1‘翠冠’梨花芽休眠期碳水化合物变化及其相关基因表达研究显示文摘以‘翠冠’梨(Pyruspyrifolia Nakai)花芽为试材,连续两年研究了其在休眠过程中的碳水化合物含量变化及编码α-淀粉酶、β-淀粉酶和α-葡聚糖磷酸化酶等碳水化合物代谢相关基因的表达模式。两年的结果显示,随着‘翠冠’梨花芽休眠的加深,可溶性糖含量逐渐下降,11月15日降到最低。此后,在内休眠及其解除过程中可溶性糖含量呈上升趋势。花芽中淀粉含量随着休眠的加深逐渐下降,最低含量出现的时间在年度间有所不同。随着休眠开始解除,淀粉含量逐渐增加,在完全解除前达到最大值,其后则逐渐下降。‘翠冠’梨花芽的PpAMY、PpBAM1、PpBAM2和PpPHS在内休眠阶段均出现一个表达高峰,然后逐渐下降直到内休眠完全解除前。之后,除PpBAM2外,其余3个基因再次上调表达。‘翠冠’梨花芽休眠期间,淀粉含量的变化与PpAMY、PpBAM1、PpBAM2和PpPHS的表达变化之间存在联系,由此推测这些基因可能参与了‘翠冠’梨花芽休眠期碳水化合物代谢的调控。郑鹏华 刘国琴 Sayed Hussain 滕元文 2013园艺学报2013,40,2:11
2梨两个休眠相关MADS-box基因特征及在其休眠过程中的表达分析显示文摘PpMADS1和PpMADS2是从‘酥梨’(Pyrus pyrifolia white pear group‘Suli’)休眠芽转录组文库筛选的两个与休眠相关的MADS-box基因序列。为了解序列的特征,对其进行了相关生物信息学分析,并以‘翠冠’和‘圆黄’梨为试材,用实时定量PCR技术分析其花芽休眠不同阶段的表达变化。结果表明:两个基因都具有MADS-box家族的特征序列MIKC-基序,PpMADS1和PpMADS2分别与日本梨(P.pyrifolia‘Kosui’)休眠相关的两个MADS-box基因PpMADS13-1和PpMADS13-2聚在一起,且单独聚为一支,与李属植物休眠相关的MADS-box基因关系最近。在两个品种的休眠过程中,PpMADS1和PpMADS2的表达呈现相似的变化趋势,都有一个表达高峰,但‘翠冠’梨PpMADS1和PpMADS2的表达高峰均出现在11月15日,而‘圆黄’梨PpMADS1的表达高峰延后到12月30日,PpMADS2的表达高峰出现在12月15日。两个品种PpMADS1和PpMADS2的表达高峰都出现在内休眠解除之前,随内休眠解除其表达量下调,在生态休眠阶段表达量维持在较低的水平。据此推测PpMADS1和PpMADS2的表达对梨芽内休眠的解除具有调控作用。刘国琴 郑鹏华 Sayed Hussain 滕元文 2013园艺学报2013,40,4:11
3Stevenleaf from Gynostemma Pentaphyllum inhibits human hepatoma cell(HepG2)through cell cycle arrest and apoptotic induction显示文摘The anticancer activity of stevenleaf(SV)on the basis of cell viability,cell cycle,and apoptosis induction in HepG2 cancer cells were evaluated.SV controlled the growth of HepG2 cells with IC50 of 139.82μmol/L for 24 h,IC50 of 119.12μmol/L for 48 h and cell cycle arrested at G0/G1 phase,induced cell apoptosis and enhanced intracellular ROS generation.For cell cycle arrest,the mRNA expression levels of p21,p27 and p53 were up-regulated,while the expression levels of Cyclin A,Cyclin D1,Cyclin E and CDK1/2 were downregulated.SV efficiently up-regulated TNF R1,TRADD1 and FADD and down-regulated Caspase8 for cell death receptors;similarly,up-regulated Bax,Bak,Cytc,Apaf1,Caspase3 and Caspase9,and down-regulated Bcl2,Bcl xl and Bad for mitochondrial signal pathway.SV induced the mTOR-mediated cell apoptosis in HepG2 cells via activation of Akt and AMPK.The mechanistic explanation for the anticancer activity of SV as functional food can be derived from above results.Sayed Sajid Hussain Fan Zhang Yuanyuan Zhang Kiran Thakur Mahrukh Naudhani Carlos L.Cespedes-Acuna Zhaojun Wei 2020Food Science and Human Wellness2020,9,3:3
4Influence of heat stress on leaf ultrastructure, photosynthetic performance, and ascorbate peroxidase gene expression of two pear cultivars(Pyrus pyrifolia)显示文摘Plants encounter a variety of stresses in natural environments. One-year-old pot-grown trees of pear(Pyrus pyrifolia Nakai cv. Cuiguan and Wonhwang) were exposed to two heat stress regimes. Under constant short-term heat stress, chloroplasts and mitochondria were visibly damaged. Relative chlorophyll content and maximum photochemical efficiency of photosystem II were significantly decreased, which indicated that the leaf photosynthetic capability declined. Under chronic heat stress, mesophyll cell ultrastructure was not obviously damaged, but leaf photosynthetic capability was still restrained. As chronic heat stress was a simulation of the natural environment in summer, further study of the responses under this stress regime was undertaken. Ascorbate peroxidase(APX) activity was increased in ‘Cuiguan', but not in ‘Wonhwang'. Inducible expression of PpAPX genes in the cytoplasm, chloroplasts and peroxisomes was consistent with increased APX activity in ‘Cuiguan', whereas only weak induction of PpAPX genes was observed in ‘Wonhwang'. 