| 3 | Recovery of serum testosterone following neoadjuvant and adjuvant androgen deprivation therapy in men treated with prostate brachytherapy显示文摘AIM: To investigate the time course of testosterone(T) recovery after cessation of androgen deprivation therapy(ADT) in patients treated with brachytherapy. METHODS: One-hundred and seventy-four patients treated between June 1999 and February 2009 were studied. Patients were divided into a short-term usage group(≤ 12 mo, n = 91) and a long-term usage group(≥ 36 mo, n = 83) according to the duration of gonadotropin-releasing hormone agonist therapy. Median follow-up was 29 mo in the short-term group and was 60 mo in the long-term group.RESULTS: Cumulative incidence rates of T recovery to normal and supracastrate levels at 24 mo after cessationwere 28.8% and 74.6%, respectively, in the long-term usage group, whereas these values were 96.4% and 98.8% in the short-term usage group. T recovery to normal and supracastrate levels occurred significantly more rapidly in the short-term than in the long-term usage group(P < 0.001 and P < 0.001, respectively). Five years after cessation, 22.6% of patients maintained a castrate T level in the long-term usage group. On multivariate analysis, lower T levels(< 10 ng/d L) at cessation of ADT was significantly associated with prolonged T recovery to supracastrate levels in the longterm usage group(P = 0.002). CONCLUSION: Lower T levels at cessation of ADT were associated with prolonged T recovery in the longterm usage group. Five years after cessation of longterm ADT, approximately one-fifth of patients still had castrate T levels. | 4 | Bioprinted Vascularized Mature Adipose Tissue with Collagen Microfibers for Soft Tissue Regeneration显示文摘The development of soft tissue regeneration has recently gained importance due to safety concerns about artificial breast implants.Current autologous fat graft implantations can result in up to 90%of volume loss in long-term outcomes due to their limited revascularization.Adipose tissue has a highly vascularized structure which enables its proper homeostasis as well as its endocrine function.Mature adipocytes surrounded by a dense vascular network are the specific features required for efficient regeneration of the adipose tissue to perform host anastomosis after its implantation.Recently,bioprinting has been introduced as a promising solution to recreate in vitro this architecture in large-scale tissues.However,the in vitro induction of both the angiogenesis and adipogenesis differentiations from stem cells yields limited maturation states for these two pathways.To overcome these issues,we report a novel method for obtaining a fully vascularized adipose tissue reconstruction using supporting bath bioprinting.For the first time,directly isolated mature adipocytes encapsulated in a bioink containing physiological collagen microfibers(CMF)were bioprinted in a gellan gum supporting bath.These multilayered bioprinted tissues retained high viability even after 7 days of culture.Moreover,the functionality was also confirmed by the maintenance of fatty acid uptake from mature adipocytes.Therefore,this method of constructing fully functional adipose tissue regeneration holds promise for future clinical applications. | Fiona Louis Marie Piantino Hao Liu Dong-Hee Kang Yoshihiro Sowa Shiro Kitano Michiya Matsusaki | 2021 | Cyborg and Bionic Systems2021,,1: | 0 |
| 5 | Pyogenic liver abscess after choledochoduodenostomy for biliary obstruction caused by autoimmune pancreatitis显示文摘A 68-year-old man underwent cholecystectomy and choledochoduodenostomy for biliary obstruction and nephrectomy for a renal tumor. Based on clinical and histopathologic findings, autoimmune pancreatitis (AIP) was diagnosed. The renal tumor was diagnosed as a renal cell cancer. Steroid therapy was started and thereafter pancreatic inflammation improved. Five years after surgery, the patient was readmitted because of pyrexia in a preshock state. A Klebsiella pneumoniae liver abscess complicated by sepsis was diagnosed. The patient recovered with percutaneous abscess drainage and administration of intravenous antibiotics. Liver abscess recurred 1 mo later but was successfully treated with antibiotics. There has been little information on long-term outcomes of patients with AIP treated with surgery. | 6 | High-throughput drug screening models of mature adipose tissues which replicate the physiology of patients’Body Mass Index(BMI)显示文摘Obesity is a complex and incompletely understood disease,but current drug screening strategies mostly rely on immature in vitro adipose models which cannot recapitulate it properly.To address this issue,we developed a statistically validated high-throughput screening model by seeding human mature adipocytes from patients,encapsulated in physiological collagen microfibers.These drop tissues ensured the maintenance of adipocyte viability and functionality for controlling glucose and fatty acids uptake,as well as glycerol release.As such,patients’BMI and insulin sensitivity displayed a strong inverse correlation:the healthy adipocytes were associated with the highest insulin-induced glucose uptake,while insulin resistance was confirmed in the underweight and severely obese adipocytes.Insulin sensitivity recovery was possible with two type 2 diabetes treatments,rosiglitazone and melatonin.Finally,the addition of blood vasculature to the model seemed to more accurately recapitulate the in vivo physiology,with particular respect to leptin secretion metabolism. | Fiona Louis Yoshihiro Sowa Shiro Kitano Michiya Matsusaki | 2022 | Bioactive Materials2022,7,1: | 0 |
