1PRMT1 promotes the proliferation and metastasis of gastric cancer cells by recruiting MLXIP for the transcriptional activation of theβ-catenin pathway显示文摘Protein arginine methyltransferase 1(PRMT1),a type I PRMT,is overexpressed in gastric cancer(GC)cells.To elucidate the function of PRMT1 in GC,PRMT1 expression in HGC-27 and MKN-45 cells was knocked down by short hairpin RNA(shRNA)or inhibited by PRMT1 inhibitors(AMI-1 or DCLX069),which resulted in inhibition of GC cell proliferation,migration,invasion,and tumorigenesis in vitro and in vivo.MLX-interacting protein(MLXIP)and Kinectin 1(KTN1)were identified as PRMT1-binding proteins.PRMT1 recruited MLXIP to the promoter ofβ-catenin,which inducedβ-catenin transcription and activated theβ-catenin signaling pathway,promoting GC cell migration and metastasis.Furthermore,KTN1 inhibited the K48-linked ubiquitination of PRMT1 by decreasing the interaction between TRIM48 and PRMT1.Collectively,our findings reveal a mechanism by which PRMT1 promotes cell proliferation and metastasis mediated by theβ-catenin signaling pathway.Feng Wang Shitong Chen Shihan Peng Xujun Zhou Houyi Tang Hanghua Liang Xi Zhong He Yang Xiaoxue Ke MuHan Lü Hongjuan Cui 2023Genes & Diseases2023,10,6:0

3USP1-regulated reciprocal differentiation of Th17 cells and Treg cells by deubiquitinating and stabilizing TAZ显示文摘The balance between inflammatory T helper type 17(Th17)and immunosuppressive regulatory T(Treg)cells is critical for maintaining immune homeostasis in the human body and is tightly regulated under healthy conditions.An increasing number of studies have reported that deubiquitinases(DUBs)play a vital role in regulating Th17-and Treg-cell differentiation.However,the biological functions of only a small fraction of DUBs in Th17-and Treg-cell differentiation are well defined.In this study,we identified ubiquitin-specific peptidase 1(USP1)as a vital regulator of CD4+T-cell differentiation.USP1 promoted Th17-cell differentiation but attenuated Treg-cell differentiation,thereby promoting the development of inflammatory diseases.Mechanistically,USP1 in CD4+T cells enhanced the activity of RORγt but promoted the proteasomal degradation of Foxp3 through deubiquitination and stabilization of TAZ in vitro and in vivo.Notably,ML323,a specific inhibitor of the USP1/UAF1 deubiquitinase complex,inhibited Th17-cell differentiation and promoted Treg-cell differentiation in vitro and in vivo,indicating that ML323 might be a promising candidate for the treatment of diseases associated with an imbalance between Th17 and Treg cells.Our study highlights the critical role of USP1 in regulating adaptive immune responses and suggests that USP1 might be a drug target for the treatment of diseases associated with an imbalance between Th17 and Treg cells.Xiaotong Zhu Peng Wang Xiaoxia Zhan Yuping Zhang Junli Sheng Shitong He Yitian Chen Dingnai Nie Xiaolong You Haiyan Mai Qinghong Yu Laisheng Li Ligang Jie Shengfeng Hu 2023Cellular & Molecular Immunology2023,20,3:0
4Angle-uniform parallel coordinates显示文摘We present angle-uniform parallel coordinates,a data-independent technique that deforms the image plane of parallel coordinates so that the angles of linear relationships between two variables are linearly mapped along the horizontal axis of the parallel coordinates plot.Despite being a common method for visualizing multidimensional data,parallel coordinates are ineffective for revealing positive correlations since the associated parallel coordinates points of such structures may be located at infinity in the image plane and the asymmetric encoding of negative and positive correlations may lead to unreliable estimations.To address this issue,we introduce a transformation that bounds all points horizontally using an angleuniform mapping and shrinks them vertically in a structure-preserving fashion;polygonal lines become smooth curves and a symmetric representation of data correlations is achieved.We further propose a combined subsampling and density visualization approach to reduce visual clutter caused by overdrawing.Our method enables accurate visual pattern interpretation of data correlations,and its data-independent nature makes it applicable to all multidimensional datasets.The usefulness of our method is demonstrated using examples of synthetic and real-world datasets.Kaiyi Zhang Liang Zhou Lu Chen Shitong He Daniel Weiskopf Yunhai Wang 2023Computational Visual Media2023,9,3:0
