, andneonatal intensive care units to treat a variety of commonly seen pain conditions. 3Uplift, climate and biotic changes at the Eocene-Oligocene transition in south-eastern Tibet显示文摘The uplift history of south-eastern Tibet is crucial to understanding processes driving the tectonic evolution of the Tibetan Plateau and surrounding areas.Underpinning existing palaeoaltimetric studies has been regional mapping based in large part on biostratigraphy that assumes a Neogene modernization of the highly diverse,but threatened,Asian biota.Here,with new radiometric dating and newly collected plant-fossil archives,we quantify the surface height of part of the south-eastern margin of Tibet in the latest Eocene(~34 Ma)to be^3 km and rising,possibly attaining its present elevation(3.9 km)in the early Oligocene.We also find that the Eocene–Oligocene transition in south-eastern Tibet witnessed leaf-size diminution and a floral composition change from sub-tropical/warm temperate to cool temperate,likely reflective of both uplift and secular climate change,and that,by the latest Eocene,floral modernization on Tibet had already taken place,implying modernization was deeply rooted in the Palaeogene.Tao Su Robert A. 4No association between cyclooxygenase-2 and uridine diphosphate glucuronosyltransferase 1A6 genetic polymorphisms and colon cancer risk显示文摘AIM:To investigate the association of variations in the cyclooxygenase-2(COX2) and uridine diphosphate glucuronosyltransferase 1A6(UGT1A6) genes and non-steroidal anti-inflammatory drugs(NSAIDs) use with risk of colon cancer.METHODS:NSAIDs,which are known to reduce the risk of colon cancer,act directly on COX2 and reduce its activity.Epidemiological studies have associated variations in the COX2 gene with colon cancer risk,but others were unable to replicate this finding.Similarly,enzymes in the UGT1A6 gene have been demonstrated to modify the therapeutic effect of NSAIDs on colon adenomas.Polymorphisms in the UGT1A6 gene have been statistically shown to interact with NSAID intake to influence risk of developing colon adenomas,but not colon cancer.Here we examined the association of tagging single nucleotide polymorphisms(SNPs) in the COX2 and UGT1A6 genes,and their interaction with NSAID consumption,on risk of colon cancer in a population of 422 colon cancer cases and 481 population controls.RESULTS:No SNP in either gene was individually statistically significantly associated with colon cancer,nor did they statistically significantly change the protective effect of NSAID consumption in our sample.Like others,we were unable to replicate the association of variants in the COX2 gene with colon cancer risk(P > 0.05),and we did not observe that these variants modify the protective effect of NSAIDs(P > 0.05).We were able to confirm the lack of association of variants in UGT1A6 with colon cancer risk,although further studies will have to be conducted to confirm the association of these variants with colon adenomas.CONCLUSION:Our study does not support a role of COX2 and UGT1A6 genetic variations in the development of colon cancer.Cheryl L Thompson Sarah J Plummer Alona Merkulova Iona Cheng Thomas C Tucker Graham Casey Li Li 2009World Journal of Gastroenterology2009,15,18:11

7Spatiotemporal variation of drought characteristics in the Huang- Huai-Hai Plain, China under the climate change scenario显示文摘Understanding the potential drought characteristics under climate change is essential to reduce vulnerability and establish adaptation strategies,especially in the Huang-Huai-Hai Plain(3 H Plain),which is a major grain production area in China. In this paper,we investigated the variations in drought characteristics(drought event frequency,duration,severity,and intensity) for the past 50 years(1961–2010) and under future scenarios(2010–2099),based on the observed meteorological data and the Representative Concentration Pathway(RCP) 8.5 scenario,respectively. First,we compared the applicability of three climatic drought indices:the standardized precipitation index(SPI),the standardized precipitation evapotranspiration index based on the Penman-Monteith equation(SPEI-PM) and the same index based on the Thornthwaite equation(SPEI-TH) to correlate the recorded agricultural drought areas. Then,we analyzed the drought characteristics using ‘run theory' for both historical and the future RCP 8.5 scenario based on the best performing index. Correlation analyses between drought indices and agricultural drought areas showed that SPEI-PM performed better than SPI and SPEI-TH in the 3 H Plain. Based on the results of SPEI-PM,drought risks including duration,severity and intensity during 1961–2010 showed an decreasing trend. However,under the RCP 8.5 scenario,drought is expected to rise in frequency,duration,severity,and intensity from 2010–2099,although drought components during the 2010–2039 are predicted to be milder compared with historical conditions. This study highlights that the estimations for atmospheric evaporative demand would create differences in the prediction of long-term drought trends by different drought indices. 