| 1 | Functional optoacoustic neuro-tomography for scalable whole-brain monitoring of calcium indicators显示文摘Non-invasive observation of spatiotemporal activity of large neural populations distributed over entire brains is a longstanding goal of neuroscience.We developed a volumetric multispectral optoacoustic tomography platform for imaging neural activation deep in scattering brains.It can record 100 volumetric frames per second across scalable fields of view ranging between 50 and 1000 mm^(3) with respective spatial resolution of 35–200μm.Experiments performed in immobilized and freely swimming larvae and in adult zebrafish brains expressing the genetically encoded calcium indicator GCaMP5G demonstrate,for the first time,the fundamental ability to directly track neural dynamics using optoacoustics while overcoming the longstanding penetration barrier of optical imaging in scattering brains.The newly developed platform thus offers unprecedented capabilities for functional whole-brain observations of fast calcium dynamics;in combination with optoacoustics'well-established capacity for resolving vascular hemodynamics,it could open new vistas in the study of neural activity and neurovascular coupling in health and disease. | X Luís Deán-Ben Gali Sela Antonella Lauri Moritz Kneipp Vasilis Ntziachristos Gil G Westmeyer Shy Shoham Daniel Razansky | 2016 | Light(Science & Applications)2016,5,1: | 5 |

| 3 | Correction-free remotely scanned two-photon in vivo mouse retinal imaging显示文摘Non-invasive fluorescence retinal imaging in small animals is an important requirement for an array of translational vision applications.The in vivo two-photon imaging of the mouse retina may enable the long-term investigation of the structure and function of healthy and diseased retinal tissue.However,to date,this has only been possible using relatively complex adaptive-optics systems.Here,the optical modeling of the murine eye and of the imaging system is used to achieve correction-free two-photon microscopy through the pupil of a mouse eye to yield high-quality,optically sectioned fundus images.By remotely scanning the focus using an electronically tunable lens,high-resolution three-dimensional fluorescein angiograms and cellular-scale images are acquired,thus introducing a correction-free baseline performance level for two-photon in vivo retinal imaging.Moreover,the system enables functional calcium imaging of repeated retinal responses to light stimulation using the genetically encoded indicator,GCaMP6s.These results and the simplicity of the new add-on optics are an important step toward several structural,functional,and multimodal imaging applications that will benefit from the tight optical sectioning and the use of near-infrared light. | Adi Schejter Bar-Noam Nairouz Farah Shy Shoham | 2016 | Light(Science & Applications)2016,5,1: | 3 |
| 4 | 小鼠海马神经网络对情景体验进行实时编码的功能单元的发现与鉴别显示文摘为了在神经网络层次上研究大脑对情景记忆信息实时编码的原理,我们创制了96通道微电极阵列系统.利用该系统,在小鼠经历惊吓事件过程中,对其大脑海马区多达260个神经元的放电活动进行了同步观察和记录.我们发现,惊吓情景刺激促发了小鼠海马CAI区神经元的放电模式发生改变,且这种改变与惊吓事件的特性及发生环境密切相关.运用统计学中的模式判别法。这些神经元的放电活动在低维空间形成了明显的集群式编码模式.进一步运用“移动窗口扫描”法,我们不仅能观察到神经网络实时编码的动态变化过程,而且能追踪惊吓事件之后记忆痕迹在神经元网络上的再现轨迹及次数.我们的分析表明:大脑海马CA1区神经网络的编码能力来自于由一系列神经元簇组成的功能性编码单元,神经元簇中的各神经元个体通过协同放电,从而克服了单一神经元放电的随机可变性,使对外界信息的实时编码获得稳定的高信噪比.这些功能性编码单元集群的激活形式能转换为具有实时性的一串串二进制数码,这一转换使不同动物大脑对各种行为事件获得了简明而通用的分类与编码. | 林龙年 Remus Osan Shy Shoham 金文军 左文琪 钱卓 梅兵 陈桂芬 | 2005 | 华东师范大学学报(自然科学版)2005,,Z1: | 2 |
