1SARS-CoV-2 infection aggravates chronic comorbidities of cardiovascular diseases and diabetes in mice显示文摘Background:Cardiovascular diseases(CVDs)and diabetes mellitus(DM)are top two chronic comorbidities that increase the severity and mortality of COVID-19.However,how SARS-CoV-2 alters the progression of chronic diseases remain unclear.Methods:We used adenovirus to deliver h-ACE2 to lung to enable SARS-CoV-2 infection in mice.SARS-CoV-2’s impacts on pathogenesis of chronic diseases were studied through histopathological,virologic and molecular biology analysis.Results:Pre-existing CVDs resulted in viral invasion,ROS elevation and activation of apoptosis pathways contribute myocardial injury during SARS-CoV-2 infection.Viral infection increased fasting blood glucose and reduced insulin response in DM model.Bone mineral density decreased shortly after infection,which associated with impaired PI3K/AKT/mTOR signaling.Conclusion:We established mouse models mimicked the complex pathological symptoms of COVID-19 patients with chronic diseases.Pre-existing diseases could impair the inflammatory responses to SARS-CoV-2 infection,which further aggravated the pre-existing diseases.This work provided valuable information to better understand the interplay between the primary diseases and SARS-CoV-2 infection.Yuanwu Ma Dan Lu Linlin Bao Yajin Qu Jiangning Liu Xiaolong Qi Lei Yu Xu Zhang Feifei Qi Qi Lv Yunpeng Liu Xudong Shi Caixian Sun Jing Li Jie Wang Yunlin Han Kai Gao Wei Dong Ning Liu Shan Gao Jing Xue Qiang Wei Sidan Pan Hong Gao Lianfeng Zhang Chuan Qin 2021Animal Models and Experimental Medicine2021,4,1:3

3The protection of CoronaVac against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant in nude-hACE2 mice显示文摘Background:Immunocompromised individuals have an increased risk of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection and severe outcomes,but we pay less attention to these people.Athymic nude mice are a murine strain with a spontaneous deficiency of the Foxn1 gene,which can result in thymic degeneration or its absence,leading to immunosuppression and a decrease in the number of T cells,and are widely used in preclinical evaluations of disease in immunocompromised populations.Methods:We investigated the protection of the coronavirus disease 2019(COVID-19)inactivated vaccine(CoronaVac)against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant utilizing a hybrid-type nude-hACE2 mouse model.Results:Compared with nude-hACE2/W mice,the viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/WV)infected with WH-09 after vaccination significantly decreased,and the histopathological changes were also reduced.The viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/OV)infected with the Omicron variant after vaccination was lower than that in nude-hACE2/O,but histopathological symptoms did not improve significantly.Conclusion:CoronaVac provides some protection against infection of both WH-09 and the Omicron variant in the nude-hACE2 mice.Our findings aimed to provide a reference for vaccination against SARS-CoV-2 in immunocompromised populations.Kaili Lin Meixuan Liu Lu Sun Hanjun Fu Hongwei Qiao Shunyi Wang Sidan Pan Hong Gao 2023Animal Models and Experimental Medicine2023,6,4:0
4Carbon budget and balance critical processes of the regional landwater-air interface:Indicating the earth system’s carbon neutrality显示文摘Regional processes on land-water-air interface carbon(C)budget and balance that interconnect the land and sea are in fact C neutrality critical processes within the earth system.It is therefore essential to quantitatively analyze synergistic landwater-air interface C transport and C exchange processes to gain a deep understanding of the important role that terrestrial ecosystems play in“missing C sink”.This paper systematically analyzes global land-water-air interface C migration processes as well as C budget changes and associated impact mechanisms.It also investigates the important role that these C budgets and C cycling processes play in inland water bodies,respective to C budgets at the land-water-air interface.Moreover,this study reveals the regulatory mechanisms of land-water-air interface C budgets and balances under a background of global climate change.It also quantitatively evaluates the status of China’s regional land-sea-air interface C budgets as well as the C sequestration potential of its lake systems.Finally,this study concludes that the explicit quantification of anthropogenic activity impacts on land-water-air interface C transport and exchange processes is of great significance to global C balances and C neutrality.Yang GAO Shuoyue WANG Yao LU Jianbao LIU Sidan LYU Kun SUN Junjie JIA Zhaoxi LI Guirui YU 2022Science China Earth Sciences2022,65,5:0