8Data-Driven Microstructure and Microhardness Design in Additive Manufacturing Using a Self-Organizing Map显示文摘To design microstructure and microhardness in the additive manufacturing(AM)of nickel(Ni)-based superalloys,the present work develops a novel data-driven approach that combines physics-based models,experimental measurements,and a data-mining method.The simulation is based on a computational thermal-fluid dynamics(CtFD)model,which can obtain thermal behavior,solidification parameters such as cooling rate,and the dilution of solidified clad.Based on the computed thermal information,dendrite arm spacing and microhardness are estimated using well-tested mechanistic models.Experimental microstructure and microhardness are determined and compared with the simulated values for validation.To visualize process-structure-properties(PSPs)linkages,the simulation and experimental datasets are input to a data-mining model-a self-organizing map(SOM).The design windows of the process parameters under multiple objectives can be obtained from the visualized maps.The proposed approaches can be utilized in AM and other data-intensive processes.Data-driven linkages between process,structure,and properties have the potential to benefit online process monitoring control in order to derive an ideal microstructure and mechanical properties.Zhengtao Gan Hengyang Li Sarah J.Wolff Jennifer L.Bennett Gregory Hyatt Gregory J.Wagner Jian Cao Wing Kam Liu 2019Engineering2019,5,4:4

12DNNGP, a deep neural network-based method for genomic prediction using multi-omics data in plants显示文摘Genomic prediction is an effective way to accelerate the rate of agronomic trait improvement in plants.Traditional methods typically use linear regression models with clear assumptions;such methods are unable to capture the complex relationships between genotypes and phenotypes.Non-linear models(e.g.,deep neural networks)have been proposed as a superior alternative to linear models because they can capture complex non-additive effects.Here we introduce a deep learning(DL)method,deep neural network genomic prediction(DNNGP),for integration of multi-omics data in plants.We trained DNNGP on four datasets and compared its performance with methods built with five classic models:genomic best linear unbiased prediction(GBLUP);two methods based on a machine learning(ML)framework,light gradient boosting machine(LightGBM)and support vector regression(SVR);and two methods based on a DL framework,deep learning genomic selection(DeepGS)and deep learning genome-wide association study(DLGWAS).DNNGP is novel in five ways.First,it can be applied to a variety of omics data to predict phenotypes.Second,the multilayered hierarchical structure of DNNGP dynamically learns features from raw data,avoiding overfitting and improving the convergence rate using a batch normalization layer and early stopping and rectified linear activation(rectified linear unit)functions.Third,when small datasets were used,DNNGP produced results that are competitive with results from the other five methods,showing greater prediction accuracy than the other methods when large-scale breeding data were used.Fourth,the computation time required by DNNGP was comparable with that of commonly used methods,up to 10 times faster than DeepGS.Fifth,hyperparameters can easily be batch tuned on a local machine.Compared with GBLUP,LightGBM,SVR,DeepGS and DLGWAS,DNNGP is superior to these existing widely used genomic selection(GS)methods.Moreover,DNNGP can generate robust assessments from diverse datasets,including omics data,and quickly incorporate complex and large datasets into usable models,making it a promising and practical approach for straightforward integration into existing GS platforms.Kelin Wang Muhammad Ali Abid Awais Rasheed Jose Crossa Sarah Hearne Huihui Li 2023Molecular Plant2023,16,1:3

