1Seasonal Variations in Phytoplankton Community Structure in the Sanggou,Ailian,and Lidao Bays显示文摘The seasonal variations in phytoplankton community structure were investigated for the Sanggou Bay(SGB) and the adjacent Ailian Bay(ALB) and Lidao Bay(LDB) in Shandong Peninsula, eastern China. The species composition and cell abundance of phytoplankton in the bay waters in spring(April 2011), summer(August 2011), autumn(October 2011), and winter(January 2012) were examined using the Utermhl method. A total of 80 taxa of phytoplankton that belong to 39 genera of 3 phyla were identified. These included 64 species of 30 genera in the Phylum Bacillariophyta, 13 species of 8 genera in the Phylum Dinophyta, and 3 species of 1 genus in the Phylum Chrysophyta. During the four seasons, the number of phytoplankton species(43) was the highest in spring, followed by summer and autumn(40), and the lowest number of phytoplankton species(35) was found in winter. Diatoms, especially Paralia sulcata(Ehrenberg) Cleve and Coscinodiscus oculus-iridis Ehrenberg, were predominant in the phytoplankton community throughout the study period, whereas the dominance of dinoflagellate appeared in summer only. The maximum cell abundance of phytoplankton was detected in summer(average 8.08 × 103 cells L-1) whereas their minimum abundance was found in autumn(average 2.60 × 103 cells L-1). The phytoplankton abundance was generally higher in the outer bay than in the inner bay in spring and autumn. In summer, the phytoplankton cells were mainly concentrated in the south of inner SGB, with peak abundance observed along the western coast. In winter, the distribution of phytoplankton cells showed 3 patches, with peak abundance along the western coast as well. On seasonal average, the Shannon-Wiener diversity indices of phytoplankton community ranged from 1.17 to 1.78(autumn > summer > spring > winter), and the Pielou's evenness indices of phytoplankton ranged from 0.45 to 0.65(autumn > spring > summer > winter). According to the results of canonical correspondence analysis, phosphate level was the major factor that limited the occurrence of P. sulcata and C. oculus-iridis, whereas optimal temperature and low salinity were responsible for Prorocentrum blooms in summer. 2Phytoplankton communities and its controlling factors in summer and autumn in the southern Yellow Sea, China显示文摘The phytoplankton water samples were collected in two multidisciplinary investigations which were carried out during summer(June) and autumn(November to December) of 2011. Phytoplankton species composition and abundance data were accomplished by Uterm?hl method. The phytoplankton community was dominated by diatoms and dinoflagellates in the southern Yellow Sea(YS) in summer and autumn. In summer, Paralia sulcata and Prorocentrum dentatum were the predominated species, the cell abundance ranged from 0.074 to 107.733×103 cells/L with an average of 9.057×103 cells/L. Two phytoplankton high abundance appeared in northwest part of the survey area and the Changjiang River Estuary, respectively. In autumn, Par. sulcata became the predominant species, and the phytoplankton cell abundance ranged from 1.035×103 to 8.985×103 cells/L, the average was 3.263×103 cells/L. The phytoplankton abundance in surface layer presented the homogeneous distributions. Canonical Correspondence Analysis(CCA) method was applied for discovering the relationship between environmental factors and the common found phytoplankton species. The responses of phytoplankton to nutrients were varied between summer and autumn. 