1Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32

3Signaling pathways involved in ischemic stroke:molecular mechanisms and therapeutic interventions显示文摘Ischemic stroke is caused primarily by an interruption in cerebral blood flow,which induces severe neural injuries,and is one of the leading causes of death and disability worldwide.Thus,it is of great necessity to further detailly elucidate the mechanisms of ischemic stroke and find out new therapies against the disease.In recent years,efforts have been made to understand the pathophysiology of ischemic stroke,including cellular excitotoxicity,oxidative stress,cell death processes,and neuroinflammation.In the meantime,a plethora of signaling pathways,either detrimental or neuroprotective,are also highly involved in the forementioned pathophysiology.These pathways are closely intertwined and form a complex signaling network.Also,these signaling pathways reveal therapeutic potential,as targeting these signaling pathways could possibly serve as therapeutic approaches against ischemic stroke.In this review,we describe the signaling pathways involved in ischemic stroke and categorize them based on the pathophysiological processes they participate in.Therapeutic approaches targeting these signaling pathways,which are associated with the pathophysiology mentioned above,are also discussed.Meanwhile,clinical trials regarding ischemic stroke,which potentially target the pathophysiology and the signaling pathways involved,are summarized in details.Conclusively,this review elucidated potential molecular mechanisms and related signaling pathways underlying ischemic stroke,and summarize the therapeutic approaches targeted various pathophysiology,with particular reference to clinical trials and future prospects for treating ischemic stroke.Chuan Qin Sheng Yang Yun-Hui Chu Hang Zhang Xiao-Wei Pang Lian Chen Luo-Qi Zhou Man Chen Dai-Shi Tian Wei Wang 2022Signal Transduction and Targeted Therapy2022,7,8:7
4Parallel difference schemes with interface extrapolation terms for quasi-linear parabolic systems显示文摘In this paper some new parallel difference schemes with interface extrapolation terms for a quasi-linear parabolic system of equations are constructed. Two types of time extrapolations are proposed to give the interface values on the interface of sub-domains or the values adjacent to the interface points, so that the unconditional stable parallel schemes with the second accuracy are formed. Without assuming heuristically that the original boundary value problem has the unique smooth vector solution, the existence and uniqueness of the discrete vector solutions of the parallel difference schemes constructed are proved. Moreover the unconditional stability of the parallel difference schemes is justified in the sense of the continuous dependence of the discrete vector solution of the schemes on the discrete known data of the original problems in the discrete W2(2,1) (Q△) norms. Finally the convergence of the discrete vector solutions of the parallel difference schemes with interface extrapolation terms to the unique generalized solution of the original quasi-linear parabolic problem is proved. 6Simultaneous determination of indapamide,perindopril and perindoprilat in human plasma or whole blood by UPLC-MS/MS and its pharmacokinetic application显示文摘Simple and sensitive methods were developed for the determination of indapamide, perindopril and its active metabolite perindoprilat in human plasma or whole blood by hyphenated ultra-performance liquid chromatography-mass spectrometry(UPLC-MS/MS). Indapamide-d3, perindopril-d4 and perindoprilat-d4 were used as the internal standards. The separation was performed on a Thermo BDS Hypersil C_(18) column(4.6 mm×100 mm, 2.4 mm) for indapamide and perindopril simultaneously following a protein precipitation pretreatment of the biosamples. The separation of perindoprilat was achieved independently on a phenomenex PFP column(4.6 mm×150 mm, 5 mm). All the analytes were quantitated with positive electrospray ionization and multiple reactions monitoring mode. The assay exhibited a linear range of 1–250 ng/mL for indapamide, 0.4–100 ng/mL for perindopril and 0.2–20 ng/mL for perindoprilat. 8Room-temperature-densified H_(3)BO_(3) microwave dielectric ceramics with ultra-low permittivity and ultra-high Qf value显示文摘With the rapid development of mobile communication technology towards 5G and 6G,the microwave dielectric materials with ultra-low permittivity and ultra-high Qf value are urgently demanded.Here,the excellent microwave dielectric properties are reported in H3BO3 ceramics with the molecular crystal structure,whose permittivity(2.84)and density(1.46 g/cm^(3))are record-low among the low-loss ceramics.The ultra-high Qf value of 146,000 GHz(or the ultra-low dielectric loss of 1.03×10^(-4) at 15 GHz)is also distinguished.Besides,the H_(3)BO_(3) ceramics can be densified at room temperature by a simple cold sintering process in a short time of 10 min,and this brings many advantages for the integration with microwave circuits.The large molecule volume originating from the molecular crystal structure and the low dielectric polarizabilities of H^(+) and B^(3+) are responsible for the ultra-low permittivity of H_(3)BO_(3) ceramics,and more microwave dielectric materials with ultra-low permittivity and ultra-high Qf value are expected to be explored in the molecular crystals.Wen Bin Hong Lei Li Han Yan Shu Ya Wu Hang Sheng Yang Xiang Ming Chen 2020Journal of Materiomics2020,6,2:3
