| 1 | Proteomic and metabolomic profiling underlines the stage- and time-dependent effects of high temperature on grape berry metabolism显示文摘Climate change scenarios predict an increase in mean air temperatures and in the frequency,intensity,and length of extreme temperature events in many wine-growing regions worldwide.Because elevated temperature has detrimental effects on berry growth and composition,it threatens the economic and environmental sustainability of wine production.Using Cabernet Sauvignon fruit-bearing cuttings,we investigated the effects of high temperature(HT)on grapevine berries through a label-free shotgun proteomic analysis coupled to a complementary metabolomic study.Among the 2,279 proteins identified,592 differentially abundant proteins were found in berries exposed to HT.The gene ontology categories“stress,”“protein,”“secondary metabolism,”and“cell wall”were predominantly altered under HT.High temperatures strongly impaired carbohydrate and energy metabolism,and the effects depended on the stage of development and duration of treatment.Transcript amounts correlated poorly with protein expression levels in HT berries,highlighting the value of proteomic studies in the context of heat stress.Furthermore,this work reveals that HT alters key proteins driving berry development and ripening.Finally,we provide a list of differentially abundant proteins that can be considered as potential markers for developing or selecting grape varieties that are better adapted to warmer climates or extreme heat waves. | David Lecourieux Christian Kappel Stéphane Claverol Philippe Pieri Regina Feil John E.Lunn Marc Bonneu Lijun Wang Eric Gomès Serge Delrot Fatma Lecourieux | 2020 | Journal of Integrative Plant Biology2020，62，8： | 2 |
| 2 | Duodenal intraepithelial T lymphocytes in patients with functional dyspepsia显示文摘AIM: To quantify the intraepithelial lymphocytes (IELs) and to document the membrane expression of CD4, CD8, TCRγδ and adhesion and/or activation-associated molecules (CD103, CD28, CD44, CD69, HLA-DR, CD95/ Fas) in the duodenal mucosa of patients with functional dyspepsia (FD) in order to provide arguments for an immunological process in FD. METHODS: Twenty-six FD patients according to Rome Ⅱ criteria (20 were H pylori negative) were studied and compared to 12 healthy adults. IELs were isolated from five duodenal biopsy samples, then quantified by microscopy and flow cytometry while the membrane phenotypes were determined by cytofluorometry. RESULTS: Duodenal histological examination was normal. In H pylori negative patients, the number of IELs was not different from that in healthy controls. Median percentage expression of CD4, CD8, or TCRγδ and CD103, CD44, CD28, CD69 on CD3+ IELs, among the adhesion/activation associated molecules tested, was not different from that in healthy controls. In contrast, the median percentage expression of CD95/ Fas [22 (9-65) vs 45 (19-88), P = 0.03] and HLA- DR expressing CD3+ IELs [4 (0-30) vs 13 (4-42), P = 0.04] was significantly lower in the H pylori negative FD group than in healthy controls, respectively. The number of IELs was significantly greater in H pylori positive FD patients than in healthy controls [median ratiofor 100 enterocytes 27.5 (6.7-62.5) vs 10.8 (3-33.3), P = 0.02] due to a higher number of CD8+ CD3+ IELs. | 13 | Simple calculator to estimate the medical cost of diabetes in sub-Saharan Africa显示文摘AIM: To design a medical cost calculator and show that diabetes care is beyond reach of the majority particularlypatients with complications.METHODS: Out-of-pocket expenditures of patients for medical treatment of type-2 diabetes were estimated based on price data collected in Benin,Burkina Faso,Guinea and Mali. A detailed protocol for realistic medical care of diabetes and its complications in the African context was defined. Care components were based on existing guidelines,published data and clinical experience. Prices were obtained in public and private health facilities. The cost calculator used Excel. The cost for basic management of uncomplicated diabetes was calculated per person and per year. Incremental costs were also computed per annum for chronic complications and per episode for acute complications. RESULTS: Wide variations of estimated care costs were observed among countries and between the public and private healthcare system. The minimum estimated cost for the treatment of uncomplicated diabetes(in the public sector) would amount to 21%-34% of the country's gross national income per capita,26%-47% in the presence of retinopathy,and above 70% for nephropathy,the most expensive complication. CONCLUSION: The study provided objective evidence for the exorbitant medical cost of diabetes considering that no medical insurance is available in the study countries.
