| 1 | Hepatocellular Carcinoma-Cause,Treatment and Metastasis显示文摘In the recent decades, the incidence of hepatocellular carcinoma (HCC) has been found to be increasing in males in some countries. In China, HCC ranked second of cancer mortality since 1990s. Hepatitis B and C viruses (HBV and HCV) and dietary aflatoxin intake remain the major causative factors of HCC. Surgery plays a major role in the treatment of HCC, particularly for small HCC. Downstaging unresectable huge HCC to smaller HCC and followed by resection will probably be a new approach for further study. Liver transplantation is indicated for small HCC, however, some issues remain to be solved.Different modes of 'regional cancer therapy for HCC' have been tried. Systemic chemotherapy has been disappointing in the past but the future can be promising.Biotherapy, such as cytokines, differentiation inducers,anti-angiogenic agents, gene therapy and tumor vaccine will probably play a role, particularly in the prevention of tumor recurrence. HCC invasiveness is currently the major target of study. | 2 | Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. Boyden chamber in vitro invasion assay demonstrated that the number of penetrating cells through the artificial basement membrane was 137.5 - 11 .0) cellsfield for MHC_C99--H vs 17.7 - 6.3) field for MHCC97-L.The proportions of cells in GO Gl phase. | 5 | Clinical observation of salvianolic acid B in treatment of liver fibrosis in chronic hepatitis B显示文摘AIM: To evaluate the clinical efficacy of salvianolic acidB (SA-B) on liver fibrosis in chronic hepatitis B.METHODS: Sixty patients with definite diagnosis of liverfibrosis with hepatitis B were included in the trial.Interferon-γ (IFN-γ) was used as control drug. Thepatients took orally SA-B tablets or received muscularinjection of IFN-γ in the double blind randomized test,The complete course lasted 6 months. The histologicalchanges of liver biopsy specimen before and after thetreatment were the main evidence in evaluation, incombination with the results of contents of serum HA,LN, Ⅳ-C, P-Ⅲ-P, liver ultrasound imaging, andsymptoms and signs.RESULTS: Reverse rate of fibrotic stage was 36.67 % inSA-B group and 30.0 % in IFN-γgroup. Inflammatoryalleviating rate was 40.0 % in SA-B group and 36.67 %in IFN-γ group. The average content of HA and Ⅳ-Cwas significantly lower than that before treatment. Theabnormal rate also decreased remarkably. Overallanalysis of 4 serological fibrotic markers showedsignificant improvement in SA-B group as comparedwith the IFN-γgroup. Score of liver ultrasound imagingwas lower in SA-B group than in IFN-γgroup (HA 36.7 %vs80 %,Ⅳ-C 3.3 % vs23.2 %). Before the treatment,ALT AST activity and total bilirubin content of patientswho had regression of fibrosis after oral administrationof SA-B, were significantly lower than those of patientswho had aggravation of fibrosis after oraladministration of SA-B. IFN-γ showed certain sideeffects (fever and transient decrease of leukocytes,occurrence rates were 50 % and 3.23 %), but SA-Bshowed no side effects.CONCLUSION: SA-B could effectively reverse liverfibrosis in chronic hepatitis B. SA-B was better than IFN-γ in reduction of serum HA content, overall decrease of4 serum fibrotic markers, and decrease of ultrasoundimaging score. Liver fibrosis in chronic hepatitis B withslight liver injury was more suitable to SA-B in anti-fibrotic treatment. | 6 | Mesozoic subduction-accretion zone in northeastern South China Sea inferred from geophysical interpretations显示文摘A segment of Mesozoic subduction-accretion zone was inferred across the northeastern South China Sea at approximately NE45° orientation. Basic evidence includes the following: A belt of peek gross horizontal Bouguer gravity gradient (PGHGBA) is comparable in size and intensity to that of the Manila subduction-accretion zone. A belt of high positive magnetic anomalies appears to the north and sub-parallel to the PGHGBA, representing the volcanic arc associated to the subduction zone. The PGHGBA crosses obliquely both Cenozoic structures and present seafloor topography, indicating a pre-Cenozoic age. The segment is offset left-laterally by NW-running strike-slip faults, in concord with the Mesozoic stress field of South China. In addition, the existence