14HIV care continuum among newly diagnosed student and non-student youths between 2012 and 2016 in Hangzhou, China显示文摘Background: Youths are disproportionally affected by the human immunodeficiency virus (HIV) infection. We aimed to assess antiretroviral therapy (ART) initiation and viral suppression rates among student and non-student youths in Hangzhou, China. Methods: Data were taken from the Chinese HIV/acquired immune deficiency syndrome Comprehensive Response Information Management System. Youths aged 15 to 24 years who were newly diagnosed with HIV between 2012 and 2016 and were living in Hangzhou were included in the study. Comparisons between student and non-student youths were made for ART initiation within 30 days, 90 days, and 12 months of HIV diagnosis, and the viral suppression rate at 12 months of HIV diagnosis and at 12 months of ART initiation. Results: A total of 707 cases met inclusion criteria, 29.6% of which were students and 70.4% were non-student youths. The student group had a higher proportion of ART initiation compared with the non-student group within 30 days of diagnosis (45.5% vs. 37.0%, P = 0.044), and a slightly higher but not statistically significant proportion at 90 days (67.0% vs. 62.7%), and 12 months (83.7% vs. 78.5%) of HIV diagnosis. ART initiation within 30 days improved from <15% in 2012 to over 65% in 2016 in both groups, and ART initiation within 90 days improved from <30% in 2012 to >90% in 2016. A smaller proportion of students experienced viral suppression compared with the non-student group (9.6% vs. 17.1%, P = 0.011) at 12 months after HIV diagnosis, but the suppression rate was similar at 12 months of ART initiation (69.9% vs. 71.1%, P = 0.743). Conclusions: ART initiation in both student and non-student youths has significantly improved between 2012 and 2016. However, the viral suppression rate remained unacceptably low at 12 months of HIV diagnosis in both student and non-student groups. 15No association between phosphatase and tensin homolog genetic polymorphisms and colon cancer显示文摘AIM: To investigate the association between single nucleotide polymorphisms (SNPs) in the phosphatase and tensin homolog (PTEN) tumor suppressor gene and risk of colon cancer. METHODS: We utilized a population-based casecontrol study of incident colon cancer individuals (n= 421) and controls (n = 483) aged ≥ 30 years to conduct a comprehensive tagSNP association analysis of the PTEN gene. RESULTS: None of the PTEN SNPs were statistically significantly associated with colon cancer when controlled for age, gender, and race, or when additionally adjusted for other known risk factors (P > 0.05). Haplotype analyses similarly showed no association between the PTEN gene and colon cancer. 16Post-hepatectomy liver regeneration in the context of bile acid homeostasis and the gut-liver signaling axis显示文摘Background:Liver regeneration following partial hepatectomy(PHx)is a complicated process involving multiple organs and several types of signaling networks.The bile acid-activated metabolic pathways occupy an auxiliary yet important chapter in the entire biochemical story.PHx is characterized by rapid but transient bile acid overload in the liver,which constitutes the first wave of proliferative signaling in the remnant hepatocytes.Bile acids trigger hepatocyte proliferation through activation of several nuclear receptors.Following biliary passage into the intestines,enterocytes reabsorb the bile acids,which results in the activation of farnesoid X receptor(FXR),the consequent excretion of fibroblast growth factor(FGF)19/FGF15,and its release into the enterohepatic circulation.FGF19/FGF15 subsequently binds to its cognate receptor,fibroblast growth factor receptor 4(FGFR4)complexed withβ-klotho,on the hepatocyte membrane,which initiates the second wave of proliferative signaling.Because some bile acids are toxic,the remnant hepatocytes must resolve the potentially detrimental state of bile acid excess.Therefore,the hepatocytes orchestrate a bile acid detoxification and elimination response as a protective mechanism in concurrence with the proliferative signaling.The response in part results in the excretion of(biotransformed)bile acids into the canalicular system,causing the bile acids to end up in the intestines.Relevance for patients:Recently,FXR agonists have been shown to promote regeneration via the gut-liver axis.This type of pharmacological intervention may prove beneficial for patients with hepatobiliary tumors undergoing PHx.In light of these developments,the review provides an in-depth account of the pathways that underlie post-PHx liver regeneration in the context of bile acid homeostasis in the liver and the gut-liver signaling axis.Lianne de Haan Sarah Jvan der Lely Anne-Loes K.Warps Quincy Hofsink Pim B.Olthof Mark J.de Keijzer Daniel A.Lionarons Lionel Mendes-Dias Bote G.Bruinsma Korkut Uygun Hartmut Jaeschke Geoffrey C.Farrell Narci Teoh Rowan Fvan Golen Tiangang Li Michal Heger 2018Journal of Clinical & Translational Research2018,4,1:2
