| 1 | 香蕉组织培养过程中内生菌污染的控制(英文)显示文摘研究了体外培养一种孟加拉传统香蕉(Musa spp.Cv.Kanthali)的茎尖组织。茎尖的原始细胞表面经无菌处理(0.1%HgCl2处理12min),接种6~15d后外植体地下茎部分仍有微生物污染(大部分是细菌),杀死了85%的外植体。为确定无污染培养基,将等量外植体分别浸泡在舍400mg/L氨苄青霉素和200mg/L庆大霉素(两种光谱抗生素)的培养基中1h。结果表明,经抗生素处理的外植体完全没有污染,但培养3周后不能再生。进行二次继代培养后,其中一部分外植体吸收了培养基并胀大,颜色由苍白转变成浅绿或深绿。三次继代培养后数天,不再观察到外植体的生长,所有经抗生素处理过的外植体都开始死亡。在未经抗生素处理的活外植体中,单个茎发育的最佳培养基是:Ms+4.0mg/LBA+0.5mg/L KT+15%CW,平均生长时间为18~21d,但再生率很低,只有30%。茎细胞增殖的最佳培养基是:MS+4.0mg/L BA+2.0mg/L IAA+15%CW,每个茎平均只萌发3~4个芽。最后,在添加0.5mg/L IBA的一半浓度的MS培养基中,体外培养茎最大生根率达到90%。 | Soubir Titov Salil Kumar Bhowmik Md.Sadrul Alam Sarder Nasir Uddin | 2007 | 生物工程学报2007,23,6: | 6 |
| 2 | Nutrient dynamics in the Sundarbans mangrove estuarine system of Bangladesh under different weather and tidal cycles显示文摘Introduction:Seasonal and tidal variations in nutrient concentration and water quality were investigated in the western Sundarbans of Bangladesh during the post-monsoon,winter and monsoon seasons during 2010–2011.Methods:Water collected from the surface,middle and bottom layers at six locations of the Kholpetua-Arpangashia river system during high and low tides was analyzed for temperature,salinity,pH,dissolved oxygen(DO),total dissolved solids(TDS),electrical conductivity(EC)and dissolved nutrients(NO3–N,PO4–P,SO4 and NH3–N).Results:The study revealed that nutrient concentrations were influenced by seasonal changes.Mean nutrient levels during post-monsoon,winter and monsoon seasons,respectively,were in the following ranges:nitrate(0.06–0.40,0.06–0.46 and 0.08–0.46 mg/L);phosphate(0.09–0.18,0.05–0.42 and 0.10–0.16 mg/L);sulphate(58.71–86.14,68.68–119.01 and 78.15–136.47 mg/L)and ammonia(0.02–0.08,0.02–0.04 and 0.26–0.38 mg/L).Increased levels of PO4–P,SO4 and NH3–N and lower DO and salinity were recorded during the monsoon period.Most of the experimental sites showed higher NO3–N content during monsoon,whereas few elevated concentrations were observed during post-monsoon and winter periods.High and low tidal waters contained mean nutrient levels in the following ranges:nitrate(0.05–0.46 and 0.04–0.40 mg/L);phosphate(0.05–0.42 and 0.07–0.18 mg/L);sulphate(63.63–125.36 and 58.71–136.47 mg/L)and ammonia(0.02–0.38 and 0.02–0.37 mg/L)without following any distinct fluctuation patterns.The western part of the Sundarbans receives less freshwater input during the monsoon season than other areas of the ecosystem,which reduces the variability of nutrient levels and water quality components.Conclusions:This study provides considerable advances in understanding the seasonality of nutrient distribution with possible tidal influence.The data generated from this study will guide continuing efforts to support a sound management for coastal mangrove ecosystems. | Shak Md Bazlur Rahaman Lipton Sarder Md Sayadur Rahaman Alokesh Kumar Ghosh Sudhin Kumar Biswas SM Shahjahan Siraj Khandaker Anisul Huq Abul Farah Md Hasanuzzaman Shikder Saiful Islam | 2013 | Ecological Processes2013,2,1: | 2 |
| 3 | Single nucleotide polymorphisms in candidate genes are significantly associated with resistance to Haemonchus contortus infection in goats显示文摘Background: Haemonchosis is a major economic problem in goat production in humid, tropical and subtropical regions. The disease is caused by an abomasal nematode, Haemonchus contortus, which is highly pathogenic in small ruminants. The aim of this study was to identifying single-nucleotide polymorphisms(SNP) that were associated with fecal egg counts(FEC) and could be used as markers to identify resistance to H. contortus in goats.Results: Ten novel variants in the CI TA, ATP2 A3, HSPA8, STAT5 B, ESYT1, and SERPING1 genes were associated with FEC in goats with a nominal significance level of P < 0.05. Two missense mutation in the exon region of the caprine CI TA gene resulted in replacement of arginine with cysteine at position 9473550(R9473550 C) and aspartic acid with glutamic acid at position 9473870(D9473870 E). Chinese goat breeds had significantly higher FEC than Bangladeshi goat breeds within their respective genotypes. Polymorphism information content(PIC), effective al ele number(Ne), and heterozygosity(He)were greatest for the STAT5 B197A > G SNP locus in al goat breeds. Pairwise coefficients of linkage disequilibrium(D′, r2)revealed complete LD(r2= 1) between significant SNP polymorphisms in CI TA and SERPING1 and strong LD(r2= 0.93 and0.98) between polymorphisms in HSPA8 and ATP2 A3, respectively. Correlation coefficient(r) between FEC and body weight(BW) was significantly positive(r = 0.56***, P < 0.001) but that between FEC and packed cel volume(PCV) was negatively significant(r =-0.47**, P < 0.01) in the total population of goats. On the other hand, correlation coefficient(r)between BW and PCV was not significant in total population of goats. Association analysis revealed that haplotypes within ATP2 A3, HSPA8, and SERPING1 were significantly associated with FEC.