of the subduction zone is supported by wide-angle seismic data obtained in different years by different institutions. At approximate localities, a north-dipping ramp of Moho surface is indicated by records of ocean-bottom seismometers, and a strong reflector about 8 km beneath the Moho reflector is indicated by both OBS and long-cable seismic records. | 7 | Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. | 9 | Dynamic changes of typeⅠ,Ⅲand N collagen synthesis and distribution of collagen-producing cells in carbon tetrachloride-induced rat liver fibrosis显示文摘AIM To find out the relationship between the gene transcription of different types ofprocollagen and the deposition of the relevant collagens in the liver tissue and to confirm the types of collagen producing cells in liverfibrogenesis.METHODS Dynamic changes of the expression of α1(Ⅰ ), α1 (Ⅲ ) and α1 (Ⅳ) procollagen mRNAand relevant collagens and the distribution ofcollagen producing cells during liver fibrogenesis of rat induced by CCl4 (20 weeks)were investigated with Northern blot analysis,in situ hybridization and immunohistochemicaltechniques.RESULTS The increased expression of α1 (Ⅲ)procollagen mRNA by Northern blot analysis was the most predominant one among the threemRNAs during fibrogenesis. However, theenhanced expression of al (Ⅳ) procollagenmRNA occurred very early while the expressionof α1 (Ⅰ) mRNA was not enhanced much until themiddle stage of the exPeriment. D6smin (Dm)positive hepatic stellate cells (HSCs) and fewmyofibroblasts (MFs) in and around the necrotic areas expressed α1 (Ⅰ), α1 (Ⅲ) and α1 (Ⅳ)procollagen mRNA signals detected by in situhybridization at the early stage of theexperiment. All the three procollagen mRNAsignals thereafter mainly localized in fibroblasts (Fbs) and MFs in fibrotic septa during the middle and late stages of fibrosis, which distributedparallel to the corresponding collagens detected by immunohistochemical study. ln addition, the endothelial cells of sinusoids and the small blood vessels within the septa also showed α1(Ⅳ) procollagen mRNA and type Ⅳ collagen expressionCONCLUSION It is considered that 'HSC-MF-Fb' effect cell system is the major cellularsource of collagen production in liver fibrosis, in which HSCs are collagen producing precursor cells in the early liver fibrogenesis, thereafter the synthesis of type Ⅰ, Ⅲ and Ⅳ collagens (Col Ⅰ, Col Ⅲ and Col Ⅳ) mainly derives fromMFs and Fbs, which play a very important role in the progress of liver fibrosis. | 11 | TRANSPORT OF THE SUSPENDED SEDIMENT IN THE CHANGJIANG ESTUARY显示文摘The Changjiang River is characterized by the enormous volume of runoff and the great amount of sediment load with remarkable seasonal variation. The annual runoff sometimes is respondent to the amount of sediment load, and sometimes not. The amount of monthly sediment load after the month of the maximum runoff is larger than those before the month. The sediment concentration and net quantity of sediment transport in the vicinity of the river mouth varies greatly in time between the ebb and flood, spring and neap, and dry seasons and flood seasons. The three bifurcations also have differences in concentration and net quantity in space. Even in the same bifurcation they have differences in and out of the sand bar. At present, the North Channel is the main passage for water and sediment load emptying into the sea from the Changjiang River. More than 50 percent of the sediments from the river basin are deposited nearby the South Branch entrance and the main depositional area is situated in subaqueous delta off the South Channel. | 12 | Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. | 16 | Mechanism of the Spring Persistent Rains over southeastern China显示文摘The Spring Persistent Rains (SPR) in the areas to the south of middle and lower reaches of the Yangtze River or over southeastern China (SEC) is a unique synoptic and climatic phenomenon in East Asia. This study reveals a possible mechanism responsible for the climatic cause of SPR formation through climatic mean data analysis and sensitive numerical model experiments. SEC is located at the down-stream of the southwesterly velocity center (SWVC) which lies on the southeastern flank of the Tibetan Plateau (TP). As a result, there are