3Spatial-temporal variation of phytoplankton community structure in Jiaozhou Bay, China显示文摘To better understand the spatial-temporal variation in phytoplankton community structure and its controlling factors in Jiaozhou Bay, Qingdao, North China, four seasonal sampling were carried out in 2017. The phytoplankton community structure and various environmental parameters were examined. The phytoplankton community in the bay was composed of mainly diatoms and dinofl agellates, and several other species of Chrysophyta were also observed. Diatoms were the most dominant phytoplankton group throughout the year, except in spring and winter, when Noctiluca scintillans was co-dominant. High Si/N ratios in summer and fall refl ect the high dominance of diatoms in the two seasons. Temporally, the phytoplankton cell abundance peaked in summer, due mainly to the high temperatures and nutrient concentrations in summer. Spatially, the phytoplankton cell abundance was higher in the northern part of the bay than in the other parts of the bay in four seasons. The diatom cell abundances show signifi cant positive correlations with the nutrient concentrations, while the dinofl agellate cell abundances show no correlation or a negative correlation with the nutrient concentrations but a signifi cant positive correlation with the stratifi cation index. This discrepancy was mainly due to the diff erent survival strategies between diatoms and dinofl agellates. The Shannon-Wiener diversity index ( H′) values in the bay ranged from 0.08 to 4.18, which fell in the range reported in historical studies. The distribution pattern of H′ values was quite diff erent from that of chlorophyll a , indicating that the phytoplankton community structure might have high biomass with a low diversity index. 5The ER membrane protein complex subunit Emc3 controls angiogenesis via the FZD4/WNT signaling axis显示文摘The endoplasmic reticulum(ER) membrane protein complex(EMC) regulates the synthesis and quality control of membrane proteins with multiple transmembrane domains. One of the membrane spanning subunits, EMC3, is a core member of the EMC complex that provides essential hydrophilic vestibule for substrate insertion. Here, we show that the EMC subunit Emc3 plays critical roles in the retinal vascular angiogenesis by regulating Norrin/Wnt signaling. Postnatal endothelial cell(EC)-specific deletion of Emc3 led to retarded retinal vascular development with a hyperpruned vascular network, the appearance of bluntended, aneurysm-like tip endothelial cells(ECs) with reduced numbers of filopodia and leakage of erythrocytes at the vascular front. Diminished tube formation and cell proliferation were also observed in EMC3 depleted human retinal endothelial cells(HRECs). We then discovered a critical role for EMC3 in expression of FZD4 receptor of β-catenin signaling using RNA sequencing, real-time quantitative PCR(RT-q PCR) and luciferase reporter assay. Moreover, augmentation of Wnt activity via lithium chloride(Li Cl) treatment remarkably enhanced β-catenin signaling and cell proliferation of HRECs. Additionally, Li Cl partially reversed the angiogenesis defects in Emc3-c KO mice. 7Relationship Between Zooplankton Community Characteristics and Environmental Conditions in the Surface Waters of the Western Pacific Ocean During the Winter of 2014显示文摘Understanding the effects of environmental heterogeneity on zooplankton communities has been a hot topic for several decades.However,relatively little is known about the responses of zooplankton communities to environmental conditions at large scales from inshore waters to the open ocean.Here,we used the abundance,biovolume,taxa and size spectra of zooplankton col-lected from the surface waters of the western Pacific Ocean during the winter of 2014 to study the relationship between zooplankton community characteristics and environmental conditions using multiple linear regression(MLR)analysis and redundancy analysis(RDA).According to a hierarchical cluster analysis based on hydrographic conditions,the study area was classified into five water masses.Significant correlations were identified between the limited nutrients and the zooplankton abundance and biovolume from inshore waters to the open ocean.Non-metric multidimensional scaling(NMDS)revealed two distinct zooplankton assemblages.In the northern inshore,Copepoda and Euphausiacea were the dominant zooplankton taxa and in other water masses,Chaetognatha and gelatinous zooplankton were the dominant zooplankton