9Segmental ureteroileal conduit resection for the treatment of distal upper urinary tract recurrence of bladder cancer fol lowing cystectomy显示文摘Segmental ureterectomy is less invasive than radical nephroureterectomy and results in nephron preservation and satisfactory tumor control.This study was to determine the feasibility of segmental ureteroileal conduit resection(SUICR) for patients with distal upper urinary tract recurrence of bladder cancer following radical cystectomy.Four patients with high-grade distal upper urinary tract recurrence underwent SUICR 15-108 months after radical cystectomy.The surgical technique details of SUICR,operative results,and follow-up outcomes are reported.The median operation time was 280 min,and estimated blood loss was less than 100 ml One patient suffered from ileus 5 days after surgery and was managed conservatively.Histopathologic evaluation showed high-grade stages pTa-pT1 diseases for these patients,and ureteral margins were all negative.No patient suffered from tumor recurrence,with a median follow-up of 39 months.SUICR preserved the ipsilateral renal unit and conformed to oncological principles during surgery.The oncological outcome was satisfactory for these properly selected patients.This technique provides a valid alternative to nephroureterectomy for patients with imperative indications and high-grade upper urinary tract recurrence of bladder cancer following radical cystectomy.Shu‑Xiong Zeng Xin Lu Wei‑Dong Xu Zhen‑Sheng Zhang Hai‑Hang Li Ying‑Hao Sun Chuan‑Liang Xu 2016Chinese Journal of Cancer2016,35,3:3

14Immunogenicity and Efficacy of Liposome-encapsulated Influenza Split Vaccine in BALB/c Mice显示文摘The cellular immunity of current influenza split vaccine is relatively low. It is necessary to develop a novel vaccine to improve the cellular immunity. Thes of this study prepared liposome-encapsulated influenza split vaccine and tested it in BALB/c mice. The mice were immunized once with 4 μg of haemagglutinin of monovalent A/New Caledonia/20/99(H1N1) encapsulated with liposomes or the split virus vaccine only through intrastomach injection. 17Multiple Effects Tailoring the Self-organization Behaviors of Triphenylene Side-chain Liquid Crystalline Polymers via Changing the Spacer Length显示文摘Long-alkyl tail triphenylene(TP) side-chain liquid crystalline polymers(SCLCPs) with different spacer length(P-m-TP,m =2,3,4,6,8,which is the number of carbon atom in the flexible alkyl spacers) have been successfully synthesized via free radical polymerization.The differential scanning calorimetry(DSC),polarized light microscopy(POM),ultraviolet-visible spectroscopy(UVVis),wide-angle X-ray diffraction(WAXD) and small-angle X-ray scattering(SAXS) measurements were performed to investigate the influence of multiple effects on the self-organization behaviors of P-m-TP,including steric effect,decoupling effect and π-π stacking effect.The experimental results revealed that P-m-TP(m = 2,3,4) formed the columnar phase which was developed by the TP moieties and the main chain as a whole,suggesting that the side-chains had strong steric effect even though the number of spacer length(m)exceeded 4.In addition,the clearing points(Tis) of the polymers were above 300 °C.When m = 6 and 8,the polymers displayed hexagonal columnar phase and exhibited the low Tis(91 and 80 °C respectively),originating from the self-assembly of triphenylene due to the decoupling effect and π-π stacking effect.This work offers a viable and inspiring pathway to control the phase transition temperature and phase structure of TP SCLCPs via simply tailoring the spacer length and increasing the alkyl tail length of TP.Xiang-Hui Han Xi-Wen Yang Sheng Chen Hang Luo Dou Zhang Hai-Liang Zhang 2018Chinese Journal of Polymer Science2018,36,8:1
18Challenges and opportunities in remote sensing-based crop monitoring:a review显示文摘Building a more resilient food system for sustainable development and reducing uncertainty in global food markets both require concurrent and near-real-time and reliable crop information for decision making.Satellite-driven crop monitoring has become a main method to derive crop information at local,regional,and global scales by revealing the spatial and temporal dimensions of crop growth status and production.However,there is a lack of quantitative,objective,and robust methods to ensure the reliability of crop information,which reduces the applicability of crop monitoring and leads to uncertain and undesirable consequences.In this paper,we review recent progress in crop monitoring and identify the challenges and opportunities in future efforts.We find that satellite-derived metrics do not fully capture determinants of crop production and do not quantitatively interpret crop growth status;the later can be advanced by integrating effective satellite-derived metrics and new onboard sensors.We have identified that ground data accessibility and the negative effects of knowledge-based analyses are two essential issues in crop monitoring that reduce the applicability of crop monitoring for decisions on food security.Crowdsourcing is one solution to overcome the restrictions of ground-truth data accessibility.We argue that user participation in the complete process of crop monitoring could improve the reliability of crop information.Encouraging users to obtain crop information from multiple sources could prevent unconscious biases.Finally,there is a need to avoid conflicts of interest in publishing publicly available crop information.Bingfang Wu Miao Zhang Hongwei Zeng Fuyou Tian Andries B Potgieter Xingli Qin Nana Yan Sheng Chang Yan Zhao Qinghan Dong Vijendra Boken Dmitry Plotnikov Huadong Guo Fangming Wu Hang Zhao Bart Deronde Laurent Tits Evgeny Loupian 2023National Science Review2023,10,4:1