strong southwesterly wind velocity convergence and moisture con-vergence over SEC. This is the immediate climatic cause of SPR formation. In spring, the seasonal evolution of the southwesterly velocity consists with the surface sensible heating over southeastern TP, indicating that the formation of SPR is related to not only the southwesterly wind of mechanical de-flected flow of TP, but also the southwesterly wind of thermal-forced cyclonic low circulation. Sensitive numerical experiments demonstrate that, without TP, both SWVC and the SPR rain belt will disappear. The southwesterly wind velocity increases almost linearly with the amount of the total diabatic heating with TP rising. Therefore, SWVC is the result of the mechanical forcing and thermal forcing of TP. | 17 | Contemporary crustal movement of continental China obtained by global positioning system(GPS) measurements显示文摘We obtain a unified horizontal velocity field of continental China and vicinity through measurements at 81 GPS stations provided by the National Key Infrastructure Project, Crustal Movement Observation Network of China. The velocity field delineates patterns of movements and deformation of active crustal blocks in continental China under the Eurasia-fixed reference frame. The space-based geodesy also clearly shows for the first time the horizontal movementand deformation of continental China induced by indentation of the Indian plate. | 18 | Significance of cyclooxygenase-2 expression in human primary hepatocellular carcinoma显示文摘AIM: To clarify the significance of cyclooxygenase-2 (COX-2) expression in human primary hepatocellular carcinoma(HCC) and adjacent nontumorous tissues.METHODS: The COX-2 protein and mRNA were investigatedin 27 HCC tissues with adjacent nontumorous tissues, and 5histologically normal liver tissues, using immunohistochemistryand in situ hybridization.RESULTS: The well-differentiated HCC expressed COX-2protein (5.68±1.19) more strongly than moderated HCC(3.43± 1.98) and poor differentiated HCC (3.33± 1.50)(P<0.05 respectively), adjacent nontumorous tissues (4.93±1.05) and normal liver tissues (3.20±1.g2) (P<0.01respectively); More intensive staining of COX-2 in adjacentnontumorous tissues was observed than that in normal livertissues (P<0.05). There was no significant difference amongadjacent nontumorous tissues, moderately differentiated HCCand poorly differentiated HCC (P>0.05). | 19 | Sorafenib in treatment of patients with advanced hepatocellular carcinoma:a systematic review显示文摘BACKGROUND: Sorafenib has become the standard first-line treatment for patients with advanced hepatocellular carcinoma (HCC). This study aimed to assess the efficacy and safety of sorafenib in advanced HCC patients and explore its true value for specific subgroups. DATA SOURCES: A computer-based systematic search from January 2005 to June 2011 with 'sorafenib' and 'advanced hepatocellular carcinoma' as search terms was performed for possible clinical trials. Hazard ratios (HR) and their 95% confidence intervals (CI) for overall survival (OS) and time to progression (TTP), rates of partial response (PR), rates of toxicity effects, and details of subgroup analysis were extracted. Meta-analyses were done using the software Review Manager (version 5.0). RESULTS: Six trials with 1164 patients were included. Based on three randomized controlled trials, the pooled HR (sorafenib/ placebo) was 0.66 for OS (95% CI: 0.56-0.78; P<0.00001) and 0.57 for TTP (95% CI: 0.47-0.68; P<0.00001). The pooled odds ratio (OR) for PR was 2.96 (95% CI: 0.96-9.15; P=0.06). For three single-arm trials, the pooled HR was 0.69 for OS (95% CI: 0.56-0.84; P=0.0002) and 0.64 for TTP (95% CI: 0.52-0.78; P<0.00001). The pooled OR for PR in three single-arm trials was 3.56 (95% CI: 1.22-10.39; P=0.02). Subgroup analysis indicated that sorafenib was less effective in patients with extrahepatic spread (with: P=0.13 vs without: P<0.0001), with normal alpha-fetoprotein level (AFP) (P=0.15 vs elevated: P=0.0006), and with elevated level of serum bilirubin (P=0.06 vs normal: P=0.0009). Sorafenib-based therapy significantly increased the risk of grade 3/4 hand-foot skin reaction, diarrhea, fatigue, and rash/desquamation.CONCLUSIONS: Sorafenib-based therapy benefits advanced HCC patients.