taxa in addition to Copepoda.Our results suggested that,on a large scale from inshore waters to the open ocean in the western Pacific Ocean,the spatial distribution of zooplankton taxa was mainly influ-enced by environmental conditions,while in the inshore waters,it was due to the top-down effect of the dominant zooplankton taxa.Finally,the slope of the normalized biovolume size spectra(NBSS)was negatively correlated with chlorophyll-a(Chl-a)concentra-tion and PO_(4)^(3−)-P concentration in the inshore waters,which indicated that the higher the trophic level the dominant zooplankton taxa were,the steeper the NBSS slope was.TIAN Ziyang ZHENG Shan GUO Shujin ZHU Mingliang LIANG Junhua DU Juan SUN Xiaoxia 2021Journal of Ocean University of China2021,20,3:1
8Concentrations and sinking rates of transparent exopolymer particles(TEPs)in a coastal sea:the Changjiang River(Yangtze River)Estuary显示文摘Transparent exopolymer particles(TEPs)are ubiquitous throughout the oceans,and their sedimentation is considered an efficient biological carbon sink pathway.To investigate the role of coastal TEPs in sinking carbon from the upper layer,samples were collected in the spring and summer of 2011 in the Changjiang River(Yangtze River)Estuary,a typical coastal water.The concentrations and sinking rates of TEPs were measured,and potential sedimentation flux of TEPs was estimated.TEPs concentrations ranged from 40.00μg/L to 1040.00μg/L(mean=(209.70±240.93)μg/L)in spring and 56.67μg/L to 1423.33μg/L(mean=(433.33±393.02)μg/L)in summer,and they were higher at bloom stations than at non-bloom stations during both cruises.A significant positive correlation between TEPs concentration and chlorophyll a(Chl a)concentration was detected,suggesting that phytoplankton was the primary source of TEPs in this area.TEPs sinking rates ranged from 0.08 m/d to 0.57 m/d with a mean of(0.28±0.14)m/d in spring and 0.10 m/d to 1.08 m/d with a mean of(0.34±0.31)m/d in summer.The potential sedimentation flux of TEP-C ranged from 4.95 mg/(m2·d)to 29.40 mg/(m2·d)with a mean of(14.66±8.83)mg/(m2·d)in spring and 6.80 mg/(m2·d)to 30.45 mg/(m2·d)with a mean of(15.71±8.73)mg/(m2·d)in summer,which was^17.81%to 138.27%(mean=65.15%±31.75%)of sedimentation flux of phytoplankton cells in the study area.Due to the increase of TEPs concentrations and their sinking rates,sedimentation fluxes of TEPs at the bloom station were obviously higher than at the non-bloom station during both cruises.This study indicates that TEPs serve as a carbon sink in the Changjiang River Estuary,especially during bloom events,and their sedimentation should be taken into account when we study the carbon sedimentation in the coastal sea.Shujin Guo Jun Sun 2020Acta Oceanologica Sinica2020,39,10:1

11Single-cell transcriptomic analysis reveals the developmental trajectory and transcriptional regulatory networks of pigment glands in Gossypium bicki显示文摘Comprehensive utilization of cottonseeds is limited by the presence of pigment glands and its inclusion gossypol.The ideal cotton has glandless seeds but a glanded plant,a trait found in only a few Australian wild cotton species,including Gossypium bickii.Introgression of this trait into cultivated species has proved to be difficult.Understanding the biological processes toward pigment gland morphogenesis and the associated underlying molecular mechanisms will facilitate breeding of cultivated cotton varieties with the trait of glandless seeds and glanded plant.In this study,single-cell RNA sequencing(scRNA-seg)was performed on 12222 protoplasts isolated from cotyledons of germinating G.bicki seeds 48 h after imbibition.Clustered into 14 distinct clusters unsupervisedly,these cells could be grouped into eight cell populations with the assistance of known cell marker genes.The pigment gland cells were well separated from others and could be separated into pigment gland parenchyma cells,secretory cells,and apoptotic cells.By integrating the pigment gland cell developmental trajectory,transcription factor regulatory