10Role of second look endoscopy in endoscopic submucosal dissection and peptic ulcer bleeding:Meta-analysis of randomized controlled trials显示文摘BACKGROUND Second-look endoscopy(SLE)to prevent recurrent bleeding in patients with peptic ulcer disease(PUD)and those undergoing endoscopic submucosal dissection(ESD)is routinely being performed.Conflicting evidence exists regarding efficacy,risk,benefit,and cost-effectiveness.AIM To identify the role and effectiveness of SLE in ESD and PUD,associated rebleeding and PUD-related outcomes like mortality,hospital length of stay,need for endoscopic or surgical intervention and blood transfusions.METHODS A systematic review of literature databases PubMed,Cochrane,and Embase was conducted from inception to January 5,2023.Randomized controlled trials that compared patients with SLE to those who did not have SLE or evaluated the role of prophylactic hemostasis during SLE compared to other conservative interventions were included.The study was conducted per PRISMA guidelines,and the protocol was registered in PROSPERO(ID CRD42023427555:).RevMan was used to perform meta-analysis,and Mantel-Haenszel Odds ratio(OR)were generated using random effect models.RESULTS A total of twelve studies with 2687 patients were included in our systematic review and meta-analysis,of which 1074 patients underwent SLE after ESD and 1613 patients underwent SLE after PUD-related bleeding.In ESD,the rates of rebleeding were 7%in the SLE group compared to 4.4%in the non-SLE group with OR 1.65,95%confidence intervals(CI)of 0.96 to 2.85;P=0.07,whereas it was 11%in the SLE group compared to 13%in the non-SLE group with OR 0.895%CI:0.50 to 1.29;P=0.36.The mean difference in the blood transfusion rates in the SLE and no SLE group in PUD was OR 0.01,95%CI:-0.22 to 0.25;P=0.91.In SLE vs non-SLE groups with PUD,the OR for Endoscopic intervention was 0.29,95%CI:0.08 to 1.00;P=0.05 while it was OR 2.03,95%CI:0.95 to 4.33;P=0.07,for surgical intervention.The mean difference in the hospital length of stay was-3.57 d between the SLE and no SLE groups in PUD with 95%CI:-7.84 to 0.69;P=0.10,denoting an average of approximately 3 fewer days of hospital stay among patients with PUD who underwent SLE.For mortality between SLE and non-SLE groups in PUD,the OR was 0.88,95%CI:0.45 to 1.72;P=0.70.CONCLUSION SLE does not confer any benefit in preventing ESD and PUD-associated rebleeding.SLE also does not provide any significant improvement in mortality,need for interventions,or blood transfusions in PUD patients.SLE decreases the hospital length of stay on average by 3.5 d in PUD patients.Gowthami Sai Kogilathota Jagirdhar Jose Andres Perez Akshat Banga Rakhtan K Qasba Ruman K Qasba Harsha Pattnaik Muhammad Hussain Yatinder Bains Salim Surani 2024World Journal of Gastrointestinal Endoscopy2024,16,4:0