5An optical slot-antenna-coupled cavity (SAC) framework towards tunable free-space graphene photonic surfaces显示文摘The optical conductivity of single layer graphene (SLG) can be significantly and reversibly modified when the Fermi level is tuned by electrical gating. However, so far this interesting property has rarely been applied to free-space two-dimensional (2D) photonic devices because the surface-incident absolute absorption of SLG is limited to 1%–2%. No significant change in either reflectance or transmittance would be observed even if SLG is made transparent upon gating. To achieve significantly enhanced surface-incident optical absorption in SLG in a device structure that also allows gating, here we embed SLG in an optical slot-antenna-coupled cavity (SAC) framework, simultaneously enhancing SLG absorption by up to 20 times and potentially enabling electrical gating of SLG as a step towards tunable 2D photonic surfaces. This framework synergistically integrates near-field enhancement induced by ultrahigh refractive index semimetal slot-antenna with broadband resonances in visible and infrared regimes, ~ 3 times more effective than a vertical cavity structure alone. An example of this framework consists of self-assembled, close-packed Sn nanodots separated by ~ 10 nm nanogaps on a SLG/SiO2/Al stack, which dramatically increases SLG optical absorption to 10%-25% at λ = 600–1,900 nm. The enhanced SLG absorption spectrum can also be controlled by the insulator thickness. For example, SLG embedded in this framework with a 150 nm-thick SiO2 insulating layer displays a distinctive red color in contrast to its surrounding regions without SLG on the same sample under white light illumination. This opens a potential path towards gate-tunable spectral reflectors. 8Eph receptor A4 regulates motor neuron ferroptosis in spinal cord ischemia/reperfusion injury in rats显示文摘Previous studies have shown that the receptor tyrosine kinase Eph receptor A4(EphA4) is abundantly expressed in the nervous system. The EphA4 signaling pathway plays an important role in regulating motor neuron ferroptosis in motor neuron disease. To investigate whether EphA4 signaling is involved in ferroptosis in spinal cord ischemia/reperfusion injury, in this study we established a rat model of spinal cord ischemia/reperfusion injury by clamping the left carotid artery and the left subclavian artery. We found that spinal cord ischemia/reperfusion injury increased EphA4 expression in the neurons of anterior horn, markedly worsened ferroptosis-related indicators, substantially increased the number of mitochondria exhibiting features consistent with ferroptosis, promoted deterioration of motor nerve function, increased the permeability of the blood-spinal cord barrier, and increased the rate of motor neuron death. Inhibition of EphA4 largely rescued these effects. However, intrathecal administration of the ferroptosis inducer Erastin counteracted the beneficial effects conferred by treatment with the EphA4 inhibitor. Mass spectrometry and a PubMed search were performed to identify proteins that interact with EphA4, with the most notable being Beclin1 and Erk1/2. Our results showed that inhibition of EphA4 expression reduced binding to Beclin1, markedly reduced p-Beclin1, and reduced Beclin1-XCT complex formation. Inhibition of EphA4 also reduced binding to p-Erk1/2 and markedly decreased the expression of c-Myc, transferrin receptor 1, and p-Erk1/2. Additionally, we observed co-localization of EphA4 and p-Beclin1 and of EphA4 and p-ERK1/2 in neurons in the anterior horn. 9Constructing self-supporting surfaces with planar quadrilateral elements显示文摘We present a simple yet effective method for constructing 3D self-supporting surfaces with planar quadrilateral(PQ)elements.Starting with a triangular discretization of a self-supporting surface,we firstcompute the principal curvatures and directions of each triangular face using a new discrete differential geometryapproach,yielding more accurate results than existing methods.Then,we smooth the principal direction field to reduce the number of singularities.Next,we partition all faces into two groups in terms of principalcurvature difference.For each face with small curvature difference,we compute a stretch matrix that turns the principal directions into a pair of conjugate directions.For the remaining triangular faces,we simply keep their smoothed principal directions.Finally,applying a mixed-integer programming solver to the mixed principal and conjugate direction field,we obtain a planar quadrilateral mesh.Experimental results show that our method is computationally efficient and can yield high-quality PQ meshes that well approximate the geometry of the input surfaces and maintain their self-supporting properties.Long Ma Sidan Yao Jianmin Zheng Yang Liu Yuanfeng Zhou Shi-Qing Xin Ying He 2022Computational Visual Media2022,8,4:0