networks,and core transcription factor functional validation,we established a model for pigment gland formation.In this model,light and gibberellin were verified to promote the formation of pigment glands.In addition,three novel genes,GbiERF114(ETHYLENE RESPONSE FACTOR 114),GbiZAT11(ZINC FINGER OF ARABIDOPSIS THALIANA 11),and GbiNTL9(NAC TRANSCRIPTION FACTOR-LIKE 9),Were found to affect pigment gland formation.Collectively,these findings provide new insights into pigment gland morphogenesis and lay the cornerstone for future cotton scRNA-seq investigations.Yue Sun Yifei Han Kuang Sheng Ping Yang Yuefen Cao Huazu Li Qian-Hao Zhu Jinhong Chen Shujin Zhu Tianlun Zhao 2023Molecular Plant2023,16,4:0

13Carbon biomass,carbon-to-chlorophyll a ratio and the growth rate of phytoplankton in Jiaozhou Bay,China显示文摘Carbon biomass,carbon-to-chlorophyll a ratio(C꞉Chl a),and the growth rate of phytoplankton cells were studied during four seasonal cruises in 2017 and 2018 in Jiaozhou Bay,China.Water samples were collected from 12 stations,and phytoplankton carbon biomass(phyto-C)was estimated from microscope-measured cell volumes.The phyto-C ranged from 5.05 to 78.52μg C/L in the bay,and it constituted a mean of 38.16% of the total particulate organic carbon in the bay.High phyto-C values appeared mostly in the northern or northeastern bay.Diatom carbon was predominant during all four cruises.Dinoflagellate carbon contributed much less(<30%)to the total phyto-C,and high values appeared often in the outer bay.The C꞉Chl a of phytoplankton cells varied from 11.50 to 61.45(mean 31.66),and high values appeared in the outer bay during all four seasons.The phyto-C was also used to calculate the intrinsic growth rates of phytoplankton cells in the bay,and phytoplankton growth rates ranged from 0.56 to 1.96/d;the rate was highest in summer(mean 1.79/d),followed by that in fall(mean 1.24/d)and spring(mean 1.17/d),and the rate was lowest in winter(mean 0.77/d).Temperature and silicate concentration were found to be the determining factors of phytoplankton growth rates in the bay.To our knowledge,this study is the first report on phytoplankton carbon biomass and C꞉Chl a based on water samples in Jiaozhou Bay,and it will provide useful information for studies on carbon-based food web calculations and carbon-based ecosystem models in the bay.Shujin GUO Zengxia ZHAO Junhua LIANG Juan DU Xiaoxia SUN 2021Journal of Oceanology and Limnology2021,39,4:0

17Selectivity of biopolymer membranes using HepG2 cells显示文摘Bioartificial liver(BAL)system has emerged as an alternative treatment to bridge acute liver failure to either liver transplantation or liver regeneration.One of the main reasons that the efficacy of the current BAL systems was not convincing in clinical trials is attributed to the lack of friendly interface between the membrane and the hepatocytes in liver bioreactor,the core unit of BAL system.Here,we systematically compared the biological responses of hepatosarcoma HepG2 cells seeded on eight,commercially available biocompatible membranes made of acetyl cellulose–nitrocellulose mixed cellulose(CA–NC),acetyl cellulose(CA),nylon(JN),polypropylene(PP),nitrocellulose(NC),polyvinylidene fluoride(PVDF),polycarbonate(PC)and polytetrafluoroethylene(PTFE).Physicochemical analysis and mechanical tests indicated that CA,JN and PP membranes yield high adhesivity and reasonable compressive and/or tensile features with friendly surface topography for cell seeding.Cells prefer to adhere on CA,JN,PP or PTFE membranes with high proliferation rate in spheriod-like shape.Actin,albumin and cytokeratin 18 expressions are favorable for cells on CA or PP membrane,whereas protein filtration is consistent among all the eight membranes.These results further the understandings of cell growth,morphology and spreading,as well as protein filtration on distinct membranes in designing a liver bioreactor.Dongyuan Lü Yuxin Gao Chunhua Luo Shouqian Lü Qian Wang Xianghong Xu Shujin Sun Chengzhi Wang Mian Long 2015Regenerative Biomaterials2015,2,1:0
