1Cerebral fractional amplitude of low-frequency fluctuations may predict headache intensity improvement following acupuncture treatment in migraine patients显示文摘OBJECTIVE:To explore how baseline resting-state cerebral function predicts clinical outcomes of acupuncture treatment for migraine.METHODS:One hundred migraine patients and 46 healthy subjects were recruited.Patients were randomized into the acupuncture,sham acupuncture,and waiting list groups.Resting-state functional magnetic resonance imaging data were collected at baseline and after 1 month of longitudinal acupuncture treatments.Fractional amplitude of low-frequency fluctuations(f ALFF)calculations were applied to explore the associations between baseline f ALFF and changes in clinical variables in the acupuncture treatment group and the waiting list group.RESULTS:Compared with healthy subjects,migraine patients had lower f ALFF in the left rostral ventromedial medulla,right thalamus,left amygdala,and right angular gyrus.Regression analyses revealed that baseline f ALFF values in the left middle frontal gyrus,left superior temporal gyrus,and middle temporal gyrus were positively associated with headache intensity changes in the acupuncture treatment group,while baseline f ALFF values in the bilateral lingual gyrus and cuneus were negatively associated with headache intensity changes in this group.CONCLUSION:The baseline f ALFF values of brain regions associated with cognitive pain modulation,but not migraine severity,may predict future headache intensity improvement levels in migraine patients receiving acupuncture treatment.Li Zhengjie Zhou Jun Cheng Shirui Lan Lei Sun Ruirui Liu Mailan Yang Jie Gao Yujie Guo Taipin Gong Qiyong Zeng Fang Liang Fanrong 2020Journal of Traditional Chinese Medicine2020,40,6:5
2A Neural Circuit Mechanism Controlling Breathing by Leptin in the Nucleus Tractus Solitarii显示文摘Leptin,an adipocyte-derived peptide hormone,has been shown to facilitate breathing.However,the central sites and circuit mechanisms underlying the respiratory effects of leptin remain incompletely understood.The present study aimed to address whether neurons expressing leptin receptor b(LepRb)in the nucleus tractus solitarii(NTS)contribute to respiratory control.Both chemogenetic and optogenetic stimulation of LepRb-ex-pressing NTS(NTS^(LepRb))neurons notably activated breathing.Moreover,stimulation of NTS^(LepRb) neurons projecting to the lateral parabrachial nucleus(LPBN)not only remarkably increased basal ventilation to a level similar to that of the stimulation of all NTS^(LepRb) neurons,but also activated LPBN neurons projecting to the preBotzinger complex(preBotC).By contrast,ablation of NTS^pRb neurons projecting to the LPBN notably eliminated the enhanced respiratory effect induced by NTSLepRb neuron stimulation.In brainstem slices,bath application of leptin rapidly depolarized the membrane potential,increased the spontaneous firing rate,and accelerated the Ca2+transients in most NTSLepRb neurons.Therefore,leptin potentiates breathing in the NTS most likely via an NTS-LPBN-preBdtC circuit.Hongxiao Yu Luo Shi Jinting Chen Shirui Jun Yinchao Hao Shuang Wang Congrui Fu Xiang Zhang Haiyan Lu Sheng Wang Fang Yuan 2022Neuroscience Bulletin2022,38,2:1
3Circuit-Specific Control of Blood Pressure by PNMT‑Expressing Nucleus Tractus Solitarii Neurons显示文摘The nucleus tractus solitarii(NTS)is one of the morphologically and functionally defined centers that engage in the autonomic regulation of cardiovascular activity.Phenotypically-characterized NTS neurons have been implicated in the differential regulation of blood pressure(BP).Here,we investigated whether phenylethanolamine N-methyltransferase(PNMT)-expressing NTS(NTS^(PNMT))neurons contribute to the control of BP.We demonstrate that photostimulation of NTS^(PNMT)neurons has variable effects on BP.A depressor response was produced during optogenetic stimulation of NTS^(PNMT)neurons projecting to the paraventricular nucleus of the hypothalamus,lateral parabrachial nucleus,and caudal ventrolateral medulla.Conversely,photostimulation of NTS^(PNMT)neurons projecting to the rostral ventrolateral medulla produced a robust pressor response and bradycardia.In addition,genetic ablation of both NTS^(PNMT)neurons and those projecting to the rostral ventrolateral medulla impaired the arterial baroreflex.Overall,we revealed the neuronal phenotype-and circuit-specific mechanisms underlying the contribution of NTS^(PNMT)neurons to the regulation of BP.Shirui Jun Xianhong Ou Luo Shi Hongxiao Yu Tianjiao Deng Jinting Chen Xiaojun Nie Yinchao Hao Yishuo Shi Wei Liu Yanming Tian Sheng Wang Fang Yuan 2023Neuroscience Bulletin2023,39,8:0

5Nutrition in Alzheimer's disease:a review of an underappreciated pathophysiological mechanism显示文摘Alzheimer's disease(AD)is the leading cause of dementia in older individuals and is an escalating challenge to global public health.Pharmacy therapy of AD is one of the well-funded areas;however,little progress has been made due to the complex pathogenesis.Recent evidence has demonstrated that modifying risk factors and lifestyle may prevent or delay the incidence of AD by 40%,which suggests that the management should pivot from single pharmacotherapy toward a multipronged approach because AD is a complex and multifaceted disease.Recently,the gut-microbiota-brain axis has gained tremendous traction in the pathogenesis of AD through bidirectional communication with multiple neural,immune,and metabolic pathways,providing new insights into novel therapeutic strategies.Dietary nutrition is an important and profound environmental factor that influences the composition and function of the microbiota.The Nutrition for Dementia Prevention Working Group recently found that dietary nutrition can affect cognition in AD-related dementia directly or indirectly through complex interactions of behavioral,genetic,systemic,and brain factors.Thus,considering the multiple etiologies of AD,nutrition represents a multidimensional factor that has a profound effect on AD onset and development.However,mechanistically,the effect of nutrition on AD is uncertain;therefore,optimal strategies or the timing of nutritional intervention to prevent or treat AD has not been established.Thus,this review summarizes the current state of knowledge concerning nutritional disorders,AD patient and caregiver burden,and the roles of nutrition in the pathophysiology of AD.We aim to emphasize knowledge gaps to provide direction for future research and to establish optimal nutrition-based intervention strategies for AD.Jiwei Jiang Hanping Shi Shirui Jiang Anxin Wang Xinying Zou Yanli Wang Wenyi Li Yuan Zhang Mengfan Sun Qiwei Ren Jun Xu 2023Science China(Life Sciences)2023,66,